Project description:Plants have developed a complicated resistance system, and they exhibit various defense patterns in response to different attackers. However, the determine factors of plant defense patterns are still not clear. Here, we hypothesized that damage patterns of plant attackers play an important role in determining the plant defense patterns. To test this hypothesis, we selected leafminer, which has a special feeding pattern more similar to pathogen damage than chewing insects, as our model insect, and Arabidopsis thaliana as the response plants. The local and systemic responses of Arabidopsis thaliana to leafminer feeding were investigated using the Affymetrix ATH1 genome array. Damaged leaves of Arabidopsis thaliana for local damage analysis and the intact leaves on the same plant for systemic damage analysis were separately frozen by liquid nitrogen. Then, we used an Affymetrix ATH1 Arabidopsis microarray to study the expression changes pattern of Arabidopsis thaliana to pea leafminers damage, both locally (LI) and systemically (SI). We downloaded data from the web database and used hierarchical clustering to explore the relationships of Arabidopsis thaliana expression pattern to different kinds of attackers.

Project description:Plant volatiles can mediate plant-plant communication in the sense that plants attacked by herbivores can signal their unattacked neighbors of danger by emitting HIPVs. We call this the priming effect. Since the plant defense response is a systematic process involving numerous pathways and genes,to characterize the priming process, a time course study using a genome-wide microarray may provide more accurate information about the priming process. Furthermore, to what extent do the priming process and direct defense share similar gene expression profiles or pathways are also not clear. We used microarray to detect the priming effect of plant volatiles to healthy Arabidopsis thaliana, and the effect of direct leafminer feeding to Arabidopsis thalianas. A system using Lima bean plants, from which HIPVs can be effectively induced by leafminer feeding, as emitters and Arabidopsis thaliana as receivers is used to track the priming process between neighbor plants. The Arabisopsis thaliana seedlings were treated by volatiles from leafminer fed lima bean for 24h or 48h for RNA extraction and hybridization on Affymetrix microarrays. The Arabisopsis thaliana seedlings fed by leafminer directly were also collected The for RNA extraction and hybridization on Affymetrix micorarrays. We want to explore the response of Arabidopsis thaliana to priming volatiles during a 24h-48h time course. We also want to compare the effect of priming and direct leafminer feeding.

Project description:While pathogen-induced immunity is comparatively well characterized, far less is known about plant defense responses to arthropod herbivores. To date, most molecular-genetic studies of plant-arthropod interactions have focused on insects. However, plant-feeding (phytophagous) mites are also pests of diverse plants, and mites induce different patterns of damage to plant tissues than do well-studied insects (e.g., Lepidopteran larvae or aphids). The two-spotted spider mite, Tetranychus urticae, is among the most significant mite pests in agriculture. T. urticae is an extreme generalist that has been documented on a staggering number of plant hosts (more than 1,100), and is renowned for the rapid evolution of pesticide resistance. To understand reciprocal interactions between T. urticae and a plant host at the molecular level, we examined mite herbivory using Arabidopsis thaliana. Despite differences in feeding guilds, we found that transcriptional responses of A. thaliana to mite herbivory generally resembled those observed for insect herbivores. In particular, defense to mites was mediated by jasmonic acid (JA) biosynthesis and signaling. Further, indole glucosinolates dramatically increased mite mortality and development times. Variation in both basal and activated levels of these defense pathways might also explain differences in mite damage and feeding success between A. thaliana accessions. On the herbivore side, a diverse set of genes associated with detoxification of xenobiotics was induced upon exposure to increasing levels of in planta indole glucosinolates. Our findings provide molecular insights into the nature of, and response to, herbivory for a representative of a major class of arthropod herbivores. We used microarray to assess global gene expresion in Arabidopsis thaliana upon Tetranychus urticae attack in two A. thaliana accessions: Bla-2, resistant to spider mite herbivory and Kon, susceptible to spider mite herbivory. 3 week old Arabidopsis thaliana plants were subjected to Tetranychus urticae attack through application of 10 mites for various periods of time (timecourse scenario) or hundreds of mites for 1 hour (feeding site scenario).

Project description:To investigate the impact of a plant´s response to abiotic stress on plant defense against subsequent biotic stress, we determined the transcriptional response of Arabidopsis thaliana to low temperature stress (4°C) and subsequent mechanical wounding or larval feeding damage by the herbivores Mamestra brassicae (generalist) and Pieris brassicae (specialist). In total, 21%, 4% and 14% of all genes responsive to M. brassicae, P. brassicae or mechanical wounding were differentially regulated in previously cold-treated compared to untreated plants.

Project description:Verticillium dahliae is a soil-borne fungus with a broad host range, including the model plants Nicotiana benthamiana and Arabidopsis thaliana. The plant immunity can be activated by Vd-derived patterns while V. dahliae secreted effectors. We report here the RNAseq analyses of samples from N. benthamiana infected by V. dahliae strains 592 and 171 at different time points. The data showed dynamic expression patterns of genes not only from plant defense signaling but also plant developmental signaling.

Project description:Plants can prepare their defense of impending herbivory, when they previously perceived eggs deposited by herbivores insect which reliably indicates that larvae will soon hatch and feed on the plants. Using Arabidopsis thaliana and Pieris brassicae we addressed the question whether prior egg deposition affects transcriptional changes in feeding-damaged plants. We determined the transcriptomes of Arabidopsis leaves that (i) were for six days deposited with eggs and suffered larval feeding for two days, (ii) suffered larval feeding for two days without prior egg deposition or (iii) were left untreated.

Project description:We have implemented an integrated Systems Biology approach to analyze overall transcriptomic reprogramming and systems level defense responses in the model plant Arabidopsis thaliana during an insect (Brevicoryne brassicae) and a bacterial (Pseudomonas syringae pv. tomato strain DC3000) attack. The main aim of this study was to identify the attacker-specific and general defense response signatures in the model plant Arabidopsis thaliana while attacked by phloem feeding aphids or pathogenic bacteria. Defense responses and networks, unique and specific for aphid or Pseudomonas stresses were identified. Our analysis revealed a probable link between biotic stress and microRNAs in Arabidopsis and thus opened up a new direction to conduct large-scale targeted experiments to explore detailed regulatory links among them. The presented results provide a first comprehensive understanding of Arabidopsis - B. brassicae and Arabidopsis - P. syringae interactions at a systems biology level. Arabidopsis thaliana (ecotype Colombia-0) seeds were grown in 6-cm-diameter pots filled with a sterile soil mix (1.0 part soil and 0.5 part horticultural perlite), 3 plants per pot. Plants were kept in growth chambers VM-CM-6tsch VB 1514 (VM-CM-6tsch Industrietechnik GmbH, Germany) under the following conditions: a 8/16 h (light/dark) photoperiod at 22M-BM-0C/18M-BM-0C, 40%/70% relative humidity, and 70/0 M-NM-<mol m-2s-1 light intensity. After 32 days plants had 8 fully developed leaves. Each plant was infested with 32 wingless aphids [Brevicoryne Brassicae] (4 per leaf), which were transferred to leaves with a fine paintbrush. Infested plants and aphid-free controls were kept in plexiglass cylinders. Plants were harvested 72 h after infestation between the 6th and 8th hour of the light photoperiod. Four biological replicates were prepared from aphid infested and control plants, each sampled from 15 individual plants. Whole rosettes were cut at the hypocotyls and aphids were removed by washing with Milli-Q-filtered water. Differences in transcriptional responses were measured by comparing genes expression of aphid infested plants against non-infested control plants.
