In this study, the proteomic profiles of single-sex infected female (SF) worms and bisexual infected mature female (MF) worms of Schistosoma japonicum at 18, 21, 23 and 25 days post infection (dpi) were identified with isobaric tags for relative and absolute quantitation-coupled liquid chromatography-tandem mass spectrometry. Differentially expressed proteins (DEPs) were subsequently used for bioinformatic analysis.

Effect of frataxin silencing by RNAi on 4, 7 and 14 days old worms

Project description:The study aimed to investigate the mechanisms that promote compensatory pro-longevity responses during mild mitochondrial stress in Caenorhabditis elegans. This was achieved by performing a DNA microarray analysis to compare gene expression profiles of wild-type and mild mitochondrial-stressed long-lived worms at days 4, 7, and 14 after birth, in five replicates. The worms were fed with either an empty-vector control (pL4440) or vector-expressing dsRNA against nematode frataxin (frh-1) to induce mild mitochondrial stress, for 3 consecutive generation starting from eggs.

Project description:Eggs were isolated by hypochlorite treatment and allowed to hatch in the absence of food as L1 stage arrested worms in S-buffer. Worms were then transferred to large plates containing NGM alone or NGM+ resveratrol and allowed to grow till young adult stage (ca 40 hours). Worms were harvested and total RNA extracted using Trizol reagent. Poly-A+ RNA was isolated using Qiagen midi-prep kit and used for microarray hybridization. N-glycosite mapping in Brugia malayi female worms, male worms, and microfilariae

Project description:N-linked glycosylation is a critical post translational modification of eukaryotic proteins. N-linked glycans are present on surface and secreted filarial proteins that play a role in host parasite interactions. Examples of glycosylated Brugia malayi proteins have been previously identified but there has not been a systematic study of the N-linked glycoproteome of this or any other filarial parasite. In this study, we applied an enhanced N-glyco FASP protocol using an engineered carbohydrate-binding protein, Fbs1, to enrich N-glycosylated peptides for analysis by LC-MS/MS. We then mapped the N-glycosites on proteins from three host stages of the parasite: adult female, adult male and microfilariae. Fbs1 enrichment of N-glycosylated peptides enhanced the identification of N-glycosites. Our data identified 582 N-linked glycoproteins with 1273 N-glycosites. Gene ontology and cell localization prediction of the identified N-glycoproteins indicated that they were mostly membrane and extracellular proteins. Comparing results from adult female worms, adult male worms, and microfilariae, we find variability in N-glycosylation at the protein level as well as at the individual N-glycosite level. These variations are highlighted in cuticle N-glycoproteins and adult worm restricted N-glycoproteins as examples of proteins at the host parasite interface that are well positioned as potential therapeutic targets or biomarkers.

Transcriptome of wild type N2 worms at embronic stage and starved L1 stage

Project description:Transcription factors are key components of regulatory networks that control development, as well as the response to environmental stimuli. We have established an experimental pipeline in C. elegans that permits global identification of the binding sites for transcription factors using chromatin immunoprecipitation and deep sequencing. We describe and validate this strategy, and apply it to the transcription factor PHA-4, which plays critical roles in organ development and starvation stress response. We defined binding sites for PHA-4 during formation of the embryonic pharynx, and in starved larvae. PHA-4 binds to thousands of locations throughout the genome. These sites shift dramatically between embryos and larvae, from developmentally regulated genes to genes involved in metabolism. We acquired transcriptome of wild type N2 worms at embyonic and starved L1 stages using RNA-seq method in comparing to the ChIP-seq experiment to study transcription factor binding. The wild type worms were cultured the same as the transgenic worms for transcription factor binding experiment.
