Two-condition experiment, Dot1lAC vs. Dot1lf/f. Biological replicates: Total kidney RNAs from 4 mice/genotype were mixed within the same genotype.

Project description:Ubiquitylation plays an important role in the control of Na+ homeostasis by the kidney. It is well established that the epithelial Na+ channel ENaC is regulated by the ubiquitin-protein ligase NEDD4-2, limiting ENaC cell surface expression and activity. Ubiquitylation can be reversed by the action of deubiquitylating enzymes (DUBs). One such DUB, USP2-45, was identified previously as an aldosterone-induced protein in the kidney, and is also a circadian output gene. In heterologous expression systems USP2-45 binds to ENaC, deubiquitylates it and enhances channel density and activity at the cell surface. Because the role of USP2-45 in renal Na+ transport had not been studied in vivo, we investigated here the effect of Usp2 gene inactivation in this process. We demonstrate first that the USP2-45 protein has a rhythmic expression with a peak at ZT12. Usp2-KO mice did not show any differences to wild-type littermates with respect to the diurnal control of Na+ or K+ urinary excretion and plasma levels neither on standard diet, nor after acute and chronic changes to low and high Na+ diets, respectively. Moreover, they had similar aldosterone levels either at low or high Na+ diet. Blood pressure measurements using telemetry did not reveal variations as compared to control mice. Usp2-KO did neither display alternations in ENaC or Na+,Cl--cotransporter (NCC) expression, nor were there any changes in regulatory protein levels, as evidenced by immunoblotting and transcriptome analysis. We conclude that USP2-45 is not crucial for the regulation of Na+ balance or maintenance of blood pressure in vivo. We compared gene expression in kidneys from Usp2 Knock-Out versus Wild-Type. Six pairs of male littermates Usp2-KO and WT were combined in two pools per condition (3 animals per pool) keeping the littermate pairing between pools.

Project description:Fibroblast growth factor-23 (FGF23), a circulating protein produced in bone, causes renal inorganic phosphate (Pi) wasting by down-regulation of sodium phosphate co-transporter 2a (Npt2a). The mechanism behind this action is unknown. We have previously generated transgenic mice (TG) expressing human wild-type FGF23 under the control of the α1 (I) collagen promoter. In this study we performed a large scale gene expression study of kidneys from TG mice and wild-type littermates. Several genes that play a role in Pi regulation had decreased expression levels, such as Npt2a, but also Pdzk1 which is a scaffolding protein known to interact with NPT2a. Importantly, the Klotho gene, a suggested crucial co-factor for FGF23 receptor binding and activation, was the most affected decreased gene. However, other genes proposed to regulate Pi levels, such as secreted Frizzled Related Protein 4 (sFRP4), Na+/H+ exchanger regulatory factor 1 (NHERF1) and the FGF-receptors 1-4, revealed no changes. Interestingly, expression levels of inflammatory response genes were increased and histological analysis revealed tubular nephropathy in the TG mice kidneys. In conclusion, FGF23 TG mice have altered kidney gene expression levels of several genes thought to be part of Pi homeostasis and an increase in inflammatory response genes, data supported by histological analysis. These findings may lead to further understanding of how FGF23 mediates its actions on renal Pi regulation. Experiment Overall Design: Five kidneys from FGF23 TG mice and five kidneys from WT littermates was used for Affymetrix Genechip analysis. One Genechip was used/animal. Animals were 8 weeks old when kidneys were collected.

Project description:Background: The transcriptional coactivator with a PDZ-binding motif (TAZ), a downstream effector of the Hippo signaling pathway, regulates the expression of target genes by acting as a transcription cofactor. TAZ knockout (KO) mice exhibit polycystic kidneys and polyuria. This study aimed to investigate the role of TAZ in vasopressin-induced AQP2 regulation. Methods: 1) TAZ knockdown mediated by siRNA in mpkCCDc11 cells; 2) qRT-PCR, semiquantitative immunoblotting, and immunocytochemistry of AQP2; and 3) Next Generation Sequencing (NGS) in mpkCCDc11 cells, a mouse collecting duct cell line. Results: Endogenous expression of AQP2 was induced in mpkCCDc11 cells by treatment with dDAVP (10-9 M) treatment. Treatment with dDAVP (10-9 M) for 24 h increased AQP2 mRNA (12,608 ± 177% of control) and AQP2 protein levels (287 ± 15%). On the contrary, the increase induced by dDAVP in AQP2 mRNA (9,240 ± 241% of the control) and protein levels (215 ± 23%) was significantly reduced in TAZ-KD cells. TonEBP protein levels remained unchanged in TAZ-KD cells. NGS identified several potential AQP2 transcription factors (TF), including Klf6, Irf3, Cebpb, and Nr4a1, selected based on previous in silico database analysis. Among these, Nr4a1 was chosen for further investigation due to its significantly reduced expression of mRNA in TAZ-KD cells, as confirmed by qRT-PCR. Conclusion: TAZ appears to affect dDAVP-induced AQP2 trafficking through mechanisms not mediated by the cAMP/PKA pathway, suggesting the involvement of other non-canonical pathways. TAZ may also regulate AQP2 abundance, potentially through interaction with several transcription factors, including Nr4a1.

Project description:Metabolic dysfunctions, such as fatty liver, obesity and insulin resistance, are among the most common contemporary diseases worldwide. Mimp/Mtch2 is a mitochondrial carrier protein homologue that leads to mitochondrial depolarization, localizes to the mitochondria and induces accumulation of fat vesicles. Transgenic mice overexpressing Mimp/Mtch2 develop fatty livers and kidneys and exhibit high blood glucose levels. The mechanism of lipid accumulation in the kidney has not been fully determined. In this study we performed a differential gene expression profile of fatty compared to non-fatty kidneys of Mimp/Mtch2-GFP transgenic mice, fed on high fat diet. RNA samples for microarray gene expression profiling were obtained from four Mimp/Mtch2-GFP mice, two samples of low fat kidneys and two of fatty kidneys.

Project description:Epigenetic enzymes play critical roles in embryogenesis by defining higher-order chromatin structures required for the establishment of organ-specific transcriptional networks. In this study we investigated the role of the histone 3 lysine 79 methyltrasferase Dot1L in cardiomyocytes during development. We conducted chromatin immunoprecipitation experiments for H3K79me2 in embryonic (E16.5) and neonatal (P1) cardiomyocytes from Dot1L cardiomyocyte-specific conditional knock out and control mice. Genes that bear this modification in DOT1L controls lose it in cKOs, consistent with the model of DOT1L as the sole histone K79 methyltransferase. Overall, we identified enrichment of methylation of H3K79 in gene bodies and in regulatory regions associated wih trascription factors essential for cardiac patterning at E16.5 and with genes important for cardiomyocyte proliferation and maturation in the postnatal period.

Project description:Global gene expression in the 20-week remnant kidneys of uninephrectomized mice fed either a chow or a high fat diet was compared with kidney tissue from the corresponding sham groups. Results provide insight into mechanisms underlying effects of high-fat induced obesity on gene expression in mouse kidney. Male C57/BJ mice aged 6 weeks were randomly assigned to uninephrectomy (UNX) or sham procedures and fed with a high fat diet or control chow diet. All mice were sacrificed under anesthesia 20 weeks after surgery and kidneys were harvested. 3-4 total RNA samples per group were analyzed and gene expression was compared between groups.
