ABSTRACT: This study utilized comparative global gene expression microarray analysis to evaluate the effects of two different Montanide adjuvants, ISA70 and ISA71, in promoting chicken protective immunity following vaccination with profilin, an actin-regulatory protein that is expressed by multiple Eimeria species.

Project description:This study investigated the ability of two novel adjuvant formulations, QCDC (Quil A/cholesterol/DDA/Carbopol) and QCDCR (QCDC/Bay R1005), in combination with a recombinant profilin vaccine, to modulate host protective immunity and to alter new gene expression during experimental avian coccidiosis. Four-condition experiment, Profilin only vs. Non-vaccination, Profilin only vs. Co-vaccination of QCDC plus profilin, Profilin only vs. Co-vaccination of QCDCR plus profilin, Biological replicates: 2 profilin only replicates, 2 Non-vaccination replicates, 2 QCDC plus profilin replicates, 2 QCDCR plus profilin replicates with dye-switching.

Project description:Avian coccidiosis is a major disease of poultry caused by the intestinal protozoa Eimeria. Aviagen line A and line B birds show differential susceptibility to Eimeria infection, with line B birds exhibiting higher lesion scores and mortality. The objective of this study was to examine differential intestinal gene expression between line A and B chicks in response to a challenge with Eimeria maxima. Following challenge with 1 x 10^4 oocysts/chick, greater than 40% of line A chicks had lesion scores of 0 to 1 (on 0 to 4 scale), similar to controls. In contrast, all line B challenged chicks at this same dose had lesion scores of 2 to 4.

We used microarray to comprehend the molecular signatures of mucosal adjuvants in the mouse vagina. The adjuvants studied, CpG-ODN and α-GalCer have previously been shown to be potent mucosal adjuvants in mice when administrered together with a glycoprotein from HSV-2.
