Project description:Affymetrix expression profiling was used to evaluate the association between IL13Rα2 expression, and mesenchymal, proneural, classical and neural signature genes expression for glioma subclasses defined by Verhaak et al (Cancer Cell; 2010). The goal of this study is to compare gene expression patterns between IL13Ra2-postive and IL13Ra2-negative glioma cell lines. IL13Ra2 status was determined by flow cytometry and confirmed by qPCR.

Project description:We report the high-throughput profiling of thirteen human glioblastoma and four pairs of panned(pdgfra+) and supernatant(pdgfra-) human glioma cells. The subtypes and the different expression genes(DEGs) was identified integrated genomic analysis:single sample GSEA and DESeq.We obtained the enrichment gene pathway by using Gene Ontology Enrichment analysis-Functional classification viewed in pie chart.We find that the features of Proneural were enriched generaly when comparing the four pairs of the panned(pdgfra+) and supernatant(pdgfra-) human glioma cells.

Project description:Identification of critical survival determinants of PDGF-driven proneural glioma. Results provided information about the genes and pathways that are regulated by PDGF signaling in PDGF-driven proneural glioma and led to the assessment of the importance of the USP1-ID2 axis in proneural glioma. Total RNA was obtained from untreated and Dox treated PDGF-driven glioma spheroid cells (PDGF-GSC) which had been isolated from the Gfap-tTa/Tre-PDGFB mouse model used in our study. The data were analyzed to determine the genes that are regulated by PDGF signaling in PDGF-driven glioma.

A single-cell atlas of human glioma

Project description:Although tumor-propagating cells can be derived from glioblastomas (GBMs) of the proneural and mesenchymal subtypes, a glioma stem-like cell (GSC) of the classical subtype has not been identified. It is unclear if mesenchymal GSCs (mGSCs) and/or proneural GSCs (pGSCs) alone are sufficient to generate the spectrum of cellular heterogeneity observed in GBM. This study sheds light on a long-standing debate regarding lineage relationships among GSCs.

Molecular signature of human cerebral organoids glioma model

Project description:We have developed a cancer model of gliomas in human cerebral organoids by CRISPR/Cas9 technology to target a Harvey-Ras (H-RAS) G12V-IRES-tdTomato construct by homologous recombination into the TP53 locus. 16 weeks after electrporation, these tdTomato positive transduced cells were sorted and compared to nontransduced cellls by RNA-seq and gene expression profiles were compared to each other and with human glioma Verhaak subtype signatures.

Project description:In this study, we performed miRNA profiles analysis of 84, 17, 20, 83, 718, 1123, 528 and 816 glioma spheres compared to normal progenitor sample (16wf) using microarray to evaluate their potential role in regulation of the biological properties of proneural and mesenchymal glioma spheres miRNA profiling analysis of the 9 samples including 8 patient-derived samples including glioma sphere samples (84, 17, 20, 718, 816, 528, 83 and 1123), as well as one normal progenitor sample (16wf).
