Project description:The Poly mutant was identified as a previously uncharacterised, but essential gene in Drosphila. Loss of Poly reults in lethality at the third larval instar stage, but only after a stage of extreme larval longevity. We aimed to assess the consequences of loss of the Poly gene on glocal gene expression by microarray analysis. Metabolism is altered in Poly mutants and here we find altered gene expression in insulin signaling pathways. Four biological replicates each of Poly mutant and wild-type larvae were used to assess gene expression on FL002 microarrays (GPL5135) . These data reveal that Poly is a mediatior of insulin receptor/TOR signaling.

Gene expression profiling of Drosophila melanogaster larval brains after chronich alcohol exposure

Project description:We sequenced mRNA extracted from brains of (1) D. melanogaster larvae exposed to food containing 5% ethanol (v/v) for 6 conscutive days, and (2) an age-matched untreated control larvae, that grew in regular food. Differential gene expression between the two groups was calculated and reported. Each group consisted of 3 biological replicates of 30 brains each.

Project description:Samples 1-24: Tissue-specific gene expression microarrays (Nimblegen) using dissected ring glands isolated from FOUR different time points of control (w1118, otherwise wild type) third instar larvae. Time points are 4, 8, 24 and 36 hours after the molt from second to third instar larvae. Samples 25-42: Tissue-specific gene expression microarrays (Nimblegen) using dissected ring glands isolated from TWO different time points of third instar larvae. Genotypes were phantom22-GAL4/RasV12 and phantom22/Torso-RNAi. Goal was to identify PTTH-dependent gene sets in the ring gland. Time points were 18 hours and 8 hours prior to puparium formation.

Drosophila CNS mitochondrial DNA dysfunction microarray

Project description:Mitochondrial DNA (mtDNA) encodes essential components of the respiratory chain and loss of mtDNA leads to mitochondrial dysfunction and neurodegeneration. Mitochondrial transcription factor A (TFAM) is an essential component of mtDNA replication and a regulator of mitochondrial copy number in cells. Studies have shown that TFAM knockdown leads to mitochondrial dysfunction and respiratory chain deficiencies. Using gene expression analysis, we aimed to investigate the effects of mtDNA dysfunction in the CNS at the molecular level. We used microarray analysis to investigate gene expression in cases of mitochondrial dysfunction in the CNS. RNA was purified from the late third instar larval CNS from control larvae, or larvae over-expressing mitochondrial transcription factor A (TFAM) in post-mitotic neurons using the neuron specific driver nsyb-Gal4. Three replicates are included for each condition.
