Project description:To profile the expression of microRNAs (miRNAs) in the whole blood of mice in exposure to intraperitoneal lipopolysaccharide (LPS) injection. C57BL/6 mice which received intraperitoneal injections of different concentration (ranged from 10 to 1000 μg) of LPS originated from different bacteria, including E. coli, Klebsiella pneumonia, Pseudomonas aeruginosa, Salmonella enterica, and Serratia marcescens were sacrificed at indicated survival times (2, 6, 24, and 72 h) following LPS injection. The whole blood was drawn and the tissues including lung, brain, liver and spleen were harvested for miRNAs expression analysis with miRNA array (Phalanx Mouse & Rat miRNA OneArray® 1.0). These differentially expressed miRNAs were applied for hierarchical cluster analysis using average linkage and Pearson correlation as a measure of similarity.

Project description:To examine how activation of different toll-like receptors (TLR) impacts gene expression in Autism Spectrum Disorder (ASD), we cultured peripheral blood monocytes from children with ASD, Pervasive Developmental Disorder Not Otherwise Specified (PDDNOS) or Asperger and typically developing children and treated them with either lipoteichoic acid (LTA) or lipopolysaccharide (LPS) to activate LTR2 or 4 respectively. Following 24 hours of stimulation, we then performed RNA sequencing to profile mRNA responses between non-treated (NT), LTA and LPS treated samples for each diagnosis (control or ASD).

Project description:Odontoblasts and fibroblasts are suspected to influence the innate immune response triggered in the dental pulp by micro-organisms that progressively invade the human tooth during the carious process. To determine whether they differ in their responses to oral pathogens, we performed a systematic comparative analysis of odontoblast-like cell and pulp fibroblast responses to TLR2, TLR3 and TLR4 specific agonists (lipoteichoic acid [LTA], double-stranded RNA and lipopolysaccharide [LPS], respectively). Cells responded to these agonists by differential up-regulation of chemokine gene expression. CXCL2 and CXCL10 were thus increased by LTA only in odontoblast-like cells, while LPS increased CCL7, CCL26 and CXCL11 only in fibroblasts. These data suggest that odontoblasts and pulp fibroblasts differ in their innate immune responses to oral micro-organisms that invade the pulp tissue. Project description:We used a systemic inflammatory challenge in animals with chronic mesial temporal lobe epilepsy to identify novel molecular pathways involved in seizure regulation. Mice were first rendered epileptic by a single intrahippocampal injection of kainic acid (KA). After a period of two-three weeks, mice developed spontaneous seizures and then received a single intraperitoneal injection of lipopolysaccharide (LPS) to mimic systemic inflammation, or saline as a control. Gene expression analysis by microarrays was then performed on hippocampal RNA samples extracted 4 and 24 hours after LPS or saline treatment (n=3 samples per treatment group).

Project description:The pathogenic mechanisms of common kidney glomerular diseases, including the vast majority of cases of proteinuria, remain unknown. Project description:Bifidobacterium animalis subsp. lactis BB12 is a widely used probiotic bacterium. However the molecular mechanism by which this bacterium confers positive health effects to the host is largely unknown. The importance of lipoteichoic acids (LTA) in the cross-talk between the bacteria and human enterocyte-like Caco-2 cells was investigated in the present study. A microarray-based analysis of gene expression revealed induction of genes encoding enzymes involved in the synthesis of lipoteichoic acids of BB12. This observation was confirmed in phenotypic experiments, where a specific galactofuranosyl carbohydrate epitope present in the structure of lipoteichoic acids of BB12 was used as a detection marker. An exopolysaccharide EPS/LTA extract isolated from BB12 and commercially available pure LTAs from the human pathogen S. pyogenes were tested in competitive adhesion experiments. The adhesion of B. animalis subsp. lactis BB12 to Caco-2 cells was inhibited in presence of LTA. The strongest inhibition was observed in the concentration range 0.5-10 ug/ml for LTAs from S. pyogenes and 5-10 ug/ml for EPS/LTA extract from BB12. The inhibitory concentrations in both cases likely reflect the physico-chemical properties of LTAs with the highest efficacy below the critical micelle concentration, and therefore we ascribe the main inhibitory effect to LTAs for the BB12 isolated EPS/LTA extract as well. In conclusion we present increased biosynthesis of lipoteichoic acids of BB12 co-incubated with human Caco-2 cells and evidence that LTA serve as important cell wall factors involved in adhesion to these human enterocyte-like cells.
