Project description:Bioinformatic prediction, deep sequencing of microRNA and expression analysis during phenotypic plasticity in the pea aphid acyrthosiphon pisum We developed high throughput Solexa sequencing and bioinformatic analyses of the genome of the pea aphid Acyrthosiphon pisum in order to identify the first miRNAs from a hemipteran insect. By combining these methods we identified 155 miRNAs including 56 conserved and 99 new miRNAs. Moreover, we investigated the regulation of these miRNAs in different alternative morphs of the pea aphid by analysing the expression of miRNAs across the switch of reproduction mode.

Cuticular structure proteomics analyses of the pea aphid Acyrthosiphon pisum

Project description:Aphids are sap-feeding insects with piercing-sucking mouthparts. They are serious pests in agriculture and provoke significant losses, with a formidable capacity to transmit numerous plant viruses causing diseases in almost all important crops. The majority of viruses are acquired after feeding on an infected plant, retained on the aphid’s stylet and inoculated to healthy ones with a single probing puncture, promoting viral outbreaks. We have recently discovered the acrostyle, an organ located at the tip of aphid maxillary stylets, that harbors the receptors for Cauliflower Mosaic Virus (CaMV), and likely those for numerous other viruses. To our opinion, establishing an exhaustive inventory of the cuticular proteins composing the acrostyle, defining the peptide motifs accessible at the surface, identifying virus receptors among these proteins, will contribute to better understand virus/vector and plant/insect interactions at different scales including the molecular level. In this manuscript, we identify proteins or peptides at the surface of aphid stylets that might play a key role in plant virus binding. This may help to pave the way for future alternative “environmentally safe” strategies to control viral spread and aphid infestation in important crops worldwide.

Gall aphid saliva LC-MS/MS - Proteins Identified from Saliva and Salivary Glands of the Chinese Gall Aphid Schlechtendalia chinensis

Project description:Aphid saliva plays an essential role in the interaction between aphids and their host plants. Several aphid salivary proteins have been identified and analyzed. However, none of the characterized salivary proteins are from galling aphids. Here we analyzed the salivary proteins from the Chinese gall aphid, Schlechtendalia chinensis using LS-MS/MS analysis. A total of 31 proteins were identified directly from secreted saliva collected via artificial diet, and 141 proteins were identified from protein extracts derived from dissected salivary glands. Among these identified proteins, 17 were found in both secreted saliva and dissected salivary glands. In comparison with salivary proteins identified from three other free living aphids, the most striking feature of the salivary protein from S. chinensis is the existence of high proportion of proteins with binding activity, including DNA binding, protein binding, ATP binding and ion binding proteins et al. We speculate that these binding proteins may be involved in induction of gall formation. Our results provide a framework for future research to elucidate the molecular basis for gall induction by S. chinensis.

Whole genome bisulfite sequencing data of Russian wheat aphid biotypes US-RWA1 (co-fed), US-RWA2 (co-fed) and US-RWA2 (isolated)

Project description:The aim for this study was to compare the differences in the methylome of three Russian wheat aphid populations that differ in virulence and bacterial load. Differences in the methylome of US Russian wheat aphid biotype 2 (US-RWA2) that have undergone removal of their phytobiomes through isolation of newly born nymphs from their mothers compared to nymphs who stayed in close proximity to their progenitors. US-RWA1 is the least virulent biotype and US-RWA2 is the most virulent biotype. The isolated US-RWA2 population (with a reduced bacterial load) has been shown to be less virulent than the co-fed population (Luna et al., 2018). The corresponding two methylomes were then compared to the methylome of US Russian wheat aphid biotype 1 (US-RWA1 is less virulent than US-RWA2) to ascertain the contribution (or lack thereof) of the phytobiome in regards to the increased/decreased virulence of phytobiome present and absent aphids. We investigated the overall, genic, intergenic, exonic and intronic methylation - as well as genic and intergenic hemimethylation -all three available contexts ( CpG, CHG and CHH) through use of the Bismark pipeline. These findings were used to ascertain to what effect the phytobiome (bacterial load) had on methylation within the US Russian wheat aphid biotypes.
