Project description:Investigation of genome-wide germline copy number mutations in mice after parental exposure to 3 Gy of acute X-rays The level of copy number mutations in offspring of exposed paternal mice was substantially increased when compared to the control Overall 262 samples were used of which 93 were control samples and 169 treated samples. The control samples came from offspring of parents (C57BL6/J (male) and CBA/Ca (female)) with sham -ir while the treated samples were from offspring of 3 Gy whole-body acute irradiated paternal and non-exposed maternal. The treated samples classified into two groups: post-meiotic and pre-meiotic according to exposed status of the sperm that gave rise to the offspring. The reference sample for each offspring was equal mixture of paternal and maternal DNA.

Analysis of mRNA expression between irradiated and non-irradiated keloid-derived primary cells.

Project description:This study aims to compare mRNA expression between irradiated and non-irradiated keloid-derived primary cells by RNA-Seq. In this study, keloid-derived primary cells that were isolated from patients, and then divided into three groups and each group had four repeats. The first group of cells was treated without radiation, the second group of cells were received 4 Gy delivered in 2-Gy fractions once per day over 2 days and the third group of cells were received 8 Gy delivered in 2-Gy fractions once per day over 4 days.The last two groups of cells were irradiated at a fixed rate of 1.15 Gy/min using an X-ray linear accelerator (Rad Source Technologies Inc., Suwanee, GA, USA). 24 hours post irradiation of the third group of cells, all keloid-derived primary cells were collected and subjected to mRNA expression analysis.

Project description:Exposure to ionizing radiation around the time of neural tube closure results in a number of developmental defects, including neural tube defects (e.g. exencephaly, cleft palate) and eye defects (e.g. microphtalmos, anaphtalmos). These defects result from perturbed neural tube closure and development of the presumptive eye. In order to better understand the underlying molecular mechanisms, gene expression profiling was performed in heads of mouse embryos at embryonic day 9, after they had been irradiated at embryonic day 7.5 with 1 Gy of X-rays. Non-irradiated mice served as controls for differential gene expression.

Project description:Transcriptional profiling of human lymphoblastoid TK6 cells comparing mock irradiated cells with cells exposed 24 hours previously to 1.67 Gy HZE (1 GeV/amu iron ions accelerated at the NASA Space Research Laboratory (NSRL) of Brookhaven National Laboratory) or 2.5 Gy 137Cs gamma rays. TK6 cells were mock irradiated or exposed to HZE or gamma-rays, and RNA was harvested 24 hours later. 3 biological replicates were independently grown and harvested during three different runs at the NSRL. One replicate per array.
