MiRNA microarray of mice with unilateral ureteral obstruction

ABSTRACT: To find miRNAs that involve in renal epithelial transition and renal fibrosis, we performed unilateral ureteral obstruction of mice for 7 days. After that, we harvested kidneys, and performed microarray of miRNA. Contralateral kidneys without ureteral obstruction were used as controls. miRNAs were purified from kidneys with ureteral obstruction and contralateral kidneys without ureteral obstruction. Then microarray of miRNA was performed (n=4). miRNAs up-regulated in kidneys with ureteral obsctruction compared with contralateral kidneys were sorted.

Exploring new pathways involved during experimental renal fibrogenesis in the Unilateral Ureteral Obstruction

Project description:Chronic kidney disease (CKD) is a burden for Public Health and concerns millions of individuals worldwide. Independently of the cause, CKD is secondary to the replacement of functional renal tissue by extra-cellular matrix proteins (i.e fibrosis) that progressively impairs kidney function. The pathophysiological pathways that control the development of renal fibrosis are common to most of the nephropathies involving native kidneys or kidney grafts. Unfortunately, very few treatments are available to stop renal fibrosis and most of the therapeutic strategies are often barely able to slow down the progression of fibrogenesis in native kidneys. Therefore, it is mandatory to discover new therapeutic pathways to stop renal fibrosis. Our objective is to study new pathways involved in renal fibrosis. We thus decided to use the model of Unilateral Ureteral renal Obstruction in mice, a fast and reproducible experimental model of renal fibrosis. We studied renal fibrosis using experimental model of ureteral unilateral obstruction in mice, which was performed by complete ligation of the left ureter. The control lateral right kidney served as internal control.

Expressions of MicroRNA in Mouse Kidney with Unilateral Ureteral Obstruction

Project description:Renal fibrosis is the final common pathway for chronic kidney disease. However, the detailed mechanisms and therapeutic strategies for renal fibrosis have not been established. MicroRNAs (miRNAs) are small, non coding RNAs that regulate various pathological conditions and diseases. Previous studies reported that miRNAs play a pivotal role in renal fibrosis. However, the role of miRNAs in renal fibrosis has not been completely elucidated. Therefore, in this study, miRNAs on renal fibrosis was investigated in a renal fibrosis mouse model generated by unilateral ureteral obstruction. MiRNA microarray analysis revealed 109 miRNAs that were upregulated more than 2 fold and 113 miRNAs that were downregulated less than 0.5 fold in the kidney of UUO mice compared with control mice.

Kidney Tissue Targeted Metabolic Profiling of Unilateral Ureteral Obstruction Rats by NMR

Project description:Renal interstitial fibrosis is a common pathological process in the progression of kidney disease. A nuclear magnetic resonance (NMR) based metabolomics approach was used to analyze the kidney tissues of renal interstitial fibrosis (RIF) rats, induced by unilateral ureteral obstruction (UUO). The combination of a variety of statistical methods was used to screen out 14 significantly changed potential metabolites related with multiple biochemical processes including amino acid metabolism, adenine metabolism, energy metabolism, osmolyte change and induced oxidative stress. The exploration of the contralateral kidneys enhanced the understanding of the disease, which was also supported by serum biochemistry and kidney histopathology results. In addition, the pathological parameters (clinical chemistry, histological and immunohistochemistry results) were correlated with the significantly changed differential metabolites related with RIF. This study shows that target tissue metabolomics analysis can be used as a useful tool to understand the mechanism of the disease and provide a novel insight in the pathogenesis of RIF.

Proteomics of obstructed kidneys from patients with obstructive nephropathy and unilateral ureteral obstruction mice

Project description:Here, we performed proteomics of kidneys obtained from patients with obstructive nephropathy and healthy controls (n=11/8), or from unilateral ureteral obstruction (UUO) mice and sham mice (n=6/6). Proteomic measurement of all human kidney samples resulted in a total of 6089 proteins with an average of 5780 proteins per sample. Compared to control kidneys, 614 proteins were up-regulated and 855 proteins were down-regulated (p value < 0.05, q value < 0.05, and fold change > 2 or < 0.5) in human obstructed kidneys. The up-regulated proteins were significantly enriched in collagen formation and immune response. Moreover, the human and mouse kidney proteomics reveled an aberrant NAD+ metabolism and NAD+-related metabolic pathways, including mitochondrial dysfunction, oxidative phosphorylation, and tryptophan metabolism. The expression patterns of several key enzymes catalyzing NAD+ biosynthesis and consumption were dramatically altered in obstructive nephropathy. Moreover, the major NAD+ consumer CD38 was strongly induced in obstructed kidneys which contributed to the decline of renal NAD+ levels. Functional studies demonstrated that CD38 deletion or inhibition significantly recovered renal NAD+ levels and reduced obstruction-induced renal fibrosis.

Identification of Transcripts Associated with Renal Damage Due to Ureteral Obstruction as Candidate Urinary Biomarkers

Project description:Renal obstruction is a common cause of renal failure in adults and children and is suspected when hydronephrosis is detected on imaging. Since not all cases of hydronephrosis are associated with renal damage, biomarkers are needed to guide intervention to relieve obstruction. We performed gene expression profiling on the kidneys from adult mice over a detailed time course after obstruction and compared these data to a neonatal model of bilateral high grade obstruction induced by conditional deletion of the calcineurin ß1 (Cnb1) gene. Using a model of short term (non-damaging) and long term (damaging) temporary obstruction, we identified which of the transcripts common to the adult and neonatal models were induced early and associated with renal damage. Using QPCR, elevated levels of several candidate RNA biomarkers of renal damage were detected in urine from obstructed mice and children undergoing surgery for ureteropelvic junction obstruction (UPJO).
