Androgen receptor coregulators baseline comparisons

ABSTRACT: LNCaP cells are an established androgen receptor expressing prostate carcinoma cell line. Human foreskin fibroblasts also expressing the androgen receptor were obtained from phenotypic normal male individuals. Cells were cultured either at confluency leading to G0 cell cycle state or while they were proliferating. Cells were either untreated, or treated with dihydrotestosterone (DHT) or ethanol (ETOH) which also served as the solvent for the DHT. All experimental RNA samples derived from the untreated or treated cell lines were hybridized on cDNA arrays against a common reference. This reference was composed out of common reference CRG (50%) and out of fibroblast RNA (50%). This reference is also called "mixed reference" in the description of the 26 individual experiments. A strain or line experiment design type assays differences between multiple strains, cultivars, serovars, isolates, lines from organisms of a single species. Cells were either proliferating or they were G0-arrested. Some hybridizations were performed in the type 1 design. In these cases, the hormone treated sample was hybridized against the ethanol treated sample on the same microarray. Hormone mediated induction or repression of gene transcription can directly be deduced from R/G normalized ratios on arrays. For other experiments, the ethanol treated control and the hormone treated experiment were hybridized on separate microarrays against a common reference of RNA. This reference was composed out of common reference batch CRG (50%) and a fibroblast RNA (50%). This reference was called mixed reference. For all experiments, the same batch of mixed reference was used. Regulation of gene expression by JMJD5 in LNCaP prostate cancer cells

Project description:The goal of this study was to define the cohort of genes whose expression is regulated by JMJD5 function in prostate cancer. Towards this goal, LNCaP sublines stably expressing wld-type JMJD5 (LNCaP-JMJD5) were established. In addition, a control subline(LNCaP-Vector) was generated by transfection with empty vector. Both sublines were cultured in either androgen-deprived medium or in the presence of dihydrotestosterone (DHT). Subsequently, total RNA was isolated and then subjected to microarray gene expression profiling with Affymetrix GeneChip HG-U133 Plus 2.0 Arrays. A total of 4 samples were analyzed in this study. The study included two cell lines, LNCaP-Vector and LNCaP-JMJD5, that were cultured under conditions of androgen depivation (-DHT) or supplementation with dihydrotestosterone (+DHT). The LNCaP-Vector cell line served as the control for the study.

FoxA1 specifies unique androgen and glucocorticoid receptor binding events in prostate cancer cells (mRNA)

Project description:We report the androgen receptor recruitment to the chromatin of androgen responsive prostate cancer cell lines, LNCaP-1F5 and VCaP in response to physiological androgen 5a-dihydrotestosterone (DHT) using ChIP-sequencing. We compare the AR recruitment by DHT to that by partial agonist/antagonist cyproterone acetate and mifepristone (RU486) in LNCaP-1F5 cells. We also report the role of glucocorticoid receptor recruitment in presence of dexamethasone (Dex) in androgen responsive prostate cancer cells. The AR and GR cistrome analysis is subsequently compared with gene expression data and RNA Pol II analysis. The ChIP-seq has been performed using AR, GR, RNA Pol II antibodies. Expression profiling by microarray of LNCaP-1F5 cells treated with vehicle, 100 nM 5a-dihydrotestosterone (DHT), 100 nM dexamethasone (Dex), 1000 nM cyproterone acetate (CPA), 100 nM mifepristone (RU486) and combination of 100 nM DHT,100 nM Dex for 24 hours.

Project description:Prostate cancer is dependent on androgen receptor (AR) signaling at all stages of the disease and cyclin D1 has been shown to negatively modulate the expression of the AR-dependent gene prostate specific antigen (KLK3/PSA). Unbiased gene expression profiling was performed to determine the androgen-regulated geneset that is sensitive to cyclin D1. Androgen-sensitive LNCaP cells were straved of androgen, then transduced with control green fluorescent protein (GFP) or cyclin D1 (CCND1) adenovirus, then subsequently stimulated with control (Ethanol) or dihydrotestosterone (DHT).
