Gene expression analysis of bovine oocytes at optimal coasting time combined with GnRH antagonist during the no-FSH period

ABSTRACT: The objective of the study was to analyze the effect of a GnRH antagonist used during the FSH withdrawal period (coasting) on transcriptome changes in in vivo bovine oocytes at the GV stage. Oocytes were collected from super-stimulated animals after a coasting duration of 68hrs (optimal condition, control) and also from the same animal who received the GnRH antagonist (treated) during the whole coasting period.

Overall, 6 hybridizations were done, corresponding to the three biological replicates and one comparison using a dye-swap set-up.

Project description:The objective of the study was to analyze the impact of various lenght FSH withdrawal period (coasting) after the last FSH injection on transcriptome changes in in vivo bovine oocytes at the GV stage. Oocytes were collected from super-stimulated animals after 20, 44, 68 and 92 hours of coasting. For each cow individually (biological replicate), each coasting time was compared to others (i.e. 20 hrs vs 44 hrs; 20 hrs vs 68 hrs; 20 hrs vs 92 hrs; 44 hrs vs 68 hrs; 44 hrs vs 92 hrs and 68 hrs vs 92 hrs) for a total of six comparisons. Overall, 36 hybridizations, corresponding to the three cows and six comparisons were done using a dye-swap set-up.

Transcription profiles of cumulus cells from GnRH agonists and GnRH antagonists treated oocytes at different maturity stages

Project description:Affymetrix gene expression profiling in cumulus cells (CC) retrieved from patients undergoing GnRH agonists and GnRH antagonists IVF treatment. Oocytes from three different maturity stages were considered: metaphase I oocytes (MI), nonfertilized metaphase II (MII) oocytes (MII-NF) and MII oocytes developed to blactocyst stage embryo (MII-BL). From 4 GnRH agonist treated patients, CC MI, CC MII-NF and CC MII-BL samples were collected; from 5 GnRH agonist and 6 GnRH antagonist treated patients, CC MII-NF and CC MII-BL samples were collected; and from 2 GnRH agonist and 4 GnRH antagonist treated patients, CC MI and CC MII-BL were collected. Altogether, 10 CC MI, 15 CC MII-NF and 21 CC MII-BL were collected and considered for transcriptome analysis.

Increased AIF-1-mediated TNF-α expression during the implantation phase in IVF cycles with GnRH antagonist protocol

Project description:Administration of GnRH antagonist in IVF has several advantages. However, studies have shown that GnRH antagonist protocol cycles have lower implantation and clinical pregnancy rates than GnRH agonist long protocol cycles. Endometrial receptivity rather than embryo quality is thought to account for this phenomenon, however the mechanism is still largely unknown. This microarray analysed human endometrium during ‘implantation window phase’ between three groups: 1) untreated control, 2) GnRH agonist long protocol, and 3) GnRH antagonist protocol. The differential gene between groups uncovered the molecular mechanism occurred in endometrium affected by the intake of GnRH agonist or GnRH antagonist. Further study of these differential gene promises to find the reasons for lower endometrial receptivity in patients treated with GnRH antagonist and to improve the implantation rate in GnRH antagonist protocols in IVF.

Project description:Transcriptional profiling of cows granulosa cells comparing granulosa cells from superstimulated control cows with granulosa cells from superstimulated cetrotide treated cows. Abstract:In cow, follicular growth stimulation (FSH) followed by a 44h-68h coasting (no FSH) period has been shown to be beneficial for developmental competence of aspirated oocytes. This process has been optimized but the associated physiology is not perfectly understood. During the above treatment LH pulse frequency is basal as the animal are in the luteal phase. The goal of this study was to test the existence of a beneficial effect of basal LH pulses during competence acquisition. To answer to this question, LH pulses have been pharmacologically inhibited during the optimal coasting period with GnRH antagonists. Commercial milking cycling cows (n=5) were stimulated with 3 days of FSH followed by a coasting period of 68H, with and without cetrorelix injections. At the scheduled time, transvaginal aspirations of immature oocytes were performed, followed by IVF while follicular cells were pooled for each animal and were snap-frozen. Granulosa cells RNA was extracted amplified and hybridized on Embryogene bovine microarray slides. Data were normalized with Loess, and analyzed with Limma procedure. Follicle size group proportions and granulosa cell transcriptome were studied. (1) There was a significant reduction in >10mm diameter follicles with cetrorelix comparing to without treatment. (2) Transcriptome analysis reveals 724 tanscripts potentially regulated by basal gonadotropins before LH surge. Among this LH influenced gene group, 84 transcripts were positively associated to developmental competence. This work allows the identification of subpopulations of gene signals dependant from LH or not, and permits a better comprehension of the implication of basal LH in follicle growth and in competence acquisition process.
