ABSTRACT: Jasmonates is inductively produced as a major plant hormone responsible for defense reactions in plants against both biotic and abiotic stresses, such as pathogen infection and mechanical wounding. Jasmonoyl isoleucine is known to be a bioactive compound of jasmonate and plays a pivotal role for plant defenses. We identified OsJAR1−related JA-inducible genes in osjar1 tos17 mutant (osjar1-2) rice leaves 0 - 2 h after JA treatment using 44k microarray.

CUL3(BPM) E3 ubiquitin ligases regulate MYC2, MYC3 and MYC4 stability and JA responses

Project description:The Jasmonate pathway regulators MYC2, MYC3 and MYC4 are central nodes in plant signaling networks integrating environmental and developmental signals to fine-tune jasmonate defenses and plant growth. Hence, their activity needs to be tightly regulated in order to optimize plant fitness. Among the increasing number of mechanisms regulating MYCs, protein stability is arising as a major player. However, how the levels of MYCs proteins are modulated is still poorly understood. Here, we report that MYC2, MYC3 and MYC4 are targets of BPM proteins, which act as substrate adaptors of CUL3-based E3 ubiquitin ligases. Reduction-of-function of CUL3(BPM) in amiR-bpm lines, bpm235 triple mutants and cul3ab double mutants enhance MYC2 and MYC3 stability and accumulation, and potentiates plant responses to JA such a root-growth inhibition, and MYC-regulated gene expression. BPM3 protein is stabilized by JA, suggesting a new negative feedback regulatory mechanism to control MYCs activity. Our results uncover a new layer for JA-pathway regulation by CUL3(BPM)-mediated degradation of MYC TFs.

An evolutionarily ancient Fatty Acid Desaturase is required for the synthesis of hexadecatrienoic acid, which is the main source of the bioactive jasmonate in Marchantia polymorpha

Project description:Jasmonates are fatty acid derived hormones that regulate multiple aspects of plant development, growth and stress responses. Bioactive jasmonates differ between vascular plants and bryophytes (using jasmonoyl-L-isoleucine; JA-Ile and dinor-12-oxo-10,15(Z)-phytodienoic acid; dn-OPDA, respectively), but bind an evolutionarily conserved COI1 receptor. Whilst the biosynthetic pathways of JA-Ile in the model vascular plant Arabidopsis thaliana have been elucidated, the details of dn-OPDA biosynthesis in bryophytes are still unclear. Here, we identify an ortholog of Arabidopsis Fatty Acid Desaturase 5 (AtFAD5) in the model liverwort Marchantia polymorpha and show that FAD5 function is ancient and conserved between species separated by more than 450 million years of independent evolution. Similar to AtFAD5, MpFAD5 is required for the synthesis of 7Z-hexadecenoic acid. Consequently, in Mpfad5 mutants the hexadecanoic pathway is blocked, the dn-OPDA levels almost completely depleted and normal chloroplast development is impaired. Our results demonstrate that the main source of dn-OPDA in Marchantia is the hexadecanoic pathway and the contribution of the octadecanoid pathway, i.e. from OPDA, is minimal. Remarkably, despite extremely low levels of the bioactive hormone (dn-OPDA), MpCOI1-mediated responses to wounding and insect feeding can still be activated in Mpfad5 mutants, suggesting that dn-OPDA is not the only bioactive jasmonate and COI1 ligand in Marchantia.

Project description:The plant hormone jasmonic acid (JA) has been known as a signal molecule that is induced by various stresses and mediates plant defense responses. Rice O. sativa inductively produces variety of defensive compounds upon abiotic and biotic stress conditions, such as wounding and insect attack. The bHLH transcription factor RERJ1 has previously been identified as JA-inducible factor whose expression is also rapidly induced by wounding. We identified RERJ1-dependent and wound-inducible genes by comparison with transcriptomes of wound treated wild-type and a Tos17-rerj1 defective mutant rice.

Identification of JA/COI1-regulated genes in tomato reproductive tissue and JA-treated roots.

Project description:Jasmonic acid (JA) is a fatty-acid derived signaling molecule that regulates a wide variety of plant developmental and stress-related responses. We identified a jasmonate-insensitive1 (jai1) mutant of tomato that is defective in the LeCOI1 gene, which plays an essential role in induced resistance of tomato to a broad spectrum of pests, and is also required for maternal control of seed and fruit development. The present study is aimed at identifying genes involved in two JA-signaled processes that have not been studied in other plants. The first specific aim is to identify the set of JA/COI1-regulated genes in tomato reproductive tissues, with emphasis on genes expressed in maternal tissues. We hypothesize that JA/COI1 signaling regulates maternal processes that nurture the developing embryo and seed. The second aim of the study is to identify JA/COI1-regulated genes that are specifically expressed in tomato roots. Although it has been shown that JA promotes root defenses against various pests, virtually nothing is known about the JA-regulated transcriptome in roots. Tomato (Solanum lycopersicum) cv Micro-Tom wild-type and jai1 seed were grown in peat pots and maintained in growth chambers. To identify COI1-regulated genes in reproductive tissues, flower buds, open flowers and immature fruit at 7 and 17 days post-pollination were collected from 8 to 10 week old plants. To identify COI1-regulated genes in roots, 3 week old plants were exposed to exogenous JA by a single soil drench with 0.1mM JA, and roots were collected after 1, 6, 12 and 24 hours. Plants treated with water were used for the 0hr time point. All tissue was snap-frozen in liquid nitrogen, and RNA was extracted and analyzed as described in the TIGR Potato Microarray protocol (phenol method). JA-INDUCIBLE MYC2-LIKE1 acts as a repressor and negatively regulates jasmonate signaling in Arabidopsis

Project description:Jasmonate (JA) is a plant hormone that controls trade-offs between plant growth and responses to biotic and abiotic stresses. Although recent studies uncover core mechanism for JA-induced responses in Arabidopsis thaliana, it remains elusive how plants attenuate those responses. We report here that a basic-helix-loop-helix type transcription factor named JA-INDUCIBLE MYC2-LIKE1 (JAM1) acts as a transcriptional repressor and negatively regulates JA signaling. Arabidopsis plants expressing the chimeric repressor for JAM1 exhibited a substantial reduction of JA responses, including JA-induced inhibition of root growth, accumulation of anthocyanin, and male fertility. These plants were also compromised in resistance to attack by Spodoptera exigua. Conversely, jam1-4 loss-of-function mutants showed enhanced JA responsiveness, including increased resistance to the insect attack. Competitive binding of JAM1 and MYC2 to the target sequence of MYC2 suggested negative regulation of JA signaling by JAM1 and suppression of MYC2 function. These results indicate that JAM1 plays a pivotal role in fine-tuning of JA-mediated stress responses and plant growth by negatively regulating JA signaling.
