ABSTRACT: The objective of this study was to identify porcine genes which expression was affected by experimental infection with A. pleuropneumoniae within 24 hours after experimental challenge. Project description:This study aimed at: (1) Establishing the pleuropneumoniae pathological model of artificial infection; (2) Using the Agilent swine genome-wide microarray containing 43,603 probes to detect the changes in gene expression of infected pigsM-bM-^@M-^Y lungs and hilar nodes. Infection induced gene expression in swine lung and hilar node were measured at 48 hours after being inoculated with 1 mL containing 3.8 M-CM-^W 10^7 cfu/mL pleuropneumoniae serotype I by atomizing inhalation in each nostril.

Genome wide transcriptional portrait of Actinobacillus pleuropneumoniae during invasive disease reveals new strategies for survival and persistence in the host

Project description:The transcriptional profile of the porcine lung pathogen, Actinobacillus pleuropneumoniae, was monitored during the acute phase of infection in its natural host. Bacterial expression profiles of A. pleuropneumoniae isolated from lung lesions of 25 infected pigs were compared in samples taken 6, 12, 24 and 48 hours post infection. A 84 chip study using total RNA recovered from A. pleuropneumoniae serotype 2 (4226) and serotype 6 (7712640) isolated from infected pig lung tissue during the first 48 of infection. Samples were taken 6, 12, 24 and 48 hours post infection, respectively. Before hybridization the samples were enriched for bacterial RNA and submitted to linear amplification. Each chip measures the expression level of 4,876 target genes from A. pleuropneumoniae with each gene covered by an average of 26.7 probes. Three biological replicates per sample. Microarray data from 3 pigs (no. 39, no. 43 and no. 72) were omitted from the final analysis due to poor signal intensity. A total of 75 microarrays were included in the final analysis.

Global transcriptional profiling in porcine liver and lymph node tissues during acute pleuropneumoni

Project description:Expression profiles of porcine liver and tracheobronchial lung lymph node tissues were studied to identify genes being significantly affected by bacterial infection of the lungs. Samples were taken from liver and tracheobronchial lung lymph node tissues in pigs experimentally infected with A. pleuropneumoniae, serotype 5b, and from healthy non-inoculated control pigs from the same herd. Samples were investigated for changes in gene expression by means of global cDNA microarrays. Expression profiling of samples of liver and lung lymph node tissues from ten pigs challenged with Actinobacillus pleuropneumoniae and from five healthy control pigs was conducted by hybridising all 2x15 samples against a tissue specific common reference consisting of a pool of equal amount of total-RNA from all samples of each tissue type and were balanced with respect to control and challenged animals.

Hepatic transcriptional profiling of mRNA and microRNA in pigs infected with Actinobacillus pleuropneumoniae

Project description:Actinobacillus pleuropneumoniae is a gram-negative bacterium that causes porcine pleuropneumonia, which is a widespread, highly contagious, and often fatal respiratory disease in swine. In this experiment pigs were inoculated with A. pleuropneumoniae serotype 5b. Liver samples from three non-inoculated pigs and three experimental inoculated pigs were used to characterize the expression profiles of mRNA and microRNA genes using DNA microarrays and Illumina GA deep sequencing, respectively. The microarray analysis identified a large number of genes, which significantly differed in expression in infected versus non-infected animals. MicroRNAs are short single stranded RNA molecules that regulate gene expression by sequence specific binding to the 3´ untranslated region (3´UTR) of target mRNAs. The deep sequencing analysis determined the identity and abundance of nearly 400 microRNAs, of which a portion was found to significantly differ in expression between the infected and non-infected animals. Target genes for differentially expressed microRNAs were predicted using microCosm Targets, which is based on the miRanda algorithm. Comparison on the two gene lists showed many common genes, which may suggest a causative relationship between changes in microRNA expression and target gene expression.

Actinobacillus pleuropneumoniae during a natural infection in pigs

Project description:Actinobacillus pleuropneumoniae is the etiological agent of porcine pleuropneumonia, a respiratory disease which causes great economic losses worldwide. Many virulence factors are involved in the pathogenesis, namely capsular polysaccharides, RTX toxins, LPS and many iron acquisition systems. In order to identify genes that are expressed in vivo during a natural infection, we undertook transcript profiling experiments with an A. pleuropneumoniae DNA microarray, after recovery of bacterial mRNAs from serotype 5b-infected porcine lungs. Comparative Genomic Hybridizations between Actinobacillus pleuropneumoniae serotype 5b strain L20 (ref) and serotype 5b fresh field isolate 896-07, recovered from infected pig lung tissues following natural acute infection. Two condition transcript profiling experiments : infectious 5b field strain isolated directly from lungs of naturally deceased pigs after acute infection vs infectious 5b field strain grown in BHI broth to an OD600 of 0.300.

Project description:Actinobacillus pleuropneumoniae is the etiologic agent of contagious pleuropneumonia, an economically important disease of commercially reared swine throughout the world. To cause this disease, A. pleuropneumoniae must rapidly overcome porcine pulmonary innate immune defenses. Since bronchoalveolar fluid (BALF) contains many of the innate immune components found in the lung, we examined the gene expression of a virulent serovar 1 strain of A. pleuropneumoniae after exposure to concentrated BALF. This experiment was also carried out with a malT mutant of the same strain.
