Project description:Epilepsy accompanying cognitive impairment has been verified by accumulating clinical cases, but the mechanism remains unclear. Here, we discover that persistent epileptic seizures impaired the ability of mice to recognize either novel objects or novel locations at 6 months old but not at 2 months old by utilizing the spontaneous epilepsy model of Cdh5-CreERT2; CDK5f/f mice after tamoxifen treatment. To the best of our knowledge, this is the first report that the levels of synapse-related proteins, such as NMDA receptors (NR1 and NR2B), PSD95, and phosphorylation of CaMKII, are progressively decreased during epileptic seizures in the hippocampus of spontaneous epileptic mice. Notably, we also found that valproate (VPA) augment synapse-related genes and protein expression and ameliorate progressive cognitive impairment. Hence, our study describes a mechanism of cognitive deficits in epilepsy and identifies new effects of VPA on synapses, which provides new insights into preventive or therapeutic interventions for epileptic cognitive deficits.

Project description:This study included four dog groups (group A: 10 healthy dogs, group B: 9 dogs with idiopathic epilepsy receiving antiepileptic medication-AEM, group C: 8 dogs with idiopathic epilepsy without AEM administration and group D: 7 dogs with seizures due to structural brain abnormalities). The epileptic dogs were allocated into these 3 groups after a diagnostic investigation that included laboratory testing, thoracic radiographic and abdominal ultrasonographic examination, brain imaging, CSF routine and proteomic analysis. The purpose of the current study was to investigate and compare the proteomic profile among the four groups of dogs.

Project description:<h4><strong>INTRODUCTION: </strong>Approximately 1% of the world's population is impacted by epilepsy, a chronic neurological disorder characterized by seizures. One-third of epileptic patients are resistant to AEDs, or have medically refractory epilepsy (MRE). One non-invasive treatment that exists for MRE includes the ketogenic diet, a high-fat, low-carbohydrate diet. Despite the KD's success in seizure attenuation, it has a few risks and its mechanisms remain poorly understood. The KD has been shown to improve metabolism and mitochondrial function in epileptic phenotypes. Potassium channels have implications in epileptic conditions as they have dual roles as metabolic sensors and control neuronal excitation.</h4><h4><strong>OBJECTIVES: </strong>The goal of this study was to explore changes in the lipidome in hippocampal and cortical tissue from Kv1.1-KO model of epilepsy.</h4><h4><strong>METHODS: </strong>FT-ICR/MS analysis was utilized to examine nonpolar metabolome of cortical and hippocampal tissue isolated from a Kv1.1 channel knockout mouse model of epilepsy (n = 5) and wild-type mice (n = 5).</h4><h4><strong>RESULTS: </strong>Distinct metabolic profiles were observed, significant (p < 0.05) features in hippocampus often being upregulated (FC ≥ 2) and the cortex being downregulated (FC ≤ 0.5). Pathway enrichment analysis shows lipid biosynthesis was affected. Partition ratio analysis revealed that the ratio of most metabolites tended to be increased in Kv1.1-/-. Metabolites in hippocampal tissue were commonly upregulated, suggesting seizure initiation in the hippocampus. Aberrant mitochondrial function is implicated by the upregulation of cardiolipin, a common component in the mitochondrial membrane.</h4><h4><strong>CONCLUSION: </strong>Generally, our study finds that the lipidome is changed in the hippocampus and cortex in response to Kv1.1-KO indicating changes in membrane structural integrity and synaptic transmission.</h4>

Project description:We carried out Massive Parallel Sequencing (MPS) to reveal miRNA expression profiles of rats with LiCl-pilocarpine induced epileptic status (SE) in adulthood (Postnatal day 60 -P60) and infancy (P12). Hippocampal tissues were collected at 3 stages of epileptogenesis: acute (24 hours), latent (7 days) and chronic (3 months after SE) from 10 animals after SE and 10 controls in each age group (120 in total). The sequencing platform identified 666 miRNA species, out of which up to 419 miRNAs were present with the coverage of more than 500 reads altogether in individual groups (onset-age epilepsy stage combinations). We discovered that age of SE induction has impact on miRNA profile throughout all stages of epileptogenesis and infantile-onset of epilepsy leads to changes in smaller number of miRNAs.
