RNA was isolated from 2 separate lungs (labeled as A or B) for each time point and 2 technical replicates were run for each sample (labeled as 1 or 2).

Project description:This project aims to establish the embryonic anole lung as a model for investigating lung development and more general epithelial morphogenesis by generating a set of single-cell RNA-seq (scRNA-seq) data from late-stage embryonic lungs taken from brown anoles (Anolis sagrei). The anole lung, with its simple architecture, provides a novel tool for investigating signaling in a less complex respiratory system than the murine lung, and this data set would be a large advance in making this system more widely known and accesible among developmental biologists.

Adult virgin female BALB/C mice were subjected to unilateral ureter obstruction (UUO) to induce kidney fibrosis and the fibrotic kidney was harvested for RNA extraction. Adult virgin female BALB/C mice were subjected to intratracheal administration of bleomycine to induce lung fibrosis and the fibrotic lungs was harvested for RNA extraction.

Project description:Metastasis is a complex process involving multiple steps. We were interested in the role of microRNAs (miRNAs) in the process of liver colonization by colorectal cancer cells. We hypothesized that the comparison between non-metastatic versus metastatic isogenic cell line should thus offer valuable insight to the molecular mechanisms involved in developing metastatic behavior. KM12C/KM12SM and SW480/SW620 are probably the best available models of isogenic cell lines differing in metastatic properties for colorectal cancer. Our first goal was to identify miRNAs that contribute to the metastatic traits of the isogenic colorectal cancer cell lines, KM12C/KM12SM and SW480/SW620. Total RNA was extracted from cells using the mirVana kit (Ambion). Total RNA (1 µg) from KM12C and SW480 (poorly metastatic) and KM12SM and SW620 (highly metastatic) cells was used to analyze the global miRNA expression profiling with TaqMan Megaplex human array A (v2.0) and B (v3.0) (Applied Biosystems).

Project description:Compare miRNA expression profiles in mouse CD4 T cells overexpression of Bcl-6 vs empty vector Mouse naïve CD4+ T cells were transduced with a Bcl-6-expressing retrovirus or the control vector and cultured in Th0 condition for 2 days. Cells expressing medium to high levels of GFP were sorted and total RNAs were isolated using mirVana miRNA Isolation Kit (Applied Biosystems). MiRNA expression was profiled using Agilent 8x15K mouse miRNA microarray miRNA-v1_95_May07 (miRBase Release 9.2) by Ramaciotti Centre for Gene Function Analysis (Sydney). Each sample was run in duplicate.

Project description:Some chronic myeloid leukemia (CML) patients with complete molecular response (CMR) are considered able to sustain the CMR after imatinib discontinuation (STOP-IM). Mahon et al. reported that among patients with a CMR lasting at least 2 consecutive years, the CMR was sustained in 41% after imatinib discontinuation. To more appropriately identify patients who can safely discontinue imatinib, we assessed the miRNA profiles of CML patients. We compared CML patients who sustained CMR for more than 6 months after discontinuation of imatinib (STOP-IM group) with those who were receiving imatinib with CMR (currently called as UMD: undetermined minimal disease), and with healthy volunteers (controls). Peripheral blood mononuclear cells (PBMCs) were harvested form 10ml of whole blood. Isolation of total RNA was performed using the mirVana PARIS kit (Ambion, Austin, TX, USA). The expression profile of miRNAs was determined using the Human Taqman miRNA Arrays A (Applied Biosystems). RNU6B were used as acontrol. QRT-PCR was carried out on an Applied Biosystems 7900HT thermal cycler using the manufacturerM-bM-^@M-^Ys recommended program. Finally, all the raw data from each array was run on Data Assist Software ver.3.1 (Applied Biosystems).

Project description:2 IL-15 KO AND 2 C57BL/6 mice were injected with 5X10^5 cells of the polyoma Middle T (pMT) primary breast tumor cell line intraveneously(IV). 2 days post injection, the lungs were harvested and one lobe of the lung was used to extract RNA from. We then utilized the SABBiosciences RT^2 Profiler PCR Array for Mouse cytokines and Chemokines to determine levels of cytokine/chemokines that were different in these 2 mice at this time point. Project description:Balb/cJ mouse received control RNAi or RNAi to 8-oxoguanine DNA glycosylase (Ogg1) intranasally prior to the exposure for 1 h to a) glucose oxidase (1 mU) to induce OGG1-BER or b) OGG1-BER product 8-oxoguanine (1 M-NM-<M). We used SABiosciences Mouse Inflammatory Cytokines & Receptors PCR Array (PAMM-011A) to quantitate inflammatory gene expression dependent on OGG1 and its product 8-oxoguanine. After intranasal challenge with glucose oxidase or 8-oxoguanine, mice were sacrificed and lungs were collected and processed to obtain RNA. Total pooled (n=5) RNA (1 M-NM-<g) was reverse transcribed into cDNA using SuperscriptM-BM-. III First Strand Synthesis System (Invitrogen), mixed with equal amounts of 2X SYBR Green Supermix (Qiagen) and 20 M-NM-<l of reaction mixture was added to each well of the PAM-011A array. The reaction was evaluated using an ABI PRISMM-BM-. 7000 Sequence Detection System using recommended settings by SABiosciences.
