Differential gene expression in salicylic acid-deficient transgenic plants overexpressing StTGA2.1 after viral infection.

Project description:The general aim was to better understand the role of StTGA2.1 in plant immunity by transcriptional profiling of plants overexpressing the salicylic acid pathwas related TGA transcription factor StTGA2.1 after infection with potato virus Y (PVY). Potato non-transgenic cultivar Rywal (NT), Rywal-NahG (NahG), a transgenic line unable to accumulate SA due to salicylate hydroxylase expression, and a transgenic Rywal-NahG line inducibly overexpressing StTGA2.1 (TGA2.1-NahG) using the glucocorticoid-system, in which target gene expression can be controlled by external application of dexamethasone (DEX), were used. The leaves of three plants per genotype were infected with either PVY or mock-inoculum and subsequently treated with DEX. Additionally, three TGA2.1-NahG PVY-infected or mock-inoculated plants were not treated with DEX as control. After four days, PVY infection symtoms appeared as small regions of necrotic tissue called lesions. Tissue sections, containing lesions and their immediate surrounding area, were sampled, allowing us to focus on PVY-affected cells. Total RNA was isolated, DNase treated and sent for library prep and high-throughput sequencing.

Project description:RNA-Seq of mock and PVYN-Wilga inoculated samples to study the transcriptional response of potato to viral infection in the cell death zone (pool A) and surrounding tissue (pool B) in different genetic backgrounds. Potato cv. Rywal reacts to the virus by hypersensitive response (HR), where spot necroses that limit the virus at the site of virus entry are formed. NahG-Rywal plants, compromised in salicylic acid accumulation, develop necrotic lesions that do not limit viral spread and show delayed defense response. shRBOHD plant with silenced RBOHD gene are not able to arrest the virus spread.

Transcriptional changes in potato in response to defence hormones

Project description:Phytohormones are involved in diverse aspects of plant life including the regulation of plant growth, development and reproduction, as well as governing stress and defence responses. We have generated a comprehensive transcriptional reference map of the early potato responses to exogenous application of the defence hormones abscisic acid, brassinosteroid, ethylene, salicylic acid and jasmonic acid. Amongst microarray probes representing the 39000 predicted genes, a total of 3175 and 2873 probes were significantly differentially expressed at 1 h and 6 h after hormone treatment, respectively. Marker genes identified for the early hormone responses in potato include: a homeodomain 20 transcription factor (DMG400000248) for abscisic acid; a SAUR gene (DMG400016561) induced in Epibrassinolide treated plants at 6 hours; an osmotin gene (DMG400003057) specifically enhanced by the ethylene precursor aminocyclopropanecarboxylic acid; a gene weakly similar to AtWRKY40 (DMG402007388) that was induced by salicylic acid and; a jasmonate ZIM-domain protein 1 (DMG400002930) which was specifically activated by methyl jasmonate. An online database has been set up to query the expression patterns of potato genes represented on the microarray that can also incorporate future microarray or RNAseq-based expression studies.
