Genes involved in plucking induced hair regeneration

ABSTRACT: Intra-follicular signals are thought to be important in plucking induced hair regenration. We hypothesize that extra-follicular macro-environment (including adipose and dermal tissue) also participates in this process. By using microarray, we want to identify the activating signals emit from extra-follicular macro-environment.

Project description:The integration of positive and negative intra- and extra-cellular signals dictates whether a cell will proliferate or differentiate. While it is intuitive to speculate that nutrients availability may influence this alternative, a comprehensive complement of the molecular determinants involved in this process has not been elucidated yet. In this study, we will investigate how nutrients (glucose) affect skeletal myogenesis. C2C12 cells will be cultured in high glucose and low glucose conditions, and their differenciation will be studied. Experiment Overall Design: We hypothesize that calorie restriction may have an effect on appropriate differentiation of skeletal myoblasts and this effect can be demonstrated by expression profiling.

The role of aV Integrin in regulating macrophage phenotype and function during cardiac remodeling after myocardial infarction

Project description:Macrophages infiltrate the infarcted heart and play a critical role in repair, remodeling and fibrosis. Macrophages sense changes in the extracellular matrix (ECM) environment through Integrins, thus activating signaling pathways that regulate their function. Our data show that av integrin is highly expressed in isolated bone marrow macrophages and is markedly upregulated in response to TGF-β, a growth factor known to be activated in the infarcted heart. Accordingly, we hypothesize that aV integrin induction in infarct macrophages may regulate macrophage phenotype, function and response to key macrophage-activating signals following MI.

Light- and plastid-regulated transcriptomes in Arabidopsis seedlings

Project description:Plastids emit signals that broadly affect cellular processes. Based on previous genetic analyses, we propose that plastid signaling regulates the downstream components of a light signaling network and that these interactions coordinate chloroplast biogenesis with both the light environment and development by regulating gene expression. We tested these ideas by analyzing light-regulated and plastid-regulated transcriptomes. We found that the plastid is a major regulator of light signaling, attenuating the expression of more than half of all light-regulated genes in our dataset and changing the nature of light regulation for a smaller fraction of these light-regulated genes. Our analyses provide evidence that light and plastid signaling are interactive processes and are consistent with these interactions serving as major drivers of chloroplast biogenesis and function. Four biological replicates were grown separately under the same conditions. Arabidopsis seedlings were grown in the presence (+Lin) or absence (-Lin) of lincomycin in 0.5 µmol m-2 s-1 blue plus red (BR) light for 6 days. After 6 days of growth in 0.5 µmol m-2 s-1 of BR light, seedlings were transferred to 60 µmol m-2 s-1 BR light. 50-100 seedlings were collected before (0 h) and 0.5 h, 1 h, 4 h, and 24 h after the 0.5 to 60 µmol m-2 s-1 BR-fluence-rate shift for RNA extraction and hybridization on Affymetrix microarrays.

Estimating essential phenotypic and molecular traits from integrative biodiversity data

Project description:<p>Functional ecology and biodiversity lack integrative reference data that combine the assessment of traits at different levels. Here, we integrate data from 16 field samples of complex thallose liverworts (order Marchantiales) collected from biological soil crust communities in Southern Sweden and Germany at three levels: (1) bioimaging (morphometric measurements from macro- and microscopy), (2) metabolomics (molecular computations performed on liquid chromatography high-resolution mass-spectrometry (UPLC/ESI-QTOF-MS) with data-dependent acquisition (DDA-MS) data) and (3) DNA marker sequencing. These data are used to construct a reference framework including a proposed naming scheme for the estimation of molecular traits and for demonstrating the systematic and standardized extraction of phenotypic and molecular traits for integration into the plant trait database TRY. Phylogenetic trees and partitioning around medoids are used to demonstrate the assessment of evolutionary relationships and the trait space connecting ecological hypotheses and documenting knowledge gains across domains. With our reference framework we want to encourage the combined assessment, reuse and integration of phenotypic and molecular traits into functional biodiversity research and related disciplines.</p>

Gene expression variation increase in trisomy 21 tissues

Project description:Congenital development disorders with variable severity occur in trisomy 21. However, how these phenotypic abnormalities develop with variations remains elusive. We hypothesize that the difference in euploidy gene expression variation among trisomy 21 tissues are perturbed by the presence of an extra copy of chromosome 21 and this may contribute to the phenotypic variations in Down syndrome.
