Genes involved in plucking induced hair regeneration

ABSTRACT: Intra-follicular signals are thought to be important in plucking induced hair regenration. We hypothesize that extra-follicular macro-environment (including adipose and dermal tissue) also participates in this process. By using microarray, we want to identify the activating signals emit from extra-follicular macro-environment.

Global profiling of human hair follicle scalp dermal papilla cells using Affymetrix microarrays

Project description:Dermal papilla cells isolated from the human hair follicle are capable of inducing hair growth in recipient epithelia. However, demonstrating disparity from rodent dermal papilla, human cells lose this inductive competance immediately upon growth in culture under normal growth conditions. We grew dermal papilla cells in hanging drop cultures that are morphologically akin to intact dermal papilla, and found that by enhancing the environment for aggregation, we could restore the inductive capacity of human dermal papilla cells in culture. The underlying genes that regulate the inductive potential of dermal papilla cells is not well understood, and we sought to use global profiling to identify key genes and pathways related to inductive competance within dermal papilla cells. We used Affymetrix microarrays to profile human dermal papilla cells in both hair inducing, and non-hair inducing states. Affymetrix microarrays were used to to perform profiling of human dermal papilla cells, both as intact tissues (freshly isolated from scalp), and at several stages in subsequent two dimensional culture; cell explant outgrowths (p0), cells at passage 1 (p1), passage 3 (p3) and passage 5 (p5). RNA was isolated from cultured cells 72 hours after feeding. Cells at passage 3 were also grown in hanging drops to form dermal spheroids, that were used for RNA collection 48 hours after establishment. Transcription profiling of mouse hair follicles in the 2nd telogen to anagen transition

Project description:Hair Follicle regeneration relies on both epithelial components (bulge and hair germ cells) and a mesenchymal one (dermal papilla cells). We used microarrays to detail the global programme of gene expression underlying organ regeneration at the transition between quiescent stages (early and middle telogen) and the initiation of a new growth (late telogen). Experiment Overall Design: These microarray at the 3 different stages were designed to identify signals released by the mesenchymal dermal papilla cells to activate epithelial growth, their target genes in the hair germ and bulge compartments, and to get at gene signature differences and similarities between hair germ and bulge cells.

Single-cell RNA-seq of mouse androgenetic alopecia models treated with CXCL12-neutralizing antibody

Project description:Androgenetic alopecia (AGA) is a common form of hair loss characterized by follicular miniaturization, dermal fibrosis, and low-grade immune activation, yet the precise mediators linking androgen signaling to these tissue-level changes remain poorly defined. Here, we use single-cell RNA sequencing (scRNA-seq) to map the skin microenvironment in a testosterone-induced mouse model of AGA and assess the therapeutic impact of a CXCL12-neutralizing antibody. We find that androgen receptor-positive dermal fibroblasts are the principal source of CXCL12, which acts in an autocrine manner via ACKR3 to drive TGF-β–mediated extracellular matrix deposition. In parallel, CXCL12 signals through CXCR4 to reprogram a Sox2+ Twist1+ dermal papilla cell population toward a fibrotic, low-inductive state and expand a pro-fibrotic Trem2+ macrophage subset. Blockade of CXCL12 reverses these transcriptional and cellular alterations across stromal, follicular, and immune compartments, restoring a regenerative skin niche and supporting hair regrowth. These findings position CXCL12 as a central mediator of fibroimmune remodeling in AGA and establish its neutralization as a promising therapeutic strategy.

Light- and plastid-regulated transcriptomes in Arabidopsis seedlings

Project description:Plastids emit signals that broadly affect cellular processes. Based on previous genetic analyses, we propose that plastid signaling regulates the downstream components of a light signaling network and that these interactions coordinate chloroplast biogenesis with both the light environment and development by regulating gene expression. We tested these ideas by analyzing light-regulated and plastid-regulated transcriptomes. We found that the plastid is a major regulator of light signaling, attenuating the expression of more than half of all light-regulated genes in our dataset and changing the nature of light regulation for a smaller fraction of these light-regulated genes. Our analyses provide evidence that light and plastid signaling are interactive processes and are consistent with these interactions serving as major drivers of chloroplast biogenesis and function.

We uncovered a hitherto unrecognized DP transmitter, TGFβ2, which activates Smad2/3 transiently in HFSCs concomitant with entry into tissue regeneration. We used microarrays to detect the genes specifically affected by TGFß receptor II-deficient mice upon HFSC activation.
