Genome wide analysis of genes expressed in late G1/S phase of the cell cycle in PMCA4 knockout (P4KO) and P4KO rescued with PMCA4a primary vascular smooth muscle cells

ABSTRACT: Husain lab cell proliferation in vitro experimental data showed that electroporating PMCA4a cDNA into P4KO cells rescued the G1 arrest phenotype in P4KO primary vascular smooth muscle cells (VSMC). This microarray documents the different gene expression profiles of P4KO and P4KO+PMCA4a VSMC at the late G1/S stage in serum synchronized cell populations (n=4 populations for each genotype).

This microarray documents the different gene expression profiles of PMCA4 wild type and knockout cells at the early G1 stage in serum synchronized cell populations (n=4 populations for each genotype). Serum synchronized cell populations were Hoechst labeled and flow sorted for early G1 sub-populations (PMCA4 WT n=4; PMCA4 KO n=4) and total RNA from sorted cells was used for microarray analysis.

Study of transcriptional changes in MCM7-depleted and G1/S-arrested A549 and A375 cells

Project description:We tested if the changes in cell invasion and migration observed after MCM7 depletion manifested in an altered gene expression profile. For this purpose, we performed RNAseq analysis after MCM7 knock-down in A549 and A375 cells expressing a Dox-inducible shMCM7. As controls, we used shMCM7 cells that had not been induced with Dox, as well as the parental A549 and A375 cell lines grown in the presence or absence of Dox. To exclude possible effects caused by changes in cell cycle progression, all RNA samples were extracted from double thymidine synchronized cell populations, in which >98% cells were arrested at the G1/S phase boundary.

Kc167 cells were synchronized by first arresting at the G1/S transition with hydroxyurea (HU), and then releasing from the HU arrest point for 6 hours. Goal was to evaluate changes in the genome-wide Mcm2-7 distribution throughout the cell cycle, and how this relates to the excess Mcm2-7 loaded onto chromatin in G1.

Project description:In this report, we examine transcripts on a genome-wide level between the synchronized cell cycles of hESCs and hESC-derived endoderm cells. We found 10347, 10299 and 10362 genes expressed in the G1, G1/S, and S phase of hESCs, respectively; 10333, 10227 and 10215 genes expressed at the G1, G1/S and S phase of derived endoderm. We compared the transcriptome between these data sets and identified genes with differentiated phase expression between hESCs and hESC-derived endoderm cells.
