Profiling of genes related to cross protection and competition for NbTOM1 by HLSV and TMV

ABSTRACT: To investigate the mechanism under the cross protection between HLSV and TMV, microarray analysis was conducted to examine the transcriptional levels of global host genes during cross protection, using Tobacco Gene Expression Microarray, 4x44k slides. The transcriptional level of some host genes corresponded to accumulation level of TMV. Some host genes were up-regulated only by HLSV.

The transcriptional level of some host genes corresponded to accumulation level of TMV. Some host genes were up-regulated only by HLSV. The gene expression in Nicotiana benthamiana during cross protection was measured at 12 days post HLSV or mock buffer inoculated as well as TMV infected N. benthamiana which pre-inoculated HLSV or mock bufer at 3 dpi (which is equal to 15 dpi by HLSV).

The transgenic plants before resistance break (BRB-6 weeks), after resistance break (ARB-8 weeks) and wild type tobacco plants infected with TMV (TMVi-9weeks) leaves were analyzed. Tomato response to TMV after 1h of infection

Project description:TMV-resistant, TMV-susceptible and enhanced susceptibility mutant tomato were grown in asceptic conditions. At approximately 5 weeks, leaves were mock inoculated, TMV inoculated, or left untreated. Leaf tissue was collected 1hour after treatment. RNA was isolated and purified; hybridizations were performed using 130ug of total RNA from plants mock inoculated versus TMV inoculated and untreated. Each RNA sample is from two pooled plants. TMV-derived Small RNAs Reveals Role for Arabidopsis thaliana RNA-dependent RNA Polymerases in Viral siRNA Biogenesis

Project description:Using a crucifer-infecting strain of Tobacco Mosaic Virus (TMV-Cg) and Arabidopsis thaliana as a model system, we analyzed the viral small RNA profile in wild-type plants as well as rdr mutants by applying small RNA deep sequencing technology. Over 100,000 TMV-Cg-specific small RNA reads, mostly of 21- (78.4%) and 22-nucleotide (12.9%) in size and originating predominately (79.9%) from the genomic sense RNA strand, were captured at an early infection stage, yielding the first high-resolution small RNA map for a plant virus. The TMV-Cg genome harbored multiple, highly reproducible small RNA-generating hot spots that corresponded to regions with no apparent local hairpin-forming capacity. Significantly, both the rdr1 and rdr6 mutants exhibited globally reduced levels of viral small RNA production as well as reduced strand bias in viral small RNA population, revealing an important role for these host RDRs in viral siRNA biogenesis. In addition, an informatics analysis showed that a large set of host genes could be potentially targeted by TMV-Cg-derived siRNAs for posttranscriptional silencing, raising the interesting possibility for a hidden layer of widespread virus-host interactions that may contribute to viral pathogenicity and host specificity. Profiling of TMV-Cg derived small RNAs in systemically infected tissues of wild type (Col-0) Arabidopsis as well as the rdr1and rdr6 mutants, at 3 days post-infection.

Proteomic analysis of TMV and TMV 24A+UPD infected N. benthamiana plants by iTRAQ reveal differences in stress signalling, cell death and response to intracellular iron

Project description:TMV was mutated by introducing different lengths of internal poly (A) tract in front of the upstream pseudo knotted domain .Twenty four nucleotides were inserted into the TMV genome, this mutant is designated TMV 24A +UPD TMV 24+UPD shows more severe leaf necrosis and stem collapse, leading to plant death within a few days. The difference in rate of virus replication and plant death may be because the poly (A) tract introduced into TMV may interact with UPD and affect its interaction with host factors. To understand TMV 24A+UPD interaction with the host and the events leading to the altered pathogenesis, we opted to evaluate its proteome profile and compare it to TMV using Isobaric tag for relative and absolute quantitation( iTRAQ).
