Project description:Analysis of the differences between NPC derived astrocytes and Olig2PC derived astrocytes at gene expression level. Olig2PC-Astros show differences in functions and gene expression profiles from NPC-Astros in vitro, providing clues of superior neuroprotection effect of Olig2PC-astrocytes. Total RNA obtained from cultured NPC_astrocytes and Olig2PC_astrocytes. One hundred ng total RNA was used for amplification and hybridization to Illumina Human HT12_V4 chip according to the Manufacturer's instructions (Illumina). Array data were processed using Illumina GenomeStudio software (Illumina). Background was subtracted and arrays were normalized using quantile. Gene expression levels were considered significant only when their detection p-value ≤0.01.

Project description:Analysis of differential expression between NPC and non-NPC tissues at miRNA expression level. We used a miRNA microarray platform to investigate the profile between NPC tissues and normal nasopharyngeal tissues to evaluate the relationship between miRNA expression and NPC. Recognition of the aberrant miRNAs and enrichment pathway analysis of predicted target gene may provide some clues to estimate the mechanism of miRNA effects on NPC carcinogenesis mediated by miRNA-mRNA regulation. Total RNA was extracted from 8 NPC tissues compared to 4 nasopharyngeal tissues. We peformed the Illumina miRNA microarray to identify the differentially expressed miRNA between NPC tissues and normal nasopharyngeal tissues.

Differentiation of human fetal multipotential neural progenitor cells to astrocytes reveals susceptibility factors for JC Virus

Project description:Viral infections of the CNS are of increasing concern, especially among immunocompromised populations. Rodent models are often inappropriate for studies of CNS infection, as many viruses, including JC Virus (JCV) and HIV, cannot replicate in rodent cells. Consequently, human fetal brain-derived multipotential CNS progenitor cells (NPCs) that can be differentiated into neurons, oligodendrocytes, or astrocytes, have served as a model for CNS studies. NPCs can be non-productively infected by JCV, while infection of progenitor-derived astrocytes (PDAs) is robust. We profiled cellular gene expression at multiple times during differentiation of NPCs to PDAs. Several activated transcription factors show commonality between cells of the brain in which JCV replicates and lymphocytes in which JCV is likely latent. Bioinformatic analysis determined transcription factors that may influence the favorable transcriptional environment for JCV in PDAs. Functional astrocytes differentiated from hiPSCs

Project description:Growing evidence implicates the importance of glia, particularly astrocytes, in neurological and psychiatric diseases. Here, we describe a rapid and robust method for the differentiation of highly pure populations of astrocytes from human induced pluripotent stem cells (hiPSCs), via a neural progenitor cell (NPC) intermediate. Using this method, we generated hiPSC-derived astrocyte populations (hiPSC-astrocytes) from 42 NPC lines (derived from 30 individuals) with an average of ~90% S100β-positive cells. Transcriptomic analysis demonstrated that the hiPSC-astrocytes are highly similar to primary human fetal astrocytes and characteristic of a non-reactive state. hiPSC-astrocytes respond to inflammatory stimulants, display phagocytic capacity and enhance microglial phagocytosis. hiPSC-astrocytes also possess spontaneous calcium transient activity. Our novel protocol is a reproducible, straightforward (single media) and rapid (<30 days) method to generate homogenous populations of hiPSC-astrocytes that can be used for neuron-astrocyte and microglia-astrocyte co-cultures for the study of neuropsychiatric disorders.

Proteomic characterization and protective effect against brain ischemia produced by adult mouse neural progenitor cells-derived extracellular vesicles.

Project description:Hypoxia produced by stroke activates the proliferation of neural progenitor cells (NPC) in the subventricular zone of the adult mouse brain. Newly generated cells seem to participate in the protection of the ischemic brain through the release of extracellular vesicles (EVs) that might target different cell types and modify their function by delivering proteins, lipids, and nucleic acids. We investigated the possible communication route mediated by EVs between NPC and neurons under conditions of cerebral ischemia using an in vitro model of oxygen and glucose deprivation (OGD). We collected OGD-activated NPC-derived EVs by differential centrifugation of the conditioned media in which NPC were grown for 12 h after OGD, and as a control, we harvested EVs released by cells cultured under normoxic conditions. EVs were characterized by shape and size with transmission electron microscopy and NanoTracking assays, and the presence of typical exosome markers was assessed by western blot. The proteomic profiling of NPC-derived EVs was carried out after resolving the proteins by polyacrylamide gel electrophoresis and performed in-gel digestion with trypsin, then the identity of the products was determined by tandem mass spectrometry (ESI-IMS-MS). Overrepresentation analyses of the proteomic data allowed us to identify executioners of signaling pathways involved in neuroprotection. (Supported by DGAPA-PAPIIT IN207020 and CONACYT A1-S-13219).

RNA-seq of iPSC-derived astrocytes that are capable of exerting pro-maturational effect on synaptic networks

Project description:Astrocytes interact closely with neurons and facilitate neuronal maturation and function by providing trophic support, regulating the extracellular environment, and engaging in a variety of inter-cellular signalling mechanisms. We have described the generation of human astrocytes and neurons from a common cortical progenitor pool, thereby recapitulating aspects of in vivo development. Firstly, we show that the iPSC-derived astrocytes exhibit many of the key molecular and functional hallmarks predicted of astrocytes. Secondly, we provide functional and transcriptomic analyses of the astrocytes’ capacity to interact with neurons both through rapid regulation of ongoing synaptic transmission as well as exerting pro-maturational effects on the synaptic networks. Transcriptional analysis included the comparison of the present RNA-seq of iPSC-derived astrocytes to other iPSC-derived astrocyte datasets as well as fetal and adult human astrocytes. We investigated differences in gene expression between our maturation-enhancing iPSC-astrocytes and iPSC-astrocytes previously shown to be unable to promote synaptic network maturation. By integrating transcriptomic data, functional genomic annotations and protein-protein interaction resources we support that astrocytic extracellular signalling is important for the ability of astrocytes to mediate functional neuronal maturation. This work provides a foundation for future investigations into astrocyte-neuron interactions in human health and disease.
