Paired subcloned human large cell lung carcinoma cell lines: low-metastatic 95C vs high-metastatic 95D

ABSTRACT: The paired low-metastatic 95C and high-metastatic 95D cell lines were subcloned from a low differentiated human large cell lung carcinomacell line PLA-801. The cells were well authenticated and published by several research groups. The cells were kindly provided by Professor Ying-Lin Lu, Department of Pathobiology, Institute of Basic Medical Sciences, Academy of Military Medical Sciences, Beijing, China on Dec. 5，2009. As described The 95C and 95D cells were cultured in RPMI 1640 (Invitrogen, USA) with 100 units/mL penicillin, 100 μg/mL streptomycin and 10% calf bovine serum, and grown at 37° C in atmosphere with 5% CO2. miRNAs differential expression between 95C and 95D was measured using a miR human_01_H10.1_080277 miRNA array (LC Sciences Houston, USA).

Transcriptomics changes in liver metastatic biopsies of colorectal carcinoma between pre-treated and acquired resistance to Cetuximab [miRNA]

Project description:Two patients who were newly diagnosed and pathologically confirmed metastatic colorectal cancer were screened for eligibility between August 2011 and December 2013 in Affiliated Hospital, Academy of Military Medical Sciences. They were treated with Cetuximab in combination with FOLFOX regimen and obtained continuous partial responses in more than six months. CT scans of liver lesions were performed every two to eight weeks. The four biopsies were evaluated by two pathologists and were confirmed that the tumor purities were greater than 50%. Each biopsy was subjected to high-throughput RNA and small RNA sequencing to obtain expression profiles of genes and miRNAs.

Project description:ARPE-19 RPE cells obtained from the Type Culture Collection of the Chinese Academy of Sciences (Shanghai, China) were cultured in DMEM medium with 10% FBS (Thermo Fisher Scientific, Waltham, USA) at the incubator with 37 °C, 5% CO2 and 100% humidity. Final concentration of 15 μM curcumin (C7727, Sigma-Aldrich, St. Louis, USA) and 100 μM CoCl4 (10007216, Sinopharm Chemical Reagent, Shanghai, China) were added within serum-free medium for 24 h and 4 h respectively. RNA-seq was performed to investigate the transcriptome alteration.

TMT-labeled quantitative protein profiling of hippocampus during human aging

Project description:Human hippocampus is one of the critical brain regions affected by aging. However, a fully understanding of aging-related changes in protein profiles in human hippocampus has not been well described, especially for the Chinese population. With the newly founded human brain bank in Chinese Academy of Medical Sciences & Peking Union Medical College, we performed a 4-plex TMT labeled proteomic study in the hippocampus of postmortem human brains. The ages of death ranged from 22-98, and were grouped into 4 aging groups: 20-50, 50-70, 70-90, and over 90 (n=4 each). None of the donors was diagnosed with neurodegenerative diseases according to their medical history. Our data identified 4582 proteins, among which 99 proteins were upregulated and 42 proteins were downregulated during the aging process.

Preparation of tirapazamine-resistant hepatoma cell line

Project description:The aim of the project was to obtain murine hepatoma MH22a [1] cell subline, resistant to anticancer agent 3-amino-1,2,4-benzotriazine-1,4-dioxide (tirapazamine, TPZ), and examine the changes of the protein expression as compared with parental cells, with the main emphasis on the changes of redox proteins. MH22a cells obtained from the Institute of Cytology of the Russian Academy of Sciences (St. Petersburg, Russia), were grown at 37Â°C in DMEM medium, supplemented with 10% fetal bovine serum and antibiotics in T25 flasks until reaching 70-80% confluence. The medium is then changed to an analogous medium with 5.0 Î¼M TPZ, and, depending on cell viability, 50% of the medium with TPZ is changed 1-2 times a week, repeating that procedure for 3.5 months to form a stable monolayer. The resulting monolayer is inoculated 1:2 and further grown by changing 50% of the culture medium twice a week until re-inoculation. 20 passages were made over 2.5 months in total. We were unable to obtain a cell subline in a stepwise fashion by growing the cells in increasing concentrations of TPZ, because its concentration increase up to 10 Î¼M caused cell death after 1 month. For analysis, one flask per each sample, 3 samples of parental cell line and 3 samples of TPZ-resistant cell line (between 15th and 20th passages), were selected. Cells were rinsed three times with PBS, detached with TrypLE Express (ThermoFisher Scientific, Waltham, MA, USA) and cell concentration and viability was determined with Trypan blue and Countess 2 (ThermoFisher Scientific, Waltham, MA, USA) automated cell counter. Cells were rinsed by centrifuging 500xg for 5 min and suspending in PBS three times. Finally, cells were flash frozen in liquid nitrogen and transferred to -86Â°C freezer for storage. Identification of long non-protein coding RNAs in chicken skeletal muscle by Solexa sequencing

Project description:Deep sequencing as a high-throughput technology has been widely used in the transcriptome profiling in mammals. In the present study, we aimed to identify chicken lncRNAs ranging from 300-1600 nt long. Total RNAs from chicken skeletal muscle at the embryonic stage were fractionated by 6% urea PAGE. Selected RNA fractions (300-1600 nt) were sequenced by Solexa technology. Chicken eggs (White Leghorn) were incubated at 37.5°C for 10 days (E10), 12 days (E12), 14 days (E14), 18 days (E18). Skeletal muscles (pectoralis) were collected from chicken embryo incubated at above four stages. All muscle samples were immediately frozen in liquid nitrogen and stored at -80°C until RNA isolation. All embryonic manipulations were conducted in accordance with the protocols of the Chinese Academy of Medical Sciences and the Institutional Animal Care and Use Committee of Peking Union Medical College. The single 'Chicken_muscle' Sample represents muscle pooled from all four stages.

Inferior Temporal Gyrus in Human Ageing: a Quantitative Protein Profiling Work based on Chinese Human Brain Bank

Project description:The temporal lobe is the cerebral cortex with critical function. The superior and middle gyrus of temporal lobe have been well studied, however, present perceptions on inferior temporal gyrus remains limited. The understanding of age-related protein profile change in human inferior temporal gyrus has not yet been well established. This 3-plex TMT labeled proteomic study is performed based on the human brain bank at the Chinese Academy of Medical Sciences & Peking Union Medical College. Age distribution of the donors ranges from 22 to 90 years old, and were assigned to three age groups: 20-50, 50-70, and 70-90 years of death age. In this ageing cohort, no neurodegenerative disorders or major stroke events were identified via standard neuropathological classification. Proteomics and bioinformatics strategies were applied to identify the perturbations of protein expression and associated pathways. Among all the ITG samples, 3113 proteins were isolated, with 37 proteins upregulated and 21 proteins downregulated.
