Project description:The Uppsala Longitudinal Study of Adult Men is a population-based study aimed at identifying risk factors for cardiovascular disease. At the time of biopsy all subjects were ~ 70yr of age We extracted RNA from muscle tissue taken from 129 subjects, when they were aged ~70yr and characterised as disease-free (note the above average longevity in Swedes born circa 1920 compared with US and UK populations). From these samples, 108 yielded RNA of sufficient quality to profile on Affymetrix gene-chips. Only survival data are used in the paper. There are no data from cardiovascular disease subjects; we only profiled the healthy subjects and followed for 20yrs.

Project description:The Uppsala Longitudinal Study of Adult Men (ULSAM) study is an ongoing, longitudinal, epidemiologic study started in 1970 and based on men born between 1920 and 1924 in Uppsala, Sweden which were invited to participate at age 50 (N=2,841) [PMID: 9484991]; 81.7% (N=2,322) participated. Individuals were reinvestigated at the ages of 60, 70, 77, 82 and 88 years. Information collected includes a medical questionnaire, blood pressure and anthropometric measurements, glucose tolerance test and 24-hour ambulatory blood pressure. Metbaolomics has been performed on plasma samples from age 70. In the final analysis we included samples from 1,138 individuals.

Project description:Genome wide DNA methylation profiles of whole blood from HIV positive men. The Illumina Infinium 450k Human DNA methylation Beadchip v1.2 was used to obtain DNA methylation profiles across approximately 480,000 CpGs. Dataset included 120 subjects from the USA but because of missing clinical characteristics only 109 subjects were used in the scientific publication. The study analyzed the effect of HIV viral load on host DNA methylation levels. Bisulphite converted DNA from the 120 HIV positive men were hybridised to the Illumina Infinium 450k Human Methylation Beadchip. Subjects had different levels of HIV viral load. This dataset reports DNA methylation data set on 120 subjects that were generated in 2013 (while the other data set of 24 subjects reports DNA methylation data generated in 2012). Details on what each sample characteristics and their values represent are provided in the 'characteristics_readme.txt' file.

Project description:Type I interferons (IFN-I) are important mediators of antiviral immunity and autoimmune diseases. Plasmacytoid dendritic cells (pDCs) from females exert an elevated capacity to produce IFN-I in response to activation of toll-like receptor 7 (TLR7) compared to male pDCs, and both hormones and genes encoded by the X chromosome have been implicated in these sex-specific differences. Using longitudinal samples collected from a cohort of trans men receiving testosterone injections as gender-affirming hormone therapy (GAHT), the impact of testosterone on TLR7-mediated IFN-I production by pDCs was assessed. GAHT induced testosterone and estradiol levels within male reference ranges and increased hemoglobin and hematocrit levels, demonstrating biological activity of testosterone. scRNA seq of pDCs showed downregulation of IFN-I-related gene expression signatures, but also revealed inter-donor heterogeneity on the transcriptional level. Longitudinal quantification of IFN-I protein production by pDCs following TLR7-stimulation showed significant and continuous reduction of IFN-I production in trans men, and reduced expression of IFN-I-stimulated genes. These longitudinal studies in trans men demonstrate that testosterone can reduce IFN-I production by pDCs, and provide novel insights into the immune-modulatory role of testosterone in sex differential IFN-I-mediated immune responses.

Project description:In this study, we used a precision medicine approach to examine molecular profiles and cellular stress responses in an important segment of AA and EA men: men undergoing prostate biopsy. The primary goal in the present study was to demonstrate the ability to assess the prostate transcriptome with a single biopsy core while further examining molecular differences in gene expression patterns relevant to prostate cancer disparities between AA and EA men in a new patient cohort presenting with early stage prostate cancer. To further this objective, we undertook a series of genome wide expression profiling experiments using high throughput RNA sequencing (RNA-Seq). RNA was purified from core biopsy prostate tissue specimens obtained from deidentified subjects enrolled in the study. Transcriptional profiles of each of the deidentified patient’s tissue samples were generated and systems level analyses were performed. The results of these analyses were compared across racial groups.
