We used microarrays to identify global patterns of gene activity during somatic embryogenesis in a wild type (Col-0) and vitamin C deficient mutant (vtc2) RNA was extracted and amplified from four stages of somatic embryogenesis (bent cotyledon, day 7 induction, day 14 induction, and mature somatic embryos) in a wild type (Col-0) and vitamin C deficient mutant (vtc2) before being hybridized to the Arabidopsis ATH1 GeneChip in duplicate (two biological replicates).

Project description:A soybean ortholog of the Arabidopsis MADS-domain transcription factor (called GmAGL15) enhanced somatic embryogenesis from immature cotyledon explants of soybean when expressed via the 35S promoter compared to non transgenic tissue (cultivar Jack). To better understand how this occurs an expression microarray experiment was performed. publication: Q. Zheng and S.E. Perry. (2014). Alterations in the transcriptome of soybean in response to enhance somatic embryogenesis promoted by orthologs of AGAMOUS-Like 15 and AGAMOUS-Like 18. Plant Physiology, in press. Comparison of transcriptomes of non-transgenic tissue to 35S:GmAGL15 tissue at the time of explant preparation (0 days) and 3 and 7 days after induction of somatic embryogenesis by placement on medium containing 2,4-D.

Transcription factor transcriptome during somatic embryogenesis (SE) in Arabidopsis thaliana

Project description:To identify transcription factor (TF) genes exhibiting preferential expression during SE, expression profiles of 1,880 TFs were compared in two genotypes with largely different capacities for SE induction in IZE culture on auxin medium, namely the highly embryonic Col-0 accession and the tanmei mutant unable to form somatic embryos. Our study revealed 729 TFs whose expression changes during the 10-day incubation period of SE; 141 TFs displayed distinct differences in expression patterns in embryogenic versus non-embryogenic cultures. This study provides comprehensive data focused on the expression of TF genes during SE and provides guidelines for further research on functional genomics of SE. The experiment was designed to monitor the expression of 1,880 TF genes at three distinctive stages of IZE-derived embryogenic culture, which refer to: (i) freshly isolated explants competent to undergo embryogenic transition (0 d), (ii) explant tissue subjected to SE induction (5 d), and (iii) the advanced phase of embryogenesis related to somatic embryo formation (10 d).
