Transcription profiling of doxorubicin sensitive and resistant human myeloma cell lines after treatment with doxorubicin to explore the role of the STAT1 signaling pathway in drug and radiation resistance

Project description:The myeloma cell line RPMI 8226/S and its doxorubicin resistant subline 8226/Dox40 were used as models to explore the potential importance of the STAT1 signaling pathway in drug and radiation resistance. The 40-fold doxorubicin resistant subline 8226/Dox40 was found to be crossresistant to single doses of 4 and 8 Gy of radiation. A genome-wide mRNA expression study comparing the 8226/Dox40 cell line to its parental line was performed to identify the underlying molecular mechanisms. Seventeen of the top 50 overexpressed genes have previously been implicated in the STAT1 signaling pathway. STAT1 was over expressed both at the mRNA and protein level. Moreover, analyses of nuclear extracts showed higher abundance of phosphorylated STAT1 (Tyr 701) in the resistant subline. Preexposure of the crossresistant cells to the STAT1 inhibiting drug fludarabine reduced expression of overexpressed genes and enhanced the effects of both doxorubicin and radiation. These results show that resistance to doxorubicin and radiation is associated with increased STAT1 signaling and can be modulated by fludarabine. The data support further development of therapies combining fludarabine and radiation.

Identification of differentially expressed miRNAs in drug resistant and drug sensitive breast cancer tissues

Project description:To identify miRNAs involved in drug resistance of human breast cancer, a miRNA microarray was performed on 5 cases of drug resistant tissues and 5 cases of drug sensitive tissues.The expression levels of totally 2019 miRNAs in 5 pairs of matched, drug resistant and drug sensitive tissues were examined by microarray. There were 27 differentially expressed miRNAs between drug resistant and drug sensitive tissues were identified of which there were 11 significantly up-regulated while the other 16 were down-regulated in drug resistant tissues compared to drug sensitive tissues. It was found that miR-489 was one of the most downregulated miRNAs in drug resistant tissues.

Project description:Cisplatin and carboplatin are the primary first-line therapies for the treatment of ovarian cancer. However, resistance to these platinum-based drugs occurs in the large majority of initially responsive tumors, subsequently resulting in a poor long-term prognosis. To model the onset of drug resistance, and investigate the DNA methylation alterations associated with cisplatin resistance, we treated clonally derived, drug-sensitive A2780 epithelial ovarian cancer cells with increasing concentrations of cisplatin. After several cycles of drug selection, the isogenic drug-sensitive and -resistant pairs were subjected to global CGI methylation microarray analyses. Comparative Transcriptome Analyses of Drug-sensitive and Drug-resistant Strains of Eimeria tenella by RNA-sequencing

Project description:To gain insights into the molecular mechnism governing the drug resistance of Eimeria tenella, two drug-resistant strains of E. tenella, maduramicin-resistant (MRR) strain and diclazuril-resistant (DZR) strain were induced.We carried out comparative transcriptome analyses of a drug-sensitive strain (DS) and two drug-resistant strains (MRR and DZR) of E. tenella by RNA-seqencing. A total of 1070 DEGs, 672 upregulated and 398 downregulated, were identified in MRR vs. DS; and 379 DEGs, 330 upregulated and 49 downregulated, were detected in DZR vs. DS. Functional annotation analysis identified several DEGs coding for proteins associated with catalytic activity in the DZR strain that were involved in glycolysis and the tricarboxylic acid (TCA) cycle. Other DEGs were associated with ion binding and ion transmembrane transporter activity in the MRR strain. Some DEGs coded for surface antigens that were downregulated in two drug-resistant strains involved invasion, pathogenesis, and host–parasite interactions.These results contribute to developing rapid molecular methods to detect drug resistance of Eimeria spp. in poultry.
