ABSTRACT: To characterize the ecological interactions among S. cerevisiae strains coming from the same geographical area, we examined the fitness of two natural isolates from San Giovese grapes, alone or in competition, in synthetic wine must (SWM). We performed genome-wide analyses in order to identify the genes involved in yeast competition and cooperation.

Project description:Grapes are a valuable fruit and an important economic crop in the world, where wine production is a major industry. In grapevine, the environmental regulation of bud dormancy varies among its diverse genotypes. Certain grapevine genotypes become dormant in response to decreasing photoperiod and others require low temperature or both environmental cues to induce dormancy. This study used a proteomic approach to gain an understanding of the underlying molecular events involved in bud dormancy commitment.

The RNA binding protein Swm is critical for Drosophila melanogaster intestinal progenitor cell maintenance

Project description:The regulation of stem cell survival, self-renewal, and differentiation is critical for the maintenance of tissue homeostasis. Although the involvement of signaling pathways and transcriptional control mechanisms in stem cell regulation have been extensively investigated, the role of post-transcriptional control is still poorly understood. Here we show that the nuclear activity of the RNA-binding protein Second Mitotic Wave Missing (Swm) is critical for Drosophila melanogaster intestinal stem cells (ISCs) and their daughter cells, enteroblasts (EBs), to maintain their progenitor cell properties and function. Loss of swm causes ISCs and EBs to stop dividing and instead detach from the basement membrane, resulting in severe progenitor cell loss. swm loss is furthermore characterized by nuclear accumulation of poly(A)+ RNA in progenitor cells. Swm associates with transcripts involved in epithelial cell maintenance and adhesion, and the loss of swm, while not generally affecting the levels of these Swm-bound mRNAs, leads to elevated expression of proteins encoded by some of them, including the fly ortholog of Filamin. Taken together, this study indicates a nuclear role for Swm in adult stem cell maintenance, raising the possibility that nuclear post-transcriptional gene regulation plays vital roles in controlling adult stem cell maintenance and function.

Comparative genomic hybridization of wild type yeast isolated from clinical samples and wine fermentations to investigate genetic variability in yeast from different ecological niches

Project description:Using microarrays targeting the complete ORFeome of Saccharomyces cerevisiae S288c, this study investigates the genetic variability present in wild type yeast from different ecological niches by comparative genome hybridization on array (aCGH). Genomic DNA of Saccharomyces strains isolated from clinical samples and strains from wine fermentations, either commercially available or isolated form spontaneous wine fermentations, was compared to genomic DNA of the sequenced laboratory strain S288c, using a common reference experimental design, with two dye-swap replicate hybridizations for each strain and six S288c self-self hybridizations for background noise estimation, in a total of 38 hybridizations.

Shotgun proteomics study on Cabernet sauvignon cells exposed to hot and cold temperature stresses

Project description:Grapes are a valuable fruit and an important economic crop in the world, where wine production is a major industry. Drought, salinity and extreme temperatures are abiotic stresses that can trigger significant complex responses in grapevines. This project investigates plant protein reactions in response to abiotic stresses, with particular reference to proteomic changes induced by the impact of hot and cold temperature stress on cultured Cabernet sauvignon cells. The aim of this quantitative label-free shotgun proteomics experiment is to provide insights into the targeted proteins, metabolic pathways and regulatory networks that are related to temperature stress in grapevine and futuristically assist in marker assisted selection.
