Project description:Microarrays were used to detail the global programme of gene expression comparing wild-type and RNAi knock-down plants of SPT4-1 and SPT4-2 Arabidosis seeds were stratified for 2 days and grown for 10 days on solid (0.8% Agar) MS in an plant incubator in a 16 hours day, 8 hours night rythm. Only the aerial part was harvested 2h after turning on the light.

Project description:Housekeeping degradation of normal ribosomal RNA in eukaryotes is completed by the RNase T2 family of ribonucleases. This family of ribonucleases is found through eukaryotes, and the Arabidopsis RNase T2 which degrades ribosomal RNA is RNS2. A null mutant in RNS2, rns2-2, has higher levels of RNA, vacuolar accumulation of rRNA and constitutive autophagy. We used microarray to detail the global changes in gene expression in response to mutation of RNS2 (rns2-2) and identified key biological processes differentially regulated in response to this mutation.

Project description:9-day-old Sertoli cells have a stronger proliferative activity compared with 21-day-old Sertoli cells. We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated and down-regulated genes during this process. Rat Sertoli cell were selected at postnatal day 9 and postnatal day 21 for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain the differentially expressed genes that were involved in cell proliferation, cell cycle, and cell death which were associated with the function of Sertoli cell.
