Genome-wide expression profiling of 30 uM curcumin treatment of immortalized prostate fibroblasts (PShTert-AR)

ABSTRACT: Identifying the effect of the natural agent curcumin on global transcriptional activity of PShTert-AR prostate fibroblast cells over time with a view to understanding mechanisms of action over time. Treatment of PShTert-AR cells for 4, 8, 12 and 16 hours in PShTert-AR cells significantly altered the expression of approximately 1200 genes. Through non-hierarchical cluster analysis, these genes were classified into 3 distinct groups depending on their pattern of temporal response.

PShTert-AR cells were treated with vehicle control (16 hours DMSO) or 30 uM curcumin (4, 8, 12 or 16 hours). Total RNA was extracted. Triplicate RNA samples were pooled and hybrydised to Affymetrix Human Gene 1.0 ST array chips.

Curcumin intake affects miRNA signature in murine melanoma with mmu-miR-205-5p most significantly altered.

Project description:Melanoma is the most aggressive form of skin cancer with estimated 48,000 deaths worldwide. The polyphenol curcumin derived from the plant Curcuma longa is well known for its anti-inflammatory and anti-cancerogenic properties. Accordingly, dietary intake of this compound may be suitable for melanoma prevention. However, how this compound affects basic cellular mechanisms in developing melanoma still remains elusive. Therefore, the aim of this study was to investigate for the first time the impact of oral curcumin administration on the miRNA signature of engrafting melanoma. For this purpose, the effects of a 4% curcumin diet on murine B78H1 melanoma were tested in a flank model. Curcumin diet or standard chow (control) was administered two weeks prior to tumor initiation until termination of the experiment. Highly significant chip-based miRNA array analysis was deployed to detect alterations in the miRNA signature of the tumors. Curcumin treatment significantly reduced the growth of the flank tumors. Furthermore the miRNA expression signature in tumors was substantially altered by curcumin intake with mmu-miR-205-5p over 100 times higher expressed when compared to controls. Putative targets of curcumin-induced up-regulated miRNAs were enriched in o-glycan biosynthesis, endoplasmatic reticulum protein processing and different cancer-related pathways. These findings demonstrate a profound alteration of the miRNA expression signature in engrafting curcumin-treated melanoma with mmu-miR-205-5p being up-regulated most significantly.

Phosphotyrosine profiling of curcumin-induced signaling

Project description:Curcumin, derived from the rhizome of Curcuma longa is a natural anti-cancer agent and has been shown to inhibit proliferation and survival of tumor cells. Although the anti-cancer effects of curcumin are well established, detailed understanding of the signaling pathways altered by curcumin is still unclear. In this study, we carried out SILAC-based quantitative proteomic analysis of CAL 27 cell line, a HNSCC cell line to investigate tyrosine signaling in response to curcumin. Using high resolution Orbitrap Fusion™ Tribrid™ Fourier transform mass spectrometer, we identified 627 phosphotyrosine sites mapping to 357 proteins. We observed alterations in the level of phosphorylation of 307 sites corresponding to 204 proteins upon curcumin treatment.

Integrating network pharmacology, transcriptomics to reveal neuroprotective of curcumin activate PI3K / AKT pathway in Parkinson’s disease

Project description:Background: Parkinson’s disease (PD), a severe threat in aging society, is a significant cause of disable. Curcumin, a polyphenol with hydrophobic properties, has been proved to against Parkinson. Our previous study provides an initial understanding of the neuroprotective mechanisms of curcumin in treating Parkinson’s disease. However, further exploration in this area is needed. The present study was designed to investigate the potential mechanism of curcumin for treating PD. Methods: The therapeutic efficacy of curcumin was evaluated using behavioral tests, immunofluorescence of tyrosine hydroxylase (TH). Network pharmacology and transcriptomics predicted the active pathway and bioprocess of curcumin in PD. Activation of the PI3K / AKT signaling pathway was confirmed by quantitative real-time PCR and immunofluorescence. Result: Curcumin restored the dyskinesia and dopaminergic neurons damage of MPTP-induced mice. The results of network pharmacology and transcriptomics showed that curcumin against Parkinson's disease by regulating inflammation, oxidative stress, and aging. The mechanisms of these were associated with activation of PI3K / AKT pathway. Conclusion: In conclusion, the mechanism of curcumin against PD was revealed by our study which lays a foundation for the application of curcumin in treating.

Expression profiling of Human SUP-T1cells under the treatment with HIV, curcumin and in combination of HIV and curcumin both.

Project description:Expression analysis was performed with human SUP-T1 cell line infected with HIV alone, treated with curcumin alone and infected with HIV in presence of curcumin. The control untreated sample was compared with all the three types of treatments to calculate the fold change. The purpose of the experiment was to identify set of genes that are dys-regulated during HIV infection and shows normal expression when treated with curcumin. This experiment is useful in identifying the mechanism of anti-HIV activity of curcumin at genetic level.

RNA-Seq Analysis of Tetranychus cinnabarinus in Response to Curcumin

Project description:RNA-seq was employed to analyze the transcriptome changes in T. cinnabarinus treated with curcumin or solvent. A total of 105, 706, 297 clean sequence reads were generated from the sequencing with more than 90% of the reads were successfully mapped to the reference sequence. The RNA-seq identified 111 and 96 differentially expressed genes between curcumin and solvent treated mites at 24 h and 48 h post treatment, respectively. GO enrichment analysis showed that differentially expressed genes from 24 h post treatment were categorized into 35 GO functional groups and differentially expressed genes from 48 h post treatment were categorized into 25 GO functional groups. The cellular process under the category of biological process was dominant throughout the GO classification at both time points. Finally, we screened 23 differentially expressed genes that are functionally identical or similar to the targets of common insecticide/acaricides or associated with mite detoxification and metabolism. Overall our results revealed the global transcriptional changes in T. cinnabarinus upon curcumin treatment and will enable the further identification of the targets of curcumin on mite.
