Three time points were established in order to collect all the 2-cell embryos present at each moment to contrast only the two exteme embryo subpopulations: fast against slow. 4 biological replicates of fast (29.5 hpf) vs. slow (46 hpf) 2-cell embryos were contrasted. Each replicate was subjected to a dye-swap procedure.

Expression data from early preimplantation Bovine embryos

Project description:The oviduct is a specialized organ playing crucial roles in the success of early reproductive events and it provides an optimal microenvironment for early embryonic development. However, changes in oviductal environment due to estrus synchronization and superovulation hormonal treatments and subsequent influence on embryos transcriptome profile are not yet investigated. For that, the objective of this study was to investigate differences in developmental rate and transcriptome profile of bovine blastocysts cultured under superovulation or synchronization oviductal environment. Influence of oviductal environment on transcriptome abundance of produced blastocysts was examined using the Affymetrix GeneChip Bovine Genome Array. Eighteen Simmental heifers were synchronized, superovulated and artificially inseminated, then nine of them were flushed at day 2 by transvaginal endoscopic means and 2-cell stage embryos were recovered and endoscopicaly transferred to only synchronized recipients. The remaining half of superovulated heifers and the synchronized recipients were flushed at day 7 to collect blastocysts which developed either in superovulated or synchronized oviduct

Interleukin-6 supplementation improves bovine conceptus elongation and transcriptomic indicators of developmental competence

Project description:A high incidence of pregnancy failures occurs in cattle during the second week of pregnancy as blastocysts transition into an elongated conceptus. This work explored whether interleukin-6 (IL6) supplementation during in vitro embryo production would improve subsequent conceptus development. Bovine embryos were treated with 0 or 100 ng/mL recombinant bovine IL6 beginning on day 5 post-fertilization. At day 7.5 post-fertilization, blastocysts were transferred into estrus synchronized beef cows (n=5 recipients/treatment, 10 embryos/recipient). Seven days after transfer (day 14.5), cows were euthanized to harvest reproductive tracts and collect conceptuses. Individual conceptus lengths and stages were recorded before processing for RNA-sequencing. Increases in conceptus recovery, length, and the proportion of tubular and filamentous conceptuses were detected in conceptuses derived from IL6-treated embryos. The IL6 treatment generated 591 differentially expressed genes (DEG) in conceptuses (n=9-10/treatment). Gene ontology enrichment analyses revealed changes in transcriptional regulation, DNA-binding, and antiviral actions. Only a few DEG were associated with extraembryonic development, but several DEG were associated with embryonic regulation of transcription, mesoderm and ectoderm development, organogenesis, limb formation, and somatogenesis. To conclude, this work provides evidence that IL6 treatment before embryo transfer promotes pre-implantation conceptus development and gene expression in ways that resemble the generation of a robust conceptus containing favorable abilities to survive this critical period of pregnancy.

The 15-min Subcellular Proteome

Project description:This project integrated data-independent acquisition (DIA) with capillary electrophoresis electrospray ionization mass spectrometry (CE-ESI-MS) to enable fast single-cell proteomics. With <15 min of effective separation time in a bottom-up proteomics setting, this technology returned ~1,200 proteins from a single HeLa-cell-equivalent proteome amount. Furthermore, using capillary microsampling to collect the proteome from identified cells in a cleavage-stage embryo of the vertebrate Xenopus laevis (frog) embryo, ~1,400 proteins were identified by measuring ~5 ng of the subcellular proteome content aspirated from live Xenopus embryos. CE-ESI-MS with DIA enhanced proteome detection sensitivity and improved analytical throughput.

Time course of transcriptional reprogramming in bovine nuclear transfer embryos

Project description:Somatic cell nuclear transfer (SCNT) into an enucleated oocyte can reprogram a differentiated nucleus to a totipotent state. However, the time course of transcriptional reprogramming has not been determined. To monitor the inactivation of somatic genes after SCNT in the bovine model system, we determined transcript levels of 159 genes highly expressed in fetal fibroblast nuclear donor cells, but not in metaphase II recipient oocytes. For 18 of these genes significantly higher transcript levels were found in four-cell SCNT embryos compared with four-cell embryos derived by in vitro fertilization (IVF), indicating incomplete silencing of somatic genes at this stage. RNA sequencing revealed that nearly 600 genes with no transcripts in oocytes were activated in eight-cell IVF embryos, in agreement with major embryonic genome activation (EGA). De novo transcription in SCNT embryos was delayed (16 genes before the 16-cell stage, 300 and >800 genes at the 16-cell and blastocyst stages). Intron-containing primary transcripts as another option to detect embryonic gene transcription revealed activation of >2,200 genes in IVF embryos at the eight-cell stage, while only 828 genes were activated in SCNT embryos. At the 16-cell stage, a higher number of activated genes was found in SCNT (1,917) than in IVF embryos (738). Self-organizing tree algorithm clustering confirmed that in SCNT embryos transcription of genes that are normally activated during major EGA is delayed. Our study provides detailed insight into the timing of genome activation in cloned embryos that is substantially different from IVF embryos in spite of similar developmental kinetics.
