Effect of Sunitinib treatment withdrawal on Immunodeficient mice bearing MDA-MB231 tumors

ABSTRACT: Immunodeficient RAG1-/- mice bearing MDA-MB231-LUC+ tumors of 50-70 mm3 were daily administrated by gavage with vehicle (containing, 0.5% carboxymethylcellulose, 1.8 % NaCl, 0.4 % Tween-80, 0.9 %. benzyl alcohol, and ultrapure water adjusted to pH 6.0) or sunitinib (40mg/kg/day) for 30 days followed by treatment withdrawal for 3-4 weeks to allow the tumor regrowth until reaching the same volume as that of the vehicle treated group. Mice were sacrificed when tumor reached the volume of 350-400 mm3 and RNAs of two tumors from two different mice treated with sunitinib or vehicle were extracted.

Development of gene expression profiles for murine breast tumours treated with Sunitinib

Project description:In order to establish a profile of gene expression of tumours treated with sunitinib, 4T1 breast tumours were allowed to establish in Balb/C mice for 1 week. The mice were then treated with 40 mg/kg Sunitinib in PBS and 3.72% DMSO (treatemt group) or PBS-DMSO vehicle alone (no treatment group), daily for the period of the experiment, after which the tumours were resected homogenised and subjected to CD31 microbead separation of endothelial cells and bulk tissue, both of which were subjected to agilent microarray analysis. Treated tumours greater than 250 mm3 (by caliper measurement) after 8 days of treatment were considered resistant to treatment, those smaller than this were considered sensitive to sunitinib treatment. Total RNA was extracted from each tumour sample using the Qiagen miRNAeasy kit, processed into cRNA using the Agilient low input, one colour labelling kit, hybridised onto the 8 x 66k whole mouse genome expression microarrays. Data was normalised in R64 and comparison matrices between treated and untreated endothelum and bulk fractions were constructed. Genes whose expression was significantly altered by sunitinib treatement were taken forward for further analysis.

Intensity-based absolute quantification (iBAQ) of proteins in MDA-MB231

Project description:To determine the absolute copy number of proteins in MDA-MB231 breast cancer cells, we employed IBAQ mediated absolute quantification of proteins based on (Schwanhäusser et al., Nature, 2011), with some modifications. Maqquant calculated iBAQ values were calibrated using spike-in standards, and used to calculate copy numbers for each identified protein within the dataset. Copy numbers for a total of 3,584 proteins were calculated in MDA-MB231 cells.

The transcriptome analysis of Peri-infarct areas of infarction for sham, PT, and Pushen treatment

Project description:We investigated the functional recovery of in ischemic stroke by Photothrombotic stroke (PT) model. RNA-Seq of the peri-infarct cortex at 28 day after PT to clarify the potential mechanisms of pushen capsule against ischemic stroke. Pushen (drug approval number: Z20040074; batch number: 20082403; Suzhong pharma) was dissolved in the vehicle of ultrapure water, and it was administered by oral gavage at 24 hours after surgery continuously for 28 days. Mice in the Sham and Vehicle group were administered with a vehicle of ultrapure water equal to the Pushen group. By applying the cutoﬀs of P <0.05 and |log2 (fold change)|>1, 2369 differentially expressed genes (DEGs) were identified between sham vs PT, and 231 differentially expressed genes (DEGs) were identified between PT vs Pushen.

The effect of MFNG knockdown on gene expression profile of xenograft tumor derived from MDA-MB231 cells

Project description:Mfng, a modulator of Notch signaling, is highly expressed in human claudin-low breast cancer (CLBC). To determine Mfng’s roles in CLBC pathogenesis,we knocked down Mfng in a CLBC cell line MDA-MB231, and found that Mfng knockdown altered Notch activation, decreased tumor sphere formation in vitro, and reduced tumor growth in xenograft model. To identify the potential downstream targets of Mfng during CLBC tumorigenesis, we compared the gene expression profiles between xenografts tumor derived from of MDA-MB231 cells carrying Mfng shRNA and the control vector. Mfng, a modulator of Notch signaling, is highly expressed in human claudin-low breast cancer (CLBC).
