Project description:Successful ex vivo expansion of hematopoietic stem cells (HSCs) would greatly benefit the treatment of disease and the understanding of critical questions of stem cell biology. Microarray studies showed that the HSC supportive mouse fetal liver CD3+ cells specifically expressed angiopoietin-like 2 (Angptl2) and angiopoietin-like 3 (Angptl3). When highly enriched HSCs were cultured in the presence of Angptl2 or Angptl3 together with saturating levels of other growth factors for 10 days, a 24 or 30 fold net expansion of long-term HSCs was observed by reconstitution analysis. The coiled-coil domain of Angptl2 mediated the effects of Angptl2 on HSC expansion. Furthermore, angiopoietin-like 5, angiopoietin-like 7, and microfibril-associated glycoprotein 4 also supported expansion of HSCs in culture. Project description:Cerebral accumulation of inflammatory senescent cells (SnC) may compromise the continuum between endothelial (EC) and neuronal function, leading to vascular cognitive impairment (VCI). We hypothesized that targeted elimination of vascular SnC will preserve cerebrovascular and neuronal functions in atherosclerotic mice of both sexes, demonstrating the causal contribution of SnC in cognitive decline. Male and female atherosclerotic mice (ATX; LDLR-/-;hApoB+/+, n=12 per group) of 6-month of age were treated twice over 3 months with an adeno-associated virus-1 delivering a shRNA targeting vascular Angptl2, a strong inflammatory factor of SnEC; a sh-scramble was used as a control. The Morris water maze test assessed cognition. In males, targeting vascular SnC had no effect on short-term memory but improved long-term memory (p<0.05 vs. sh-scramble); in contrast, in females, the treatment reduced short-term memory (p<0.05), and had no effect on long-term memory. Endothelium-dependent flow-mediated dilations in pressurized cerebral arteries were improved (p<0.05) by the treatment in both males and females. Single-nuclei RNA-sequencing (hippocampal region) revealed a strong sexual dimorphism in transcriptomics profile, with the overexpression of senescence pathways only in male vascular cells. The analysis also revealed that male sex predicted the response to the sh-Angptl2 in oligodendrocytes progenitor cells and neurons (AUC>0.8). Accordingly, the sh-Angptl2 reduced the number of EC expressing angptl2 in males (-75%), but not in females (-10%). In conclusion, these data demonstrate a sexual dimorphism in the causal contribution of vascular senescence in cerebrovascular dysfunction and cognitive impairment: only in male atherosclerotic mice, vascular senescence contributes to VCI.

Project description:Endothelial cells cover the lining of different blood vessels and lymph nodes, and have major functions including the transport of blood, homeostasis, inflammatory responses, control of infiltration of circulating cells into the tissues, and formation of new blood vessels. Therefore, understanding these cells is of major interest. The morphological features, phenotype and function of endothelial cells varies according to the vascular bed examined. The sialomucin, CD34, is widely used as an endothelial marker. However, CD34 is differentially expressed on endothelial cells in different organs and in pathological conditions. Little is known about regulation of endothelial CD34 expression or function. This marker is also strongly modified in-vitro in endothelial cell models, including human umbilical vein endothelial cells (HUVEC) and endothelial progenitor cells (EPC). We have therefore selected CD34high and CD34low endothelial cell subpopulations and examined their CD34 expression and function. Results: Transcriptomic analysis showed that CD34 gene and protein expressions are highly correlated, that CD34high cells are less proliferative, and have a stronger expression of IL-33 and Angiopoietin 2, compared to CD34low cells. Higher secretion levels of IL-33 by CD34high HUVECs was confirmed by ELISA. Finally, when endothelial cells were allowed to interact with peripheral blood mononuclear cells (PBMCs), CD34high endothelial cells induced a stronger proliferation of regulatory T lymphocytes (Tregs) compared to CD34low cells whereas expansion of other CD4+-T cell subsets was equivalent. These results suggest that CD34 expression by endothelial cells in-vitro associates with their ability to proliferate and with an immunogenic ability that favours the tolerogenic response.
