Project description:To understand the regulation of cytotoxicity by decidual CD8+ T cells (CD8+ dT) at the maternal-fetal interface, gene expression analysis of CCR7-CD45RA- effector-memory CD8+ T cells isolated from peripheral blood of unrelated healthy controls and decidual tissue from first trimester (6-12 weeks) and term (>37 weeks) pregnancy was performed. Furthermore, first trimester decidual effector-memory CD8+ T cells were stimulated with anti-CD3/CD28 in the presence of IL-2 for 12 hours or 72 hours. RNA was isolated and gene expression profiles were generated by employing Affymetrix HG_U133_Plus 2 arrays on the Affymetrix Geneatlas system.

scRNA-seq of induced pluripotent stem cell derived decidual natural killer cells

Project description:Decidual natural killer (dNK) cells are the most abundant immune cells at the maternal-placental interface in early gestation. The exploration of dNK cell heterogeneity and function is currently an active area of research, particularly as these differ across gestation and different regions of the placenta. We applied proteomic and transcriptomic single cell definitions of dNK cells and subtypes to characterize these cells at the chorioamniotic membranes (CAM) and basal plate (BP, maternal surface) regions of the placenta at term. We found that there is reduced abundance of low-effector-function dNK1 and increased abundance of high-effector-function dNK3 in term decidual compartments. At term, in comparison to BP-dNK, CAM-dNK had greater abundance of moderate-effector-function dNK2, decreased abundance of dNK3, and lower expression of inhibitory receptor CD9. At the same time, using our knowledge of dNK heterogeneity, we developed a protocol to differentiate induced pluripotent stem cells (iPSC) into dNK cells. In this manuscript, we detail a protocol to differentiate iPSC into dNK-like cells as characterized by scRNA-seq analyses and CD45+CD56brightCD16- protein expression. We further developed the protocol to enrich for dNK2 – the most abundant dNK cell type in first trimester and term CAM – while inducing expression of dNK marker proteins CD9 and CD103 with the addition of TGFß. This study reveals the shift in dNK subtypes underlying functional changes in dNK cells between first trimester and term and detail a protocol with which we can methodically differentiate iPSC into dNK cells and specifically induce the dNK2 subtype for mechanistic perturbation.

Expression data from CD11cHI and CD11cLO decidual macrophage populations.

Project description:Decidual macrophage populations, CD11cHI and CD11cLO cells were analyzed for expression profiles and unique characteristics. We used microarrays to detail the global program of gene expression and to determine differences between these two unique decidual macrophage populations. First trimester decidual tissue was digested in order to release immune cells. Cells were stained for CD14 and CD11c and purified by MoFlo sorting. Cells were immediately lysed for RNA extraction and hybridization on Affymetrix microarrays. We utilized 8 patient paired samples (total of 16 microarrays).

Transcriptional and Functional Programming of Decidual Innate Lymphoid Cells

Project description:A successful pregnancy requires many physiological adaptations from the mother, including the establishment of tolerance towards the semiallogeneic fetus. Innate lymphoid cells (ILCs) have arisen as important players in immune regulation and tissue homeostasis at mucosal and barrier surfaces. Dimensionality reduction and transcriptomic analysis revealed the presence of two novel CD56Bright decidual ILCs, C10 and C2, that express divergent levels of Eomes. We found that decidual C10, C2, and cNKs correlated highly with previously identified first trimester decidual dNKs (dNK1-3). Functional analysis revealed polyfunctional profiles that depend on the nature of the stimulus, with a preference for IFNgamma and VEGF production. Overall, our data suggests persistence of pregnancy-specific decidual innate lymphocytes across pregnancy.

Epigenetic characterization of NKG2C high cells of primigravid decidual NK, multigravid decidual NK, nulligravid endometrial NK and multigravid endometrial NK

Project description:Natural killer cells (NKs) are abundant in the human decidua. Several diseases of poor placental development are associated with first pregnancies, and we thus looked to characterize differences in decidual NKs in first versus repeated pregnancies. We discovered a population unique to repeated pregnancies, which possesses a novel transcriptome and epigenetic signature, characterized with high expression levels of the receptors NKG2C and LILRB1. Activation of these receptors leads to increased production and secretion of IFNγ and VEGFa, the latter found to support vascular sprouting and trophoblast-tumor growth. We propose that this population represents NK “memory” of pregnancy, and have named these cells Pregnancy Trained decidual NK cells (PTdNKs). We further suggest that the precursors of PTdNKs are found in the endometrium. PTdNKs may prove useful in understanding and treating disorders of poor placentation.
