Expression data from pheochromocytoma (PHEO) and paraganglioma (PGL) tumor samples

Project description:Genotype specific differences in expression profiles have been evaluated using human HuGene1.0-ST Gene Chips. In this dataset we include expression data obtained from 8 normal adrenal medulla and 45 PHEOs/PGLs patient samples. Viable appearing tissue from the center of the lesions was collected and snap frozen for RNA extraction. Each of the 45 PHEO/PGL samples was examined by pathologist upon resection. Patients PKh_27 and PKh_28 with SDHB mutation were from the same patient with samples taken from two different locations at different times. Diagnosis of PHEO/PGL has been confired in all cases histopathologically. The tissues were grouped according to genetic/syndromic background and tumor location into SDHB (n = 18), SDHD-A/T (n = 6), SDHD-HN (n= 8), and VHL (n = 13).

Project description:Mean age of patients was 53.2 years old (range, 18–80). Twenty-two patients were males, and 7 were females. Tissues were snap-frozen in liquid nitrogen within 5 min of harvesting, and stored thereafter at －80℃. Samples were specifically re-reviewed by a board-certified pathologist in our institution, using observation of sections of paraffin-embedded tissues that were adjacent or in close proximity to the frozen sample from which the RNA was extracted. Transcription profiling of rat at pheochromocytoma cell line PC12 treated with trimethyltin at low and high doses

Project description:The rat pheochromocytoma cell line PC12 cells were cultured in complete DMEM till 80% confluence, then placed at 5000 cells per squared cm. Cells were then plated in 24-well plates for cell viability assay and in T75 flasks for RNA isolation. Medium was replaced with serum-free fresh medium for 12 hours prior to TMT treatment. Gene expression patterns were then analysed using Rat Expression Array 230A Experiment Overall Design: In this study we analize gene expression patterns in PC12 cells treated with Trimethyltin (TMT). RNA-seq right ventricle congenital heart disease paediatric patients

Project description:RV samples were collected from discarded myocardial tissue from paediatric patients under 16 years of age undergoing their first cardiac surgery at Bristol Royal Hospital for Children. Fresh biopsies (10 mg net weight) were collected from surgery from the apex of the RV immediately after institution of cardiac pulmonary bypass and snap frozen upon harvesting. Strand-specific bulk RNA sequencing was performed on whole tissue snap frozen RV biopsies via Genewiz (Azenta). Briefly, total RNA from RV was isolated from 10mg of tissue, with poly-A selection. Sequencing was conducted on Illumina NovaSeq, yielding 2x150bp paired end reads, 30 million read pairs per sample, with >80% of bases with Q30 or higher.

Prolactinoma is a benign pituitary tumor that produces excessive prolactin resulting in hyperprolactinemia and another disease.Tissues were microdissected and removed using the surgical microscope, rinsed in sterile saline, snap-frozen in liquid nitrogen, and stored (-80 ℃) until analysis. Results provide insight into understanding the difference of gene expression in prolacinomas.
