Expression data from Col-0 and sp1,spx2 under phosphate starvation stress and recovery after resupplying phosphate

ABSTRACT: We performed a transcriptomic analysis of Pi-starvation and recovery after resupplying Pi in Arabidopsis thaliana (Columbia-0) wild type plants and double mutant spx1,spx2. Results show that SPX1 is a Pi-dependent inhibitor of the transcription factor PHR1, a central regulatory protein in the control of transcriptional responses to Pi starvation.

Then, plants were transferred to complete medium for 4 hours to analyze the effect of resupplying plants with Pi.

Project description:We performed a transcriptomic analysis of Pi starvation responses in Arabidopsis thaliana (Columbia-0) wild type plants under phosphate starvation stress and in plants with altered PHR1(-like) activity, comparing mutants of phr1 and phr1-phl1 grown in phosphate-lacking medium. Results show the central role of PHR1 and functionally redundant members of its family in the control of transcriptional responses to Pi starvation. The analysis was performed in wild-type plants grown for seven days in complete (+Pi) and Pi-lacking (-Pi) Johnson solid media and the single phr1 and double phr1-phl1 mutants grown for 7 days in –Pi medium. Three independent biological samples of total RNA from shoot and root were hybridized separately.

Project description:We performed a transcriptomic analysis of Pi starvation responses in Arabidopsis thaliana (Columbia-0) phr1 mutant plants expressing PHR1 in presence of cicloheximide, that inhibit protein translation, thus preventing any effect of PHR1 on the expression of indirect targets. Results show the primary target genes of PHR1 in the responses to Pi starvation. The analysis was performed in phr1 plants and phr1 plants overexpressing a fusion GR:PHR1 (OXGR:PHR1) whose activity is postranslationally controlled by dexamethasone (DEX), that allows studying gene expression upon PHR1 activation and the concomitant inhibition of translation with cycloheximide (CHX), thus preventing any effect of PHR1 on the expression of indirect targets. To perform this study, phr1 and OXGR:PHR1 phr1 plants were grown for seven days in complete Johnson liquid media (+Pi), 2 days in Pi-lacking media (-Pi) and then supplemented with 5 µM DEX and 10 µM CHX for 6 hours before harvesting. Three independent biological samples of total RNA from shoot and root were hybridized separately.

Inositol pyrophosphate InsP8 acts as intracellular phosphate signal in Arabidopsis

Project description:The maintenance of cellular phosphate (Pi) homeostasis is of paramount importance in living organisms. Little is known about how cells sense intracellular Pi status in multi-cellular eukaryotes. Here, we report the identification of inositol pyrophosphate InsP8, a ligand that binds to the Pi sensor protein SPX1 to inhibit PHR1, the central regulator of Pi starvation responses in Arabidopsis. The concentration of InsP8, synthesized by diphosphoinositol pentakisphosphate kinases VIH1 and VIH2, depends on cellular Pi level and the double mutants accumulated higher amounts of Pi. The paradox of Pi toxicity and induction of Pi starvation inducible genes indicated that the cells could not sense cellular Pi without InsP8. Our study reveals that InsP8 acts as a Pi signaling molecule and binds to the receptor SPX1 to regulate Pi homeostasis.

Role of cis-zeatin in root responses to phosphate starvation

Project description:In this work we show that root illumination influences Pi starvation response, enhancing the root and shoot growth arrest and limiting the root/shoot ratio as well as root hair elongation. A transcriptomic study in dark-grown roots (DGR) roots identifies several genes that respond to Pi deficiency that were not previously reported. Hormone profiling revealed that Pi starvation reduces the level of trans-Zeatin (tZ) but significantly increases the amount of cis-Zeatin (cZ) and cis-Zeatin Riboside (cZR). We have analyzed the transcriptomic response of Arabidopsis roots to phosphate starvation combined with cis- or trans-zeatin treatments for 8 hours.

Sugarcane transcriptome - Phosphate starvation

Project description:Rooted sugarcane plantlets, originated from in vitro meristem culture (genotype SP80-3280, CTC, Brazil), were greenhouse acclimatized by initial cultivation on 1/20th strength Hoagland and Arnon (1950) nutrient solution. Nutrient solutions were aired from an oil-less compressor and replaced every 7 days, increasing nutrient concentration to ¼ strength in 3 weeks. Plants were then individually transferred to 2.8 L pots filled with fresh ¼ strength nutrient solution. After one week, half of the plants were transferred to fresh solution containing 250 µM Pi, while the other half was transferred to nutrient solution deprived of phosphate (Pi), with H2PO4 being replaced by H2SO4 (Muchhal et al., 1996). Roots from six plants from each treatment (0 and 250 µM Pi) were harvested 6, 12, 24 and 48 h after exposure to phosphate starvation and immediately frozen in liquid nitrogen. For each time point and each treatment, root samples were aggregated in three pools of two samples each. Extraction of total RNA was performed separately on each sample pool. Dynamic responses of barley root quantitative succinyl-proteome to short-term phosphate starvation and recovery

Project description:Improving phosphate (Pi) acquisition and utilization efficiency is a crucial challenge for the sustainability of agriculture worldwide. The understanding of plant how to response and cope with Pi-limitation, and its underlying molecular mechanisms is key to discovering strategies of efficient Pi acquisition and utilization. Barley (Hordeum vulgare L.) is one of the major cereal crops, has distinct advantage for studying mechanisms of tolerance of phosphorus deficiency due to low Pi demand. Recent studied reveal that post-translational modification (PTM) by phosphorylation, ubiquitination, and glycosylation are play important roles in cellular regulation the plant phosphate starvation response (PSR). Lysine succinylation occurs frequently in the proteins associated with metabolic pathways, which may participate in the regulation of the plant PSR process. However, succinylation precisely modulates the metabolic pathways of proteins in response to PSR in barley remain largely unknown
