Human Uterine Leiomyomata microRNA gene expression

ABSTRACT: MicroRNAs (miRNAs) are a class of small, non-coding RNAs. MiRNAs regulate developmental activity and may be aberrantly expressed in human diseases, including cancer. Expression of miRNAs is tissue or organ specific. Human uterine leiomyomas (ULMs) are the most common neoplasms of women, characterized by the ovarian sex hormone dependence, worse morbidity in black women and genetic heterogeneity. The high incidence of the disease may be associated with developmental defects in uteri. In this study, we examined miRNA expression in 55 ULMs with matched myometrium using spotted microarray platform. Keywords: Disease state analysis

RNA was hybridized to Affymetrix U133A 2.0 transcription microarrays. Differentially expressed genes and pathways were identified using standard methods.

Profiling and Functional Analyses of MicroRNAs and Their Target Gene Products in Human Uterine Leiomyomas

Project description:Human uterine leiomyomas (ULM) are characterized by dysregulation of a large number of genes and non-coding regulatory microRNAs. In order to identify microRNA::mRNA associations relevant to ULM pathogenesis, we examined global correlation patterns between the altered microRNA expression and the predicted target genes in leiomyomas and matched myometria. We found that some but not all of the predicted and inversely correlated target genes in ULMs can be directly regulated by microRNAs in vitro. Our findings provide a broad overview of molecular events underlying the tumorigenesis of uterine leiomyoma and identify select genetic and regulatory events that alter microRNA expression and may play important roles in ULM pathobiology by positively regulating tumor growth while maintaining the non-invasive character of ULMs Keywords: Differential gene expression Paired normal (myometrium) and tumor (ULM) samples derived from 5 patients were used

Expression profiling of lncRNAs, miRNAs and mRNAs and their differential expression in leiomyoma using next generation RNA sequencing

Project description:Total RNA was isolated from leiomyoma and paired myometrium (N=8) and samples from three pairs were subjected to RNA sequencing. After analysis, 5941 lncRNAs (2813 up- and 3128 down-regulated at ≥1.5 fold), 148 miRNAs (56 up- and 96 down-regulated at ≥1.5 fold) and 3855 mRNAs (2030 up- and 1855 down-regulated at ≥1.5 fold) were differentially expressed in leiomyomas. Using QRT-PCR we further confirmed the expression of HULC, lnc-MEG3, LINC00890, TSIX, LINC00473, lnc-KLF9-1 and lnc-POTEM-3 (lncRNA-ATB) in leiomyoma and matched myometrium (N=8). Our results presented here provide a comprehensive expression profile of lncRNAs in leiomyomas with concurrent integrated expression of miRNAs and mRNAs in leiomyomas.

Proteogenomic Landscape of Uterine Leiomyomas from Hereditary Leiomyomatosis and Renal Cell Cancer Patients

Project description:Pathogenic mutations in fumarate hydratase (FH) drive hereditary leiomyomatosis and renal cell cancer (HLRCC) and increase the risk of developing uterine leiomyomas (ULMs). An integrated proteogenomic analysis of ULMs from HLRCC (n=16; FH-mutation confirmed) and non-syndromic (NS) patients (n=12) identified a significantly higher protein:transcript correlation in HLRCC (R=0.35) vs. NS ULMs (R=0.242, MWU p=0.0015). Co-altered proteins and transcripts (228) included antioxidant response element (ARE) target genes, such as thioredoxin reductase 1 (TXNRD1), and correlated with activation of NRF2-mediated oxidative stress response signaling in HLRCC ULMs. We confirm 185 transcripts previously described as altered between HLRCC and NS ULMs, 51 co-altered at the protein level and several elevated in HLRCC ULMs are involved in regulating cellular metabolism and glycolysis signaling. Furthermore, 367 S-(2-succino)cysteine peptides were identified in HLRCC ULMs, of which sixty were significantly elevated in HLRCC vs. NS ULMs (LogFC=1.86, MWU p<0.0001). These results confirm and define novel proteogenomic alterations in uterine leiomyoma tissues collected from HLRCC patients and underscore conserved molecular alterations correlating with inactivation of the FH tumor suppressor gene.
