Expression data from thioglycollate-elicited peritoneal macrophages from 129S6/SvEvTac and DBA2/J mice

ABSTRACT: Strain differences influence susceptibility to atherosclerosis. Apolipoprotein E-null mice on a DBA/2J genetic background (DBA-apoE) are highly susceptible to atherosclerosis in the aortic root area compared with those on a 129S6/SvEvTac background (129-apoE). To explore strain-specific differences affecting the susceptibility to atherosclerosis, we performed microarray analysis of macrophages from wild type mice of each strains.

Project description:Gene expression patterns were determined from five brain regions (bed nucleus of the stria terminalis, hippocampus, hypothalamus, periaqueductal gray, and pituitary gland) in six mouse strains (129S6/SvEvTac, A/J, C57BL/6J, C3H/HeJ, DBA/2J, and FVB/NJ). eQTL study of bone marrow derived macrophages and endothelial cells from DBA2xAKR F2 mice

Project description:An eQTL analysis between DBA/2J and AKR/J mice strains on the ApoE null background was done in bone marrow derived macrophages and endothelial cells to identify modifier genes that affect atherosclerosis susceptibility. DBA/2J mice were crossed with AKR/J mice and their offspring crossed with each other to yield an F2 generation. Bone marrow derived macrophages were cultured and gene expression quantified using Illumina's MouseRef-8 v2 arrays in 170 F2 mice for an eQTL study. In addition primary endothelial cells derived from the aorta of 48 male F2 mice were also assayed using arrays for eQTL mapping. Lesion area in the aortic root was also quantified in these samples.

Strain-specific differences in brain gene expression in a hydrocephalic mouse model with motile cilia dysfunction

Project description:Analysis of strain-specific differences in gene expression in brains from a hydrocephalic mouse model of primary ciliary dyskinesia. The results identify genes that are differentially expressed between C57BL6/J and 129S6/SvEvTac brains. These genes encode proteins that function in a variety of cellular processes and include some that are relevant to hydrocephalus and cilia function, providing insight into the mechanisms underlying susceptibility to hydrocephalus.

Characterization of cholesterol loading effect on the gene expression of bone marrow derived macrophages of AKR and DBA/2 ApoE-deficient mice

Project description:Strain effects on the response to cholesterol loading in bone marrow derived macrophages (BMM) are not well studied. The atherosclerosis susceptible strain, DBA/2 on the ApoE deficient background and the atherosclerosis resistant AKR strains show differences in cholesterol handling when loaded with AcLDL. To understand the molecular basis for these differences, BMM from both strains were loaded with acetylated low desnisty lipoprotein (AcLDL) and gene expression quantified. BMMs from AKR greatly upregulated transcripts involved in the lysosomal trafficking pathway. This experiment strongly suggest that upregualtion of the lysosomal pathway in response to AcLDL maybe protective against atherosclerosis. In addition we find that transcripts containing the sterol response element (SRE) motif had strong strain-loading interactions suggesting a strain response to SRE response.

Host genetic background strongly affects pulmonary miRNA expression before and during evolution of influenza A virus infection

Project description:Background: The role of host encoded microRNAs in genetically determined host susceptibility to influenza A virus (IAV) infection has not been explored. We therefore compared changes in pulmonary miRNA expression during IAV infection in two inbred mouse strains with differential susceptibility to IAV infection. Results: miRNA expression profiles were determined in lungs of the more susceptible mouse strain DBA/2J and the less susceptible strain C57BL/6J within 120 hours post infection (hpi) with IAV (H1N1) PR8. Even the miRNomes of uninfected lungs differed substantially between the two strains. After a period of relative quiescence, major miRNome reprogramming was detected in both strains by 48 hpi and increased through 120 hpi. Distinct groups of miRNAs regulated by IAV infection could be defined: (1) miRNAs (n= 39) whose expression correlated with HA mRNA expression and represented the general response to IAV infection independent of host genetic background; (2) miRNAs (n=20) whose expression correlated with HA mRNA expression but differed between the two strains; and (3) remarkably, miRNAs (n=8) whose abundance even in uninfected lungs differentiated nearly perfectly (area under the ROC curve >0.99) between the two strains throughout the time course. Higher abundance of miRNAs with anti-apoptotic functions and lower abundance of miRNAs regulating the PI3K-Akt pathway were associated with the more susceptible DBA/2J strain. Conclusions: These results suggest that differences in host miRNA abundance before and during IAV infection are in part determined genetically and contribute to susceptibility to IAV infection in this experimental mouse model, and likely in humans.
