Project description:In order to identify direct or indirect miR-27a modulated genes, RH36 cells transfected with miR-27a precursors (pre-miR-27a or pre-control), analyzed 48h post-transfection, were used to perform microarray analysis. Gene expression profiling was carried out in ERMS cell line (RH36) transfected with miR-27a or with a negative pre-control. We performed the experiments in triplicate. We used the ‘‘Whole Human Genome Oligo Microarray’’ (Agilent), consisting of 41.000 (60-mer) oligonucleotide probes, which span conserved exons across the transcripts of the targeted full-length genes.

Gene expression in human umbilical cord endothelial cells following premiR-100 overexpression

Project description:To search for genes regulated by microRNA-100 in endothelial cells, we transfected human umbilical cord endothelial cells (HUVECs) with miR-100 precursor oligonucelotides. Human umbilical vein endothelial cells (HUVECs) were isolated from donated umbilical cords, pooled from two donors and cultivated up to passage 5. For transfection with pre-miR microRNA precursor molecules cells were cultured to 70% confluence and transfected with 8nM pre-miR-100 or an irrelevant control oligonucleotide (both from Ambion) using Lipofectamin RNAiMax (Invitrogen) according to the manufacturers instructions. Total RNA was isolated 48h after transfection.

MicroRNA-27a regulates lipid metabolism and inhibits hepatitis C virus replication in human hepatoma cells

Project description:Gene expression profiling was carried out in Huh-7.5 cells in which miR-27a was over- or under-expressed. Transfection of cells with pre-miR-27a and pre-miR-control, or anti-miR-27a and anti-miR-control enabled down- and up-regulated genes to be determined, respectively. Replication and infectivity of the lipotrophic hepatitis C virus (HCV) is regulated by cellular lipid status. Among differentially expressed micro (mi)RNAs, we found that miR-27a was preferentially expressed in HCV-infected compared with hepatitis B virus (HBV)-infected liver. Gene expression profiling of Huh-7.5 cells showed that miR-27a regulates lipid metabolism by targeting the lipid synthetic transcriptional factor, RXRα, and the lipid transporter, ABCA1 Carrying out a Target Scan (Release 5.2) of miR-27a predicted 921 candidate target genes, and functional gene ontology enrichment analysis of these genes by MetaCore (Thomson Reuters, NY) showed that miR-27a could target the signaling pathways of cytoskeleton remodeling and lipid metabolism . To examine whether these signaling pathways were regulated by miR-27a, gene expression profiling was carried out in Huh-7.5 cells in which miR-27a was over- or under-expressed. miR-148b modulates the expression of multiple genes

Project description:To identify which miR-148b targets were involved in tumorigenesis, a microarray analysis was performed for miR-148b over-expressing cells versus controls and 129 (49 up and 80 down) modulated genes were revealed. The effects of miR-148b on cancer progression depend on the direct and indirect regulation of multiple target genes. To identify miR-148b modulated genes, MDA-MB-231 cells were transfected with miR-148b precursors or negative controls (pre-miR-148b or control) and used 48h later for microarray and western blot (WB) analyses. When a âWhole Human Genome Oligo Microarrayâ (Agilent) platform was employed, 129 differentially expressed genes (49 upregulated, 80 downmodulated) were found at 48h, considering a fold change (FC) cut of 1.5 and a false discovery rate (FDR) of 16% (Table S4). Crossing these results with the list of putative miR-148b targets (3642) obtained by the miRecords System, we observed that 33 of the modulated genes were also miR-148b predicted targets and interestingly, 26 out of these 33 genes were downmodulated.
