Project description:Somatic cells surrounding the oocyte were sampled at the following stages: developmentally incompetent or poorly competent prophase I oocytes (NC1 oocytes), developmentally competent prophase I oocytes (C1 oocytes), and developmentally competent metaphase II oocytes (C2 oocytes). NC1 samples were collected from immature stage IV follicles, C1 samples from immature stage VI follicles, and C2 samples from in vitro matured stage VI follicles. Global transcriptional profiling was performed using somatic cells collected from xenopus ovarian follicles during in vivo oocyte developmental competence acquisition. Project description:During mammalian oocyte growth, chromatin configuration transition from the nonsurrounded nucleolus (NSN) to surrounded nucleolus (SN) type plays a key role in the regulation of gene expression and in acquisition of meiotic and developmental competence by the oocyte. Nonetheless, the mechanism underlying chromatin configuration maturation in oocytes is poorly understood. Here we show that nucleolar protein DCAF13 is an important component of the ribosomal RNA (rRNA)-processing complex and is essential for oocyte NSN–SN transition in mice. A conditional knockout of Dcaf13 in oocytes led to the arrest of oocyte development in the NSN configuration, follicular atresia, premature ovarian failure, and female sterility. The DCAF13 deficiency resulted in pre-rRNA accumulation in oocytes, whereas the total mRNA level was not altered. Further exploration showed that DCAF13 participated in the 18S rRNA processing in growing oocytes. The lack of 18S rRNA because of DCAF13 deletion caused a ribosome assembly disorder and then reduced global protein synthesis. DCAF13 interacted with a protein of the core box C/D ribonucleoprotein, fibrillarin, i.e., a factor of early pre-rRNA processing. When fibrillarin was knocked down in oocytes from primary follicles, the follicle development was inhibited as well, indicating that an rRNA processing defect in the oocyte indeed stunts chromatin configuration transition and follicle development. Taken together, these results elucidated the in vivo function of novel nucleolar protein DCAF13 in maintaining mammalian oogenesis.

Project description:Vitrification is a method for long-term biological sample cryopreservation without causing intra- and extra-cellular ice formation. We recently established a novel closed vitrification system to cryopreserve mouse ovarian follicles. Using the 3D alginate hydrogel encapsulated in vitro follicle growth (eIVFG) method, we have demonstrated that compared to freshly-harvested follicles, vitrified follicles have normal follicle and oocyte reproductive outcomes. Our recent study further demonstrated the faithful preservation of molecular signatures of follicle-stimulating hormone (FSH)-stimulated follicle maturation in vitrified follicles. However, it is unknown whether ovulation, another crucial gonadotropin-dependent follicular event, and involved ovulatory gene regulatory pathways are well conserved in vitrified follicles. Fresh and vitrified follicles grown for 8 days by eIVFG were collected at 0, 1, 4 and 8-hour post human chorionic gonadotropin (hCG) treatment for the single-follicle RNA sequencing. Principal component analysis (PCA) and Pearson’s correlation analysis revealed that vitrified follicles have similar transcriptomic profiles to fresh follicles. Furthermore, the expression of several genes essential for ovulation were comparable between vitrified and fresh follicles. In summary, these results demonstrate that our closed vitrification system preserves follicular transcriptomic dynamics during ex vivo ovulation. Together with our previous findings that vitrification preserves FSH-stimulated follicle maturation, the integration of vitrification of immature follicles and eIVFG has a great potential to serve as an additional fertilization preservation approach for young cancer patients and endangerers species. Moreover, follicle vitrification enables a high-content ovarian follicle biobank, which can greatly improve the throughput of eIVFG for studying ovulation biology, anovulatory disease, toxicology, and novel contraceptive drug development targeting ovulation.

Project description:The objective of this study was to identify miRNA expression profiles of extracellular vesicles (EVs) from porcine follicular fluids (FFs) in association with oocyte quality. Antral follicles were aspirated individually and oocytes were stained with 0.5% Lissamine Green B stain (LB), a vital stain for oocyte quality. Each oocyte was classified separately according to the stain into high-quality (unstained; HQ) and low-quality (stained; LQ). Oocyte corresponding FFs were pooled together into HQ and LQ groups and their EV-miRNAs were isolated and sequenced. Sequencing analysis revealed that a total of 295 known miRNAs were commonly detected in both groups. MiR-27b-3p, miR-140-3p, miR-29a-3p, miR-202-5p, and miR-16 were the top highly abundant miRNAs in both groups. Differentially expression (DE) analysis exhibited that 19 miRNAs (including miR-193a-5p, miR-125b, and miR-320) were up- while 22 (including miR-9, miR-6516, and miR-206) were down-regulated in the HQ compared to the LQ group.

Project description:The objective of this study was to identify the expression profile of miRNAs in porcine oocytes derived from follicles of different sizes using RNA high throughput sequencing technology. Oocytes were aspirated from large (3-6 mm) or small (<2 mm) ovarian follicles and tested for developmental competence and chromatin configurations. Small RNA libraries were constructed from both groups and then sequenced on an Illumina NextSeq500. Oocytes from large follicles showed higher developmental competence and different chromatin configuration compared to small oocyte group. In total, 167 and 162 known miRNAs were detected in large and small oocyte groups, respectively with 153 miRNAs were commonly expressed in both groups. In addition, 155 predicted novel miRNAs were detected and quantified. MiR-205, miR-16, miR-148a-3p, miR-125b, and let-7 family were among the top 10 highly abundantly expressed miRNAs in both oocyte groups. Further analysis showed that 8 miRNAs were differentially expressed (DE) between both groups (>2 fold change) with 4 up- and 4 down-regulated miRNAs in large compared to small oocyte group. Target gene prediction followed by KEGG pathway analysis revealed 46 pathways that were enriched with miRNA-target genes. Oocyte meiosis pathway and signaling pathways including FoxO, PI3K-Akt, TGFβ, and cAMP were predictably targeted by DE miRNAs. These results give more insights into the potential role of miRNAs in controlling oocyte developmental competence.
