Project description:A bovine oligo microarray platform (GPL7053) was used to evidence differences in gene expression profiles of muscle samples from five (5) dexamethazone plus estradiol treated cows and from five (5) control animals. Samples were stored under vacuum at +4°C and collected at D0 and D14. In this study, we analyzed twenty (20) bovine neck muscle samples stored under vacuum at 4°C and collected at D0 (10) and D14 (10) from five (5) dexamethazone plus estradiol treated cows and five (5) from control animals. Gene expression profiling was performed using the Agilent-015354 Bos taurus Oligo Microarray platform (GPL7053) based on single-colour detection (Cyanine-3 only). Microarrays were scanned with Agilent scanner G2565BA (barcode on the left, DNA on the back surface, scanned through the glass) at a resolution of 5 microns; all slides were scanned twice at two different sensitivity settings (XDRHi 100% and XDRLo 10%); the scanner software created a unique ID for each pair of XDR scans and saved it to both scan image files. Feature Extraction (FE) 9.5 used XDR ID to link the pairs of scans together automatically when extracting data. The signal left after all the FE processing steps have been completed is ProcessedSignal that contains the Multiplicatively Detrended, Background-Subtracted Signal.

Proteomic analysis of adipose tissue revealing differentially abundant proteins in highly efficient mid-lactating cows

Project description:The current situation of rising demand for animal products and sustainable resource usage, improving nutrient utilization efficiency in dairy cows is an important task. Understanding the biology of feed efficiency in dairy cows enables for the development of markers that may be used to identify and choose the best animals for animal production. Thus in this study, ten Holstein cows were evaluated for feed efficiency and adipose tissue samples from five high efficient and five low efficient dairy cows were collected for protein extraction, digestion and data were analyzed for differential abundant proteins enriched in feed efficiency pathways. Among the identified peptides, we found 110 DAPs and two protein networks significantly related to feed efficiency. Among the relative mRNA expression of genes involved in energy metabolism including transcription/translation (STAT2, DDX39A and RBM39) or protein transport (ITGAV), only RBM39 showed significant decrease in high efficient dairy cows. The findings presented here confirmed the Transferrin upregulated in pathways including acute phase response signaling, LXR/RXR activation, FXR/RXR activation of high efficient dairy cows supporting that these pathways are related to feed efficiency in dairy cows.
