Impact of endurance training in MHC-I over-expressed Mice

ABSTRACT: Myosine heavy chain I was overexpressed in mouse skeletal muscle. The transgenic mice, as well as the wild type mice, were endurance trained on treadmill for 5 days/week over 8 weeks at 80% peak aerobic capacity in comparison with untrained mice. The gene expression profiles of the skeletal muscles from wild-type trained (WTT) and untrained (WTUT), and transgenic trained (TGT) and untrained (TGUT) were measured using Affymetrix Mouse430A 2.0 GeneChips. Transcription profiling of right ventricule from arrhythmogenic heterozygous plakoglobin deficient mice undergone endurance training to identify transcriptional responses to training.

Project description:Endurance training accelerated development of right ventricular dysfunction and arrhythmias in plakoglobin+/- mice. Histology and electron microscopy did not identify right ventricular abnormalities. To identify differences in the transcriptional response to training gene expression profiling was performed. Experiment Overall Design: Gene expressson analyses were performed in 5 pairs of plakoglobin (+/-) mice and their wildtype littermates. Four pairs of trained mice and one pair of old untrained mice were studied. Training induced expression of several hypertrophy associated genes such as ANP but no differences between the trained plakoglobin (+/-) mice and wt mice was seen.

Transcriptional control of training adaptation in skeletal muscle

Project description:Skeletal muscle has an enormous plastic potential and adapts to various stimuli such as mechanical stress, nutrients and hormones. While morphological changes observed in endurance trained muscles are well described, the molecular underpinnings of training adaptation is poorly understood. Therefore, the aim of our study was to define the molecular signature of a trained muscle and unravel the transcriptional responses of untrained and trained muscles to an acute bout of endurance exercise. Our results reveal that even though at baseline, the transcriptome of trained and untrained muscles is very similar, training status substantially affects the transcriptional response to an acute challenge, both quantitatively and qualitatively. Therefore, we provide novel insights into the complex molecular processes that are induced in response to exercise in a temporal- and training status-dependent manner. In addition, our study reveals that the epigenetic landscape contributes to the transcriptional memory of a trained muscle.
