DNA microarray analysis following gefinitib/luteolin administration to PC-3 cells

ABSTRACT: To further investigate the molecular mechanism of cell death induction by luteolin and/or gefitinib, we performed DNA microarray analysis, which may reveal the change of gene expression pattern following treatment with these chemicals. We examined PC-3 cells after 24h of treatment with 60μM luteolin and/or 60μM gefitinib because subG1 population has not appeared yet at this timing, which helps to avoid the secondary effects of apoptosis on the alteration of the gene expression pattern.

Project description:To determine if any altered miRNA expression is actually involved in the growth inhibition and/or cell death induction by luteolin and/or gefitinib, we performed miR-array analysis using RNA from PC-3 cells after 24h of treatment with 60Î¼M luteolin and/or 60Î¼M gefitinib. PC-3 cells were treated for 24h with DMSO, 60Î¼M luteolin (Lut), 60Î¼M gefitinib (Gef) or their co-administration (Lut+Gef). Each sample was run in duplicate.

Project description:PC-9 cells were exposed in vitro to the reversible EGFR inhibitor ZD1839 (gefitinib) and resistant clones were isolated. One of these resistant clones (PC-9 GR4) was then exposed to the irreversible EGFR inhibitor PF00299804 and dual-resistant clones (PC-9 DRs), or those resistant to both ZD1839 and PF00299804, were isolated. In order to compare global genomic changes between PC-9, PC-9 GR, and PC-9 DR cells, we performed a genome-wide SNP analysis.

Project description:We report the use of transcriptome sequencing in the intervention of luteolin or quercetin in adipogenesis. We constructed the adipogenic differentiation model of mouse embryonic fibroblasts and obtained the differential genes of luteolin or quercetin regulating adipocyte differentiation by transcriptomic sequencing. We identified 2085 differentially expressed lipogenesis response genes, 181 luteolin response genes and 574 quercetin response genes. This study provides a framework for the network regulation mechanism of luteolin or quercetin inhibiting preadipocyte differentiation.

Project description:We established a gefitinib-resistant cell line (PC-9GR), by serial exposure of gefitinib to PC-9, an originally gefitinib-sensitive lung cancer cell line (PC-9na), for long period.We collected total RNA from both PC-9 and PC-9GR and examined mRNA expression profile, comprehensively. Gefitinib induced gene expression was measured in human lung cancer cell line was measured at 48 hours after exposure to 1uM Gefitinib.
