Project description:In the present study through microarray analysis we looked into the mRNA differential expression in peripheral blood of PCOS patients v/s control women. The results implicated that many signalling networks as MAPK pathway, Androgen signaling, Insulin signaling and Immune signaling are regulated in peripheral blood of PCOS patients. The data indicate that there is generic PCOS specific gene expression in peripheral blood of PCOS subjects which can reflect the same from other PCO tissues. Total RNA was extracted from peripheral blood of 4 PCOS patients and 4 control subjects and compared for mRNA diferential expression through microarray.

Project description:Pancreatic cancer's aggressiveness and poor response to conventional treatment have long necessitated a personalized medicine platform. Patient-derived pancreatic cancer organoids (PCOs) with matching genetic mutations have emerged as valuable tools for drug screening. However, PCOs, composed solely of cancer cells, may exhibit different characteristics from the original tumors due to the absence of the tumor microenvironment (TME). To address these issues, we extensively characterized the transcriptomic features of matched patient-derived mixed PCO-CAF systems, combining pancreatic cancer cells and cancer-associated fibroblasts (CAFs), compared to PCOs and the original tumors. The mixed PCO-CAF model closely resembled the patient tissue's transcriptomic traits, indicating its potential in reflecting the TME. Single-cell RNA sequencing revealed different cell populations in the mixed PCO-CAF system, including myofibroblast-like CAFs and inflammatory CAFs, influencing the essential features of cancer cells in the TME. Several growth factors and cytokines were identified in both the patient tissue and mixed PCO-CAFs, suggesting the importance of intercellular signaling communication. Importantly, disrupting these interactions could lead to antitumor responses. Given the cumulative implications of our dataset, the mixed PCO-CAF emerges as a clinically applicable paradigm for personalized therapeutics.

Project description:Prenatal testosterone treated sheep, similar to PCOS women, manifest reduced cyclicity, functional hyperandrogenism and polycystic ovary (PCO) morphology. The PCO morphology results from increased follicular recruitment and persistence of antral follicles, consequence of reduced follicular growth and atresia, and driven by cell-specific gene expression changes that are poorly understood. Therefore, utilizing RNA sequencing, cell-specific transcriptional changes were assessed in laser capture microdissection isolated antral follicular granulosa and theca cells from 21 months-of- age control and prenatal testosterone treated sheep.

Project description:Polycystic ovary syndrome (PCOS) is a common endocrine disorder in women resulting in ovulation failure and other metabolic problems. However, the underlying mechanisms of it remain largely uncertain due to its complexity of clinical manifestations. Since PCOS is believed as a systemic disorder involved endocrine, metabolism, immune system and many organs, we therefore want to know what happen in the peripheral blood of patients with PCOS. To this purpose, gene expression patterns of peripheral blood from 10 PCOS patients and 10 healthy women were profiled by microarray. The significance analysis of microarray (SAM) software was employed to screen the differentially expressed genes (DEGs) and gene ontology (GO) was used for functional enrichment analysis. Also, quantitative reverse-transcription PCR (qRT-PCR) was performed to confirm the results from microarray. 181 DEGs with a fold change >2.0 and q-value <0.05 were identified between the groups. Of these genes, 149 were up-regulated and 32 down-regulated in PCOS. Importantly, 14 genes associated with inflammatory response pathway were highly enriched in PCOS. Furthermore, qPCR assays validated the dysregulated inflammatory response genes. Our observation reinforces the hypothesis that PCOS is characterized by the presence of systemic inflammatory changes and inflammation is implicated in the pathogenesis of this entity. Further elucidation of the aberrant expression of inflammation-related genes how to affect the pathogenesis of PCOS may lead to the development of novel preventative and therapeutic strategies.

Project description:Follicles of polycystic ovaries (PCO) often become arrested in early antral stages at around 3 to 11 mm in diameter. The condition disturbs dominant follicle selection and may result in altered ovulation and anovulation. During the growth and development of human follicles, the follicular fluid (FF) constitutes the avascular microenvironment in which the oocyte develops and acts as a vehicle for hormone signaling between cues from circulation and follicular cells. Previous proteomics studies performed on FF from women with polycystic ovarian syndrome (PCOS) have revealed information on the protein changes associated with the pathophysiology of this disorder. However, these studies have been conducted on FF samples obtained in connection with oocyte pick-up during ovarian stimulation right at the time of ovulation and are limited to follicular conditions during the follicular phase of the menstrual cycle. This study aimed to detect proteomic alterations in FF from human small antral follicles (hSAF) obtained from women with PCO as compared to normal women.

Project description:Insulin resistance is a common metabolic abnormality in women with PCOS and leads to an elevated risk of type 2 diabetes. Studies have shown that thiazolidinediones (TZD) improve metabolic disturbances in PCOS patients. We hypothesized that the effect of TZD in PCOS is in part mediated by changes in the transcriptional profile of muscle favoring insulin sensitivity. Using Affymetrix microarrays, we examined the effect of pioglitazone (30 mg/day for 16 weeks) on gene expression in skeletal muscle of 10 obese women with PCOS metabolically characterized by a euglycemic-hyperinsulinemic clamp. Moreover, we explored gene expression changes between these PCOS patients before treatment and 13 healthy control women. Treatment with pioglitazone improved insulin-stimulated total, oxidative and non-oxidative glucose metabolism, and reduced fasting serum insulin (all p < 0.05). Global pathway analysis using Gene Map Annotator and Pathway Profiler (GenMAPP 2.1) and Gene Set Enrichment Analysis (GSEA 2.0.1) revealed a significant upregulation of genes involved in mitochondrial oxidative phosphorylation (OXPHOS), ribosomal proteins, mRNA processing reactome, translation factors, and proteasome complexes in PCOS patients after pioglitazone therapy. Quantitative real-time PCR suggested that upregulation of OXPHOS genes was mediated by an increase in PGC-1M-NM-1 expression (p < 0.05). Expression of genes involved in ribosomal proteins and OXPHOS was down-regulated in PCOS patients before treatment compared to matched healthy women using GenMAPP 2.1 and GSEA 2.1. These data indicate that pioglitazone therapy restores insulin sensitivity in part by a coordinated upregulation of genes involved in mitochondrial oxidative metabolism and protein biosynthesis in skeletal muscle of PCOS. These transcriptional effects of pioglitazone therapy may contribute to prevent the onset of type 2 diabetes in these women. Experiment Overall Design: Ten obese women of reproductive age with PCOS participated in the study to test the effect of pioglitazone therapy (data set 1). To test if pioglitazone ameliorate existing defects in PCOS patients, the expression profile of the 10 PCOS patients before treatment were compared to the same cohort of 13 control subjects (data set 2).

Project description:Polycystic ovary syndrome (PCOS) is a common reproductive endocrine disease characterized by persistent anovulation and hyperandrogenism, affecting approximately 8%-10% of women of childbearing age and occupying an important position in the etiology of infertility. There is increasing evidence that lncRNAs are involved in the development of PCOS, but the potential regulatory mechanism is still unclear. This study performed high-throughput lncRNA sequencing of follicular fluid exosomes in non-PCOS infertility patients and PCOS infertility patients. The sequencing results led to the identification of 1253 upregulated and 613 downregulated lncRNAs from a total of 1866 detected candidates. There was no significant difference between the PCOS patients and non-PCOS patients in body mass index (BMI) or the fasting blood glucose (FBG) level. However, luteinizing hormone (LH), estradiol (E2), testosterone (T), serum prolactin (PRL) and anti-Mullerian hormone (AMH) levels were clearly upregulated in PCOS patients compared to those in non-PCOS patients. There was also an increase in LH/FSH (>2) in the PCOS patients. Functional analysis showed that the functions of lncRNAs were related to adenine metabolism, adenine biosynthesis and cytidine triphosphate biosynthesis. These results suggest that lncRNAs may play an important role in the pathogenesis of PCOS and may be potential targets for the diagnosis and treatment of PCOS.

Project description:Using microarray, we compared the expression of miRNAs from the peripheral blood of male subjects with T1DM and T2DM with healthy controls. Healthy male controls used were age-matched to the T2DM group patients with mean(SD) 37.3 (7.1) years. Subjects with T1DM were younger [23.3(1.6) years]. Expression was compared and validated using quantitative real-time PCR. Statistical testing (ANOVA, P-value <0.05) was performed and fold changes with respect to the control were calculated Systolic BP, fasting glucose, HbA1c, total cholesterol and LDL-cholesterol levels were significantly higher in T2DM subjects compared with controls (P-value <0.05). Compared with controls, we identified 37 differentially regulated miRNAs in DM subjects. Among them, 21 miRNAs were upregulated (2-5 fold change, p-value < 0.05) and 16 miRNAs were downregulated (1.5-2 fold change, p-value < 0.05). These miRNAs had gene putative targets primarily involved in regulating pancreatic development and functions, adipocyte differentiation, insulin signaling and glucose-dependent insulin secretion. peripheral blood of male subjects with T1DM and T2DM with healthy controls
