Project description:The loss-of-function mutant of AtLEJ1 had well-developed anther cells and showed an increased accumulation of JA and ET in its flower bud clusters. Conversely, overexpression of AtLEJ1 caused a delay in the development of the floral organs, arrested anther cell development and secondary cell-wall biosynthesis in the endothecium cells, and reduced JA and ET contents in the flower bud clusters, resulting in male-sterility through anther indehiscence. We used microarrays to gain a comprehensive understanding of the transcriptional changes induced by AtLEJ1 gene expression, Experiment Overall Design: We performed a full genome microarray analysis (ATH1 Affymetrix chips) with the wild type, 35S:AtLEJ1/5-8 line and atlej1 mutant. Total RNAs were obtained separately from unopened flower buds at stages 1~12, when the anthers are not yet opened, and from open flowers at stages 13~14, when anthesis occurs.

Project description:The function of the plant hormone jasmonic acid (JA) in development of tomato flowers was analyzed with a mutant defective in JA perception (jasmonate-insensitive1-1, jai1-1). In contrast to Arabidopsis JA-insensitive plants that are male sterile, the tomato mutant jai1-1 exhibits major defects in female development resulting in female sterility. To identify putative JA-dependent regulatory components, transcriptomics was performed using isolated ovules of three different stages of flower development, from both wild type and jai1-1. Among the strongly down-regulated genes in jai1-1, one encoding a MYB transcription factor (SlMYB21) was found. Its orthologue in Arabidopsis has a crucial role in JA-regulated stamen development. SlMYB21 showed transcription factor activity in yeast, interaction with SlJAZ9 in yeast and in planta, and complemented the Arabidopsis mutant myb21-5. To analyze SlMYB21 function in tomato ovule development, CRISPR/Cas9 mutants were created and a TILLING mutant was identified, all showing female sterility and therefore corroborating a function of MYB21 in tomato ovule development. Transcriptomics from wild type, jai1-1 and myb21-2 carpels revealed processes that might be controlled by SlMYB21. The data suggest a positive regulation of JA biosynthesis by SlMYB21, but a negative regulation of the action of auxin and GA. The results demonstrate that SlMYB21 mediates at least partially the action of JA and might control the flower to fruit transition.

Project description:The coordination of pollen viability, stamen filament elongation and anther dehiscence is essential for the successful pollination and reproduction in angiosperms. The hormone jasmonic acid (JA) are crucial for these processes. However, the tight regulation of JA biosynthesis during stamen development is still largely unknown. Here, we demonstrate that the rice (Oryza sativa) ERF-associated amphiphilic repression (EAR) motif-containing protein TCP INTERACTOR CONTAINING EAR MOTIF PROTEIN1 (OsTIE1) tightly regulates JA biosynthesis by repressing TEOSINTE BRANCHED1/CYCLOIDEA/PROLIFERATINGCELLFACTORS (TCP) transcription factor OsTCP1/PCF5 during stamen development. The loss of OsTIE1 function in ostie1 mutants causes male sterility. The ostie1 mutants display inviable pollen, the early stamen filament elongation and the precocious anther dehiscence. JA biosynthesis is activated earlier and JA level is precociously increased in anthers of ostie1. The expression of OsTIE1 is developmentally regulated during stamen development. OsTIE1 is localized in nuclei and has transcriptional repression activity. We further show that OsTIE1 directly interacts with OsTCP1, and overexpression of OsTCP1 caused early anther dehiscence resembling that of ostie1. The expression of genes encoding key enzymes of JA biosynthesis including Oryza sativa lipoxygenase (OsLOX) genes is regulated by the OsTIE1-OsTCP1 complex. Our findings discover that the OsTIE1-OsTCP1 module plays critical roles in stamen development by finely tuning JA biosynthesis, and provides a new foundation to create male sterile plants for hybrid seed production.

Project description:This study was initiated with the objective of identifying the anther/tapetum specific promoters from cotton floral buds. Cotton is an important commercial crop. Hybrid cotton varieties are developed to obtain improved yield and fiber quality. Most of the hybrid seed production in cotton is carried out by hand emasculation, which requires large amount of manpower, resulting in high cost of hybrid seed. We are developing barnase-barstar based male sterility system, which would be a better alternative for hybrid development. The tapetum specific promoters are main requirement for such a system. The study was thus carried out to identify genes expressed in the anthers. Cotton bud sizes were correlated with tapetum development. RNA was isolated from following tissues: • Anther tissues from buds at pre-meiotic stage of development (Tapetum absent) • Buds without anther tissues at pre-meiotic stage of development • Anther tissues from buds during meiosis (Tapetum present) • Buds without anther tissues during meiosis • Anther tissues from buds at post-meiotic stage of development (Tapetum degenerated) • Buds without anther tissues at post-meiotic stage of development • Leaf tissues • Seedling 5 days after germination Biotin labeled cRNA was hybridized on Affymertix cotton Genechip Genome array following Affymetrix protocols. Three biological replicates were maintained.

Project description:Background: Pepper (Capsicum annuum L.) is a major cash crop throughout the world. Male sterility is an important characteristic in crop species that leads to a failure to produce functional pollen, and it has crucial roles in agricultural breeding and the utilization of heterosis. Objectives: In this study, we identified many crucial factors and important components in metabolic pathways in anther and pollen development, and elucidated the molecular mechanism related to pollen abortion in pepper. Methods: Pepper pollen was observed at different stages to detect the characteristics associated with male sterility and fertility. The phytohormone and oxidoreductase activities were detected in spectrophotometric and redox reaction assays, respectively. Proteins were extracted from male sterile and fertile pepper lines, and identified by TMT/iTRAQ (Tandem mass tags/isobaric tags for relative and absolute quantitation) and LC-MS/MS (liquid chromatograph-mass spectrometer) analysis. Differentially abundant proteins (DAPs) were analyzed based on Gene Ontology annotations and the Kyoto Encyclopedia of Genes and Genomes database according to |fold change)| > 1.3 and P value < 0.05. DAPs were quantified in the meiosis, tetrad, and binucleate stages by parallel reaction monitoring (PRM). Results: In this study, we screened and identified one male sterile pepper line with abnormal cytological characteristics in terms of pollen development. The peroxidase and catalase enzyme activities were significantly reduced and increased, respectively, in the male sterile line compared with the male fertile line. Phytohormone analysis demonstrated that the gibberellin, jasmonic acid, and auxin contents changed by different extents in the male sterile pepper line. Proteome analysis screened 1645 DAPs in six clusters, which were mainly associated with the chloroplast and cytoplasm based on their similar expression levels. According to proteome analysis, 45 DAPs were quantitatively identified in the meiosis, tetrad, and binucleate stages by PRM, which were related to monoterpenoid biosynthesis, and starch and sucrose metabolism pathways. Conclusions: We screened 1645 DAPs by proteomic analysis and 45 DAPs were related to anther and pollen development in a male sterile pepper line. In addition, the activities of peroxidase and catalase as well as the abundances of phytohormones such as gibberellin, jasmonic acid, and auxin were related to male sterility. The results obtained in this study provide insights into the molecular mechanism responsible for male sterility and fertility in pepper.

Project description:MALE STERILITY 35 (MS35/ MYB26) is a R2R3-type MYB transcription factor (Steiner-Large et al., 2003). MS35 is suggested to be a key regulator in the process of lignified secondary thickening in the anther endothecium which provides mechanical force for anther dehiscence (Dawson et al., 1999). The ms35 mutant bears non-dehiscent anthers and thus exhibits a male sterile phenotype. This microarray experiment aims to identify genes controlled by MS35 and thus establish the precise role MS35 plays during anther development. We have demonstrated that MS35 is expressed in the anther during pollen mitotic divisions. Comparisons of gene expression will be carried out between anthers from the Ler.gl (wild-type control) and ms35.gl (mutant) to identify genes with altered expression due to the ms35 mutation. The use of the gl marker will enable a prompt identification of the ms35 mutant amongst the segregating population. The wild-type control line was chosen to be Ler.gl which ensures that the changes of gene expression are not due to the gl mutation. Plants were grown in growth chambers at 22h of daylight (167 umol.m2.sec-1) at 24 C and 2h of night at 24 C. Developmental stages of anthers were defined by bud position and DAPI staining. As soon as the plants showed three fertile or sterile siliques, anther and filaments were collected from both lines at Pollen Mitosis I (PM I), bicellular, and PM II stage, separately. RNA was extracted using RNeasy Mini Kit (Qiagen, USA) and digested with DNase to eliminate genomic DNA contamination. The identified genes will be further analysed using bioinformatics and genomic (knockout) resources that are available for Arabidopsis. 12 samples were used in this experiment.
