Project description:Gene expression signatures induced by transient ROS production were characterized in both mouse back skin and tail epidermis samples. A transient in situ ROS production in the tissue was activated by an adapted photodynamic treatment, using methyl aminolevulinate as a metabolic precursor of endogenous Protoporphyrin IX. Gene expression in skin samples was measured 48 h after the photodynamic treatment. Experimental groups included: [1] Back skin control group (n=3 animals); [2] Back skin treated group (n=3 animals); [3] Tail skin control group (n=3 animals); and [4] Tail skin treated group (n=3 animals).

Silencing ferrochelatase in breast cancer cells MDA-MB-231

Project description:Aim was to identify novel targets of protoporphyrin-IX (PpIX) after silencing Ferrochelatase (FECH) enzyme. PpIX is a light sensitive metabolite of heme synthesis. Generation of PpIX is increased if FECH activity is suppressed. Here, silencing of FECH leads to down-regulation of PpIX, measured by a decrease of PpIX-specific fluorescence. PpIX is a photosensitizer used for photodynamic treatment of various carcinomas. Increasing PpIX leads to apoptosis of tumor cells.

Transcription profiling of Arabidopsis irradated with cFR (far-red) light for 18 hr and treated with 20 nM of flumioxazin

Project description:Plastids communicate with the nucleus by means of retrograde plastid signals. The far-red (FR) light insensitive Arabidopsis mutant laf6 disrupted in a plastid-localised ABC-like protein (atABC1) accumulates the plastid signal protoporphyrin IX (proto IX) and has attenuated nuclear gene expression (Moller et al.2001 Genes Dev. 15:90-103). Our data suggests that proto IX accumulation results in hypocotyl elongation in response to FR light and we have demonstrated that by inhibiting the plastid localised protoporphyrinogen IX oxidase (PPO) using flumioxazin wild-type plants phenocopy laf6 by accumulating proto IX with a concomitant loss of hypocotyl growth inhibition in a dose-dependent manner. It is at present unclear what effect increased proto IX has on nuclear gene expression and how this is integrated with photomorphogenic responses such as hypocotyl elongation.

Transcriptome analysis in petals and leaves of chrysanthemums with different chlorophyll levels

Project description:To identify the regulatory steps that control chlorophyll accumulation, we compared gene expression in petals and leaves of chrysanthemum cultivars with different chlorophyll levels. Microarray analyses showed that the expression levels of chlorophyll biosynthesis genes encoding glutamyl-tRNA reductase, Mg-protoporphyrin IX chelatase, Mg-protoporphyrin IX monomethylester cyclase, and protochlorophyllorophyllide oxidoreductase were well associated with chlorophyll content: their expression levels were lower in white petals than in green petals, and were highest in leaves. Among chlorophyll catabolic genes, expression of STAY-GREEN, encoding Mg-dechelatase, which is a key enzyme controlling chlorophyll degradation, was considerably higher in white and green petals than in leaves. We searched for transcription factor genes whose expression was well related to chlorophyll level in petals and leaves and found several such genes.

Interaction between ROS and miR-451

Project description:Reactive Oxygen Species (ROS) could be a stress factor that affects microRNA regulation and function in macrophages. The production of microRNAs (miRNA) is influenced by various stimuli, including environmental stresses. We hypothesized that ROS-associated stress could regulate macrophage miRNA synthesis. p47phox-/- mice have deficient NADPH oxidase activity resulting in decreased ROS production. We cultured bone marrow-derived macrophages (BMDM) from wild type (WT) and p47phox-/- mice and profiled miRNA expression using microarrays. The microarray data reveals that there are differences in the expression levels of different miRs, and our studies suggest functional crosstalk between ROS and miR-451 in the regulation of macrophage oxidant stress.

Our findings suggest a potential therapeutic strategy to target G-quadruplexes and decrease cognitive impairment associated with ATR-X syndrome.

Project description:The current study investigated the effect of RAS-induced ROS production on gene expression in normal human haematopoietic progenitor cells (HPC) using gene expression profiling (GEP) and assessed whether ROS-induced gene expression changes contributed to the pro-proliferative phenotype. In order to determine the ROS-specific GEP, Affymetrix Human Exon 1.0ST arrays were used for the comparison of mutant RAS and control cells cultured in the presence or absence of the NOX inhibitor, DPI, which strongly suppressed the production of ROS.
