Project description:Chronic alcohol ingestion changes the alveolar landscape. We used microarrays to characterize the change in mRNA expression following chronic alcohol ingestion in male Sprague Dawley rates (EtOH 36% of calories) We used microarrays to characterize the change in mRNA expression following chronic alcohol ingestion in male Sprague Dawley rats (EtOH 36% of calories for 6 weeks). Control animals were given an isocaloric substitution for EtOH for 6 weeks. Alveolar type 2 cells were isolated using an immuno-absorption /technique and RNA was extracted by Trizol. Four control animals and three EtOH treated animals were used for this study.

Project description:In this study, we analyzed the role of miR-21 in liver regeneration after partial hepatectomy (PHx) in chronic ethanol-treated rats. Male Sprague-Dawley rats were fed a liquid diet containing 36% of total calories derived from ethanol for 5 weeks; corresponding pair-fed calorie-matched controls were fed diets in which ethanol calories were replaced by carbohydrate. After 5 weeks, locked nucleic acid (LNA)-modified oligo antisense to miR-21 (AM21, Exiqon, Vedbaek, Denmark) was used to inhibit miRNA in vivo, and rats were subjected to 70% PHx. Liver samples were collected at 24h after the surgery. The excised liver samples at t=0 served as within-animal controls. Rat Gene 2.0 ST (Affymetrix, Santa Clara, CA) arrays were used to obtain global gene expression data from pooled liver samples (pools of 3 or 4 biological replicates/array, total 8 arrays). Gene expression was profiled in AM21 treated or untreated chronic ethanol-fed (EtOH) and carbohydrate control pair-fed (CHO) rat liver prior to (control, t=0) and 24 h after 70% partial hepatectomy (PHx).

Project description:In this study, we analyzed the effects of chronic alcohol consumption on liver repair and regeneration after partial hepatectomy (PHx). Rats were fed a liquid diet containing 36% of total calories derived from ethanol for 5 weeks; corresponding pair-fed calorie-matched controls were fed diets in which ethanol calories were replaced either by carbohydrate or by fat. After 5 weeks, rats were subjected to 70% PHx and liver samples were collected at 1, 6 and 24h after the surgery. The excised liver samples at t=0 served as within-animal controls. Project description:Alcohol (ethanol, EtOH) consumption during pregnancy can result in fetal alcohol spectrum disorders (FASDs), which are characterized by prenatal and postnatal growth restriction and craniofacial dysmorphology. The specific mechanisms by which alcohol mediates these injuries have yet to be determined. Cell-derived extracellular vesicles, including exosomes and microvesicles containing several species of RNAs (exRNAs), have recently emerged as a mechanism of cell-to-cell communication. However, EtOH’s effects on the biogenesis and function of non-coding exRNAs during fetal development have not been explored. Therefore, we studied the effects of maternal EtOH exposure on the composition of exRNAs in the amniotic fluid (AF) using a rat fetal alcohol exposure (FAE) model. Timed pregnant Sprague Dawley rats received 0 or 2.5 g/kg of 20% EtOH by oral gavage on embryonic days 5, 8, 10, 12 and 15. Through RNA-Seq analysis we identified and verified AF exosomal miRNAs with differential expression levels specifically associated with maternal EtOH exposure.

Project description:The specific role of nutritional status during gestation in exacerbating or mitigating fetal toxicity of ethanol (EtOH) remains unclear. We used an intragastric infusion model (Total enteral nutrition, TEN) to appropriately control for dietary EtOH consumption and caloric intake. Time impregnated female Sprague Dawley rats (250-300 g) were surgically cannulated with intragastric cannulae (on gestation day 4) and infused either control or EtOH-containing diets. Rats were fed 220 kcal/ kg3/4/d (NRC recommended caloric intake for gestation) or 160 kcal/ kg3/4/d (undernourished to 72% of control) diets containing either 12 g/kg/d EtOH or an isocaloric amount of carbohydrates from gestation days 6-15. Maternal hepatic gene expression profiles were assessed on GD15. The present data dissect specific effects of nutritional status during gestation and EtOH intake and the interaction thereof. Project description:Titanium dioxide nanofiber (TDNF) is widely used in the manufacture of various household products including cosmetics. As a result, there is a possibility of negative impact on human health. The goal of the present study is to investigate the short-term effects of titanium dioxide nanofiber ingestion of male Sprague Dawley rats. TDNF was fabricated via electrospinning method, followed by dissolution in water. Six- to seven-week-old male Sprague Dawley rats were exposed to a total of 0, 40, and 60 ppm of TDNF for 2 weeks via oral gavage. Lungs and other tissues were autopsied and excised immediately frozen in liquid nitrogen and stored at -80 deg C until analysis. Total RNA was converted to single-stranded sense-target, fragmented, labeled, and hybridized to the Affymetrix Rat Gene ST 2.0 microarray according to the manufacturer’s protocol.

Project description:The present research focuses on 2-aminoanthracene (2AA), recognized as a derivative of PAHs. The Centers for Disease Control and Prevention (CDC) characterizes PAHs as exhibiting properties that adversely affect the health of various living organisms. These compounds increase the risk of neoplasms in humans and animals and are implicated in the increase of genetic mutations. 2AA, an aromatic amine, has demonstrated elevated carcinogenic and mutagenic potential on animals7. In the long term, exposure to PAHs may cause damage to the liver and kidneys. Furthermore, 2AA has been associated with the onset of early diabetic symptoms9. Consumption of 2AA by Sprague-Dawley rats has shown inflammatory and genetic overregulation, increased glucose, and decreased insulin production. This study aims to examine the hepatic impact of 2AA ingestion by Sprague-Dawley Rats. Rats have anatomical structures of the organs similar to those of humans, so they are suitable for research in biomedical science.
