Project description:Senescence is a developmental process and chlorophyll is an indicator of leaf senescene. In plants cytokinin plays a role in delaying leaf senescence. Chlorophyll degradation is tightly regulated during senescence and cytokinin might interplay in the chrorophyll degradation pathway to regulate leaf greening. we used microarray to study the differential gene expression during leaf senescence and inpresence of a synthetic cytokinin (6-benzyl adenine). third leaf of 30-day-old rice plant were selected and incubated for 72 hours in solution containing 6-BA in darkness. Control condition leaves were incubated in solution without 6-BA. Three sets of control versus 6-BA treated leaves were used for RNA extraction and hybridation on Affymatrix rice gene chip.

Project description:Melatonin, a natural phytohormone present in most plants, plays multiple roles in plant growth and stress responses. Although melatonin biosynthesis-related genes have been suggested to possess diverse biological functions, their roles and functional mechanisms in regulating rice grain yield remain largely unexplored. Here, we revealed that a rice caffeic acid O-methyltransferase (OsCOMT) gene is involved in melatonin biosynthesis through in vitro and in vivo evidences. Transgenic assays show OsCOMT significantly delays leaf senescence at the grain filling stage, and then improves photosynthesis efficiency. Further experimental and transcriptomic data suggest that OsCOMT inhibits the degradation of chlorophyll and chloroplast, which in turn delay leaf senescence. Histological analysis also reveals the role of OsCOMT in the development of vascular bundle system in rice. The levels of melatonin and cytokinin were significantly increased in the culm of OsCOMT-overexpression plant relative to those of the wild-type (WT). In the OsCOMT-overexpression line, the cytokinin-biosynthesizing genes were up-regulated and the cytokinin-degrading genes were down-regulated, thereby increasing the cytokinin levels compared with the WT. Thus, OsCOMT-mediated vascular patterning may result from the crosstalk between melatonin and cytokinin. More importantly, OsCOMT significantly increased grain number and yield production of rice in various background, including Nipponbare (NIP) and Suken118 (SK118). Our findings show novel insights into melatonin-mediated leaf senescence and vascular patterning, and provide a new strategy to enhance rice yield production.

Chlorophyll catabolic pathway has been intensively studied and nearly all the enzymes involved in the pathway are characterized. However, regulatory mechanism of the pathway is largely unknown at the molecular level. In this study, we performed a yeast one-hybrid screening using a library composed only of transcription factor cDNAs to search for factors controlling expression of the chlorophyll catabolic genes. We identified a common regulator, ANAC046, that directly binds to the promoter regions of NON-YELLOW COLORING 1, STAY-GREEN 1 (SGR1), SGR2, and pheophorbide a oxygenase. Transgenic plants overexpressing ANAC046 exhibited an early senescence phenotype with lower chlorophyll content as compared with the wild-type plants, while loss-of-function mutants exhibited a delayed senescence phenotype with higher chlorophyll content. Microarray analysis of ANAC046 showed that not only chlorophyll catabolic genes but also senescence-associated genes were positively regulated by ANAC046. Here, we demonstrate that ANAC046 is a novel positive regulator of Arabidopsis leaf senescence and exerts its effect through controlling the expressions of Chl catabolic genes and senescence-associated genes. Transcriptomes of leaf of ANAC046-overexpression plant (50 days after seed sawing), seedling of ANAC046-SRDX-overexpression plant (5 days after dark treatment), and seedling of ANAC046-knockout plant (5 days after dark treatment) were measured together with their respective wild-type controls.

Project description:A gene expression analysis of the changes in in popolulus tremula during the autumn sensencece. Leaves were analyzed from late summer and late into autumn. Cytokinins are plant hormones that typically block or delay leaf senescence. We profiled 34 different cytokinins/cytokinin metabolites (including precursors, conjugates and degradation products) in leaves of a free-growing mature aspen (Populus tremula) before and after the initiation of autumnal senescence over three consecutive years. The levels and profiles of individual cytokinin species, or classes/groups, varied greatly between years, despite the fact that the onset of autumn senescence was at the same time each year, and senescence was not associated with depletion of either active or total cytokinin levels. Levels of aromatic cytokinins (topolins) were low and changed little over the autumn period. Diurnal variations and weather-dependent variations in cytokinin content were relatively limited. We also followed the expression patterns of all aspen genes implicated as having roles in cytokinin metabolism or signaling, but neither the pattern of regulation of any group of genes nor the expression of any particular gene supported the notion that decreased cytokinin signaling could explain the onset of senescence. Based on the results from this tree, we therefore suggest that cytokinin depletion is unlikely to explain the onset of autumn leaf senescence in aspen.

Several compounds showed significant activity, especially in delaying senescence in detached wheat leaves. We used microarrays to gather information about the reprogramming of gene transcription when senescent Arabidopsis leaves were treated with selected C2-substituted aromatic cytokinin ribosides that showed high activity in the senescence bioassay. Arabidopsis senescent leaves were treated with cytokinins and subsequently used for RNA extraction and hybridization on Affymetrix microarrays. 21-days old Arabidopsis leaves were treated with the appropriate cytokinin or left untreated (DMSO only).
