Expression data from mouse whole bladders of cyclophosphamide-induced cystitis

ABSTRACT: To analyze the gene expression of non-bacterial bladdder inflammation, mouse cyclophosphamide(CYP)-induced model of cystitis was adapted. Forty-eight hour after CYP or vehicle treatment, total RNA was extracted from CYP-treated group (CYP150) and control group (CYP0). We used microarrays to detail the gene expression underlying bladder inflammation.

Project description:To analyze the gene expression of non-bacterial bladdder inflammation, mouse cyclophosphamide(CYP)-induced model of cystitis was adapted. Forty-eight hour after CYP or vehicle treatment, total RNA was extracted from CYP-treated group (CYP150) and control group (CYP0). We used microarrays to detail the gene expression underlying bladder inflammation. Five-week-old female Balbc/A mice were injected intraperitoneally with 150mg/kg body weight of CYP diluted in 15ml/kg body weight of saline. Mice injected with saline alone served as controls. Treated mice were killed 48 hours after administaration and the whole bladders harvested. Total RNA was extracted and microarray analysis was performed.

Project description:During cyclophosphamide (CYP) induced ctstitis progression differential expression of microRNAs (miRs) play a role in urinary bladder inflammation and detrusor function. We identified eight UBs miRs that are known to be involved in various other bladder pathologies which exhibited altered expression in CYP mice and e predicted to have a role in inflammation and smooth muscle function (miR-34c-5p, -34b-3p, -212-3p, -449a-5p, -21a-3p, -376b-3p, -376b-5p and -409-5p).

Single-Cell RNA sequencing reveals the immune microenvironment and signaling networks in cystitis glandularis

Project description:Cystitis glandularis (CG) is a common chronic bladder hyperplastic disease. The current lack of unified diagnosis and treatment criteria makes it essential to comprehensively describe the inflammatory immune environment in CG research. Here, we performed scRNA-sequencing in CG patients for the first time, in which four inflamed tissues as well as three surrounding normal bladder mucosa tissues were included. Specifically, we isolated 18,869 cells to conduct bioinformatic analysis and performed immunofluorescence experiments. Our genetic results demonstrate that CG does not have the classic chromosomal variation observed in bladder tumors, reveal the specific effects of TNF in KRT15 epithelial cells, and identify a new population of PIGR epithelial cells with high immunogenicity. In addition, we confirmed the activation difference of various kinds of T cells during chronic bladder inflammation and discovered a new group of CD27-Switch memory B cells expressing a variety of immunoglobulins. Our study reveals, for the first time, the different kinds of cell subgroups in CG and provides the necessary basis for the clinical treatment of cystitis glandularis.

Th1/17 polarization and potential treatment by an anti-interferon-γ DNA aptamer in Hunner-type interstitial cystitis

Project description:Hunner-type interstitial cystitis (HIC) is a rare, enigmatic inflammatory disease of the urinary bladder with no curative treatments. In this study, we aimed to characterize the unique cellular and immunological factors specifically involved in HIC by comparing with cystitis induced by Mycobacterium bovis bacillus Calmette–Guérin, which presents similar clinicopathological features to HIC. Here, we show that T helper 1/17 polarized immune responses accompanied by prominent overexpression of interferon (IFN)-γ, enhanced cGAS-STING cytosolic DNA sensing pathway, and increased plasma cell infiltration are the characteristic inflammatory features in HIC bladder. Further, we developed a novel mouse anti-IFN-γ DNA aptamer and observed that intravesical instillation of the aptamer significantly ameliorated bladder inflammation, pelvic pain and voiding dysfunction in a recently developed novel murine HIC mouse model with little migration into the blood. Our study provides the plausible basis for clinical translation of the anti-IFN-γ DNA aptamer in the treatment of human HIC.
