Colorado potato beetle transcriptional response to aegerolysin treatment

Project description:Protein complexes of aegerolysins pleurotolysin A2 (PlyA2) and pleurotolysin B (PlyB) from oyster mushrooms Pleurotus sp. display targeted toxicity against Colorado potato beetle (CPB; Leptinotarsa decemlineata) larvae. This selective toxicity is achieved through aegerolysins' interaction with insect-specific membrane sphingolipids. This study explores the potential adaptive response of CPB larvae to the aegerolysin complex.

Expression profiling of Solanum chacoense in response to Colorado potato beetle herbivory

Project description:We report the transcriptional response to Colorado potato beetle herbivory in leaves of the highly beetle resistant Solanum chacoense diploid line USDA8380-1 (80-) and a susceptible F2 individual (EE501F2_093) derived from a cross between 80-1 and a beetle susceptible line S. chacoense M6. Sampling tissue in a time course during adult Colorado potato beetle feeding provides novel insight to the transcriptomic defense response to this important pest.

Plant genes affected by Solanum tuberosum and Colorado potato beetle interaction

Project description:Potato leaves From Solanum tuberosum var. Kennebec will be wounded and oral secretions from 4th instar CPB will be isolated and added to the plants as described by Kruzmane et al (2002, Physiol. Plantarum 115:577-584). The leaf from the 6th node of the potato plant will be wounded or wounded and treated with oral secretions from CPB. Unwounded leaves from node 1-5 of the wounded and wounded plus oral secretions plants will be harvested as systemic material. The leaves will be harvested after 4 hrs and RNA will be isolated. 4 hrs was chosen because this represents a time when early and late induced genes should both be present. In addition, the leaf from the 6th node will be subjected to feeding by CPB that have been raised on potato leaves and starved for 16 hrs immediately prior to infestation. Insects will be allowed to feed for 1 hr and the leaves will be harvested after 3 additional hrs. An additional set of plants will be used to infest the leaf on the 6th node for 4 hrs. Leaves from the 6th node will be collected from uninfested plants after 4 hrs as a control. Three sets of 6-12 plants will be used for each sample. Keywords: Direct comparison

Aggressive Potato virus YNTN (PVYNTN) and mild PVYN isolates trigger different specific responses in susceptible potato

Project description:Potato virus YNTN (PVYNTN) is one of the most devastating potato virus causing great losses in the potato production industry. PVYNTN induces severe symptoms on inoculated leaves and a disease known as potato tuber necrosis ringspot disease (PTNRD) develops on tubers. Closely related PVYN isolate induces only mild symptoms on inoculated potato leaves and no symptoms on tubers. The early response of sensitive potato cvs. Igor and Nadine to inoculation with PVYNTN and PVYN was analysed allowing identification of genes involved in severe symptoms induction. Microarray and quantitative-PCR analysis was carried out to identify differentially expressed genes after inoculation with both virus isolates. Two distinct groups of genes were shown to have a role in severe symptoms development – one group of genes related to energy production and a second group of genes connected with virus spread. Earlier accumulation of sugars and decrease in photosynthesis was observed in leaves inoculated with aggressive PVYNTN isolate than in leaves inoculated with milder PVYN isolate. PVYNTN isolate was shown not to activate differential expression of antioxidant metabolism and pectinmethylesterase inhibitor (PMEI) leading to a delay in plant response and on the other hand it limited callose deposition enabling faster virus spread through the plant.

Early response of potato plants to virus (PVY-NTN) inoculation

Project description:Potato virus YNTN (PVYNTN), causing potato tuber ring necrosis disease, dramatically lowers the quantity and the quality of the potato yield all over the world. The cultivar Igor is one of the most susceptible cultivars, developing severe disease symptoms on plants as well as on tubers. Finding genes differentially expressed in the early response to infection, when the host response is more defense- than infection- related, could improve our understanding of the potato - PVYNTN interaction. Differential gene expression in early response of potato cv. Igor plants to PVYNTN infection was studied using potato TIGR cDNA-microarrays. Expression was compared between mock inoculated and virus infected plants 12 hours after inoculation, in four biological replicates. Keywords: direct comparison
