Project description:Lungs were collected as soon as possible after thermal injury 30-40 mins post injury for neutrophil isolation. Lungs were minced in HBSS with 1% BSA (Sigma) and strained through a 40um nylon filter (BD). Samples were sorted using BD fusion machine. A 100um nozzle was used at a flow rate of 2.RNA was extracted from these neutrophils and microarray was performed.

Project description:We identified the microglial population in the chick anterior hypothalamus (AH) and showed that early-life thermal stress (on day 3 post-hatch) influences the inflammatory process in the AH, altering microglial transcriptomic signature in response to LPS treatment (on day 10 post-hatch).

We described alterations in genome-wide CpG methylation profile of hypothalamic microglia between heat-conditioned and non-conditioned 10-day-old chicks prior LPS challenge. We suppose that changes in CpG DNA methylation might be involved in reprogramming microglial function.

Transcriptomic profiling by array of long-term thermal acclimation (2 months) at various temperatures in liver tissue of Pachycara brachycephalum

Project description:The Antarctic eelpout, Pachycara brachycephalum, is a member of the cosmopolitan fish family Zoarcidae, which comprises 284 species distributed all over the globe. Since the Antarctic realm is a highly constant habitat, the species P. brachycephalum was employed in a global approach to characterize thermal response mechanisms in terms of long-term cold- and warm acclimation. Specimen of P. brachycephalum were incubated for 2 months at six different temperatures: -1C, 0C (control), 3C, 5C, 7C, 9C. In each group 5 fish were selected for hybridization (except control: n=7). By comparing the features of control and treatment groups we addressed temperature dependent expression profiles in liver tissue.

Muscle injury causes long-term changes in stem cell DNA methylation [RRBS]

Project description:Injury to muscle brings about the activation of stem cells, which then generate new myocytes to replace the damaged tissue. We now demonstrate that this activation causes a dramatic change in the stem cell methylation pattern that prepares them epigenetically for terminal myocyte differentiation. These demethylation and de novo methylation events occur at regulatory elements associated with genes involved in myogenesis. Local injury of one muscle brings about an almost identical epigenetic change in satellite cells from other muscles in the body, as well. Furthermore, this same methylation state is also generated in muscle stem cells of female animals following full-term pregnancy, even in the absence of any injury. In all of these cases, which appear to be mediated by circulating factors, the new methylation profile is then stably maintained in resident muscle stem cells and thus represents a molecular memory of previous physiological events that is probably programmed to provide a mechanism for adapting to the environment.
