The microarray experiment studied the gene expression of untreated and treated Listeria cells following 4h exposure to cclA. A six chip study using total RNA recovered from six separate cultures of L. monocytogenes 08-5923; three grown in TSB (untreated) and three grown in TSB with cclA (treated); Each chip measures the expression level of 2,857 genes from L. monocytogenes 08-5923 with five 60-mer probe pairs (PM/MM) per gene, with 1-fold technical redundancy.

Project description:Carnocyclin A (CCLA) is an antimicrobial peptide produced by Carnobacterium maltaromaticum ATCC PTA-5313, which can be used to control the growth of Listeria monocytogenes in ready-to-eat meat products. The aim of this research was to elucidate the cellular responses of L. monocytogenes 08-5923 exposed to a sublethal dose of CCLA. Microarray, quantitative reverse transcription-PCR, tandem mass spectrometry, and electron microscopy were used to investigate the alteration in gene expression, protein production, and morphological changes in cells of Listeria following treatment with CCLA. The genes involved in metabolism (baiE, trn, and pykA), cell wall synthesis (murZ and dacB2), and cell division (clpE and divIVA) were upregulated following a 15-min exposure to CCLA as a result of stress responses. Genes involved in cell division, cell wall synthesis, flagellar synthesis, and metabolism were downregulated after 4 h as a result of adaptation. Analysis of total soluble proteins confirmed the downregulation of pykA and gnd after 4 h of exposure to CCLA. The absence of flagella was observed in L. monocytogenes following 30 h of exposure to CCLA. A sublethal dose of CCLA induced adaptation in L. monocytogenes 08-5923 by inhibition of expression of genes and proteins critical for synthesis of cell wall structures and maintaining metabolic functions. Both the mannose- and cellobiose-specific phosphotransferase systems could be targets for CCLA.

Gene expression data of ODH-08-treated Western diet-fed murine liver

Project description:Hepatic fibrosis is a dynamic process characterized by the net accumulation of extracellular matrix resulting from chronic liver injury such as nonalcoholic steatohepatitis. During the pathogenesis of hepatic fibrosis, activation of hepatic stellate cells (HSCs) causes transdifferentiation of quiescent cells into proliferative and fibrogenic myofibroblasts. In the present study, we developed a novel RORα-selective ligand, ODH-08, based on the modification of JC1-40, a previously reported N-methylthiourea analog. Administration of ODH-08 to Western diet (WD)-fed mice improved the signs of hepatic fibrosis: decreased hepatic collagen deposition and suppression of the expression of fibrogenic markers. ODH-08 inhibits the TGF1-induced fibrogenic activation of HSCs through suppression of the TGFβ1–SMAD signaling pathway, which represents a novel mechanism for the antifibrogenic effect of RORα. Thus, ODH-08 appears to be a promising antifibrotic agent to treat hepatic fibrosis. We performed a microarray analysis in the liver tissue of ODH-08-treated WD-fed mice to anlyse differentially expressed genes under ODH-08 administration. The control group was vehicle-treated WD-fed mice.

Proteomic dataset of <i>Listeria monocytogenes</i> exposed to sublethal concentrations of free and nanoencapsulated nisin.

Project description:The cellular proteins of L. monocytogenes exposed to free and liposome-encapsulated nisin at sublethal concentration were hydrolyzed by trypsin and examined by tandem mass spectrometry (MS/MS) to obtain proteomic data. In the present study, we use the STRING v11.05 database analyze the interactions among the 78 upregulated proteins from L. monocytogenes obtained after treatment with sublethal concentrations of free and nanoliposome-encapsulated nisin. As result, from the upregulated proteins by free nisin was determined a network with 140 edges with two relevant nodes, containing ribosomal proteins and transmembrane transport proteins (SecD and ABC transport system). These two sets of proteins present biological connection as a group, with strong interactions and are related to detoxification and other Listeria response mechanisms. In addition, a high amount of membrane proteins was identified in the free nisin treatment. On the other hand, in the interaction analysis of upregulated proteins by liposome-loaded nisin, was found 156 edges with a single protein network, the same observed in free nisin, related to ribosomal proteins. Therefore, according with this analysis, the encapsulation of nisin into liposomes cause upregulation of ribosomal and decrease of L. monocytogenes response proteins as compared with free nisin.

Investigating Transcriptomic Induction of Resistance and/or Virulence in Listeria monocytogenes Cells Surviving Sublethal Antimicrobial Exposure.

Project description:The potential transcriptomic induction of resistance and/or virulence in two L. monocytogenes strains belonging to the most frequent listeriosis-associated serovars (i.e., 1/2a and 4b), following their sublethal antimicrobial exposure, was studied through qPCR determination of the relative expression of 10 selected related genes (i.e., groEL, hly, iap, inlA, inlB, lisK, mdrD, mdrL, prfA, and sigB). To induce sublethal stress, three common antimicrobials (i.e., benzalkonium chloride, thymol, and ampicillin) were individually applied for 2 h at 37 °C against stationary phase cells of each strain, each at a sublethal concentration. In general, the expression of most of the studied genes remained either stable or was significantly downregulated following the antimicrobial exposure, with some strain-specific differences to be yet recorded. Thymol provoked downregulation of most of the studied genes, significantly limiting the expression of 6/10 and 4/10 genes in the strains of ser. 1/2a and ser. 4b, respectively, including those coding for the master regulators of stress response and virulence (SigB and PrfA, respectively), in both strains. At the same time, the two genes coding for the invasion internalin proteins (InlA and InlB), with crucial role in the onset of L. monocytogenes pathogenesis, were both importantly upregulated in ser. 4b strain. The results obtained increase our knowledge of the stress physiology of L. monocytogenes under certain sublethal antimicrobial conditions that could be encountered within the food chain and in clinical settings, and may assist in better and more effective mitigation strategies.

Proteome of Listeria monocytogenes FBUNT biofilm cells and response to bacteriocin AL705

Project description:L. monocytogenes FBUNT is able to form biofilm after six days at 10 ºC. Proteome under planktonic growth, lactocin AL705-treated and untreated biofilms was analyzed. Compared to planktonic cells, protein abundance shifts associated with ribosomes function and biogenesis, cell membrane functionality, carbohydrate and amino acid metabolism and transport were observed in sessile cells. When sessile cells were treated with lactocin AL705, proteins up-regulation were mostly related to carbohydrate metabolism and nutrient transport in an attempt to compensate for impaired energy generation caused by bacteriocin interacting with the cytoplasmic membrane. Notably, transport systems as β-glucosidase IIABC (lmo0027), cellobiose (lmo2763) and trehalose (lmo1255) specific PTS proteins were highly overexpressed.
